Background: Fidgetin-like 1 (FIGNL1) is extensively overexpressed in a variety of cancers. It facilitates non‑small cell lung cancer tumor cell proliferation and hepatocellular carcinoma formation due to abnormal DNA repair. Clinically relevant data indicates that its high expression is linked with the poor prognosis of patients with renal clear-cell carcinoma, low-grade gliomas, and hepatocellular carcinoma.

A Phase Ib Study of Linperlisib in the Treatment of Patients with Relapsed and/or Refractory Peripheral T-cell Lymphoma.

Clin Cancer Res

October 2024

State Key Laboratory of Experimental Hematology, National Clinical Research Center for Blood Diseases, Institute of Hematology and Blood Diseases Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College (PUMC), Tianjin, China.

Purpose: Relapsed and/or refractory peripheral T-cell lymphoma (r/r PTCL) is an aggressive and heterogeneous hematologic malignancy with high unmet need. Previously, PI3K inhibitors were shown to be efficacious in B- and T-cell lymphomas, but as a drug class, these agents have frequently been observed to have tolerability limitations. Next-generation agents that improve the tolerability while maintaining efficacy are desirable.

Article Synopsis
  • A study aimed to investigate the link between the 'weekend warrior' physical activity pattern and all-cause mortality in adults with type 2 diabetes was conducted using data from the National Health and Nutrition Examination Survey.
  • The research included over 6,000 participants and found that both weekend warriors and regularly active individuals had lower mortality rates compared to those who were inactive.
  • The results suggest that the weekend warrior activity pattern could effectively reduce all-cause mortality in adults with type 2 diabetes, similar to more regular exercise routines.

Adenoid cystic carcinoma (ACC) is a malignant tumor primarily originating from the salivary glands, capable of affecting multiple organs. Although ACC typically exhibits slow growth, it is notorious for its propensity for neural invasion, local recurrence, and distant metastasis, making it a particularly challenging cancer to treat. The complexity of ACC's histological and molecular features poses significant challenges to current treatment modalities, which often show limited effectiveness.

Cancer-associated fibroblasts (CAFs) represent one of the major components of the tumor stroma, which might create an immunosuppressive tumor microenvironment by inducing and functionally polarizing protumoral macrophages. Previous studies indicated that exosomes derived from CAFs might transmit regulating signals and boost esophageal squamous cell carcinoma (ESCC) development. This study is designed to explore the role and mechanism of CAFs-derived exosomal microRNA-889-3p (miR-889-3p) in ESCC progression.

RNA nanotherapeutics with fibrosis overexpression and retention for MASH treatment.

Nat Commun

August 2024

State Key Laboratory of Natural and Biomimetic Drugs, School of Pharmaceutical Sciences, Peking University, Beijing, China.

Article Synopsis
  • Metabolic dysfunction-associated steatohepatitis (MASH) complicates the delivery of therapeutic proteins due to excess extracellular matrix (ECM), leading to the development of a new treatment strategy called "Fibrosis overexpression and retention (FORT)."
  • FORT utilizes specially designed retinoid-derivative lipid nanoparticles (LNPs) that enhance mRNA expression in fibrotic areas and include modifications to promote protein anchoring in the ECM.
  • The approach has shown a tenfold increase in protein expression and improved retention of engineered therapeutic proteins in fibrotic lesions, proving effective in various animal models of MASH while reducing toxicity.

Article Synopsis
  • A phase III clinical trial was conducted to evaluate the effects of different radiation doses (high vs. standard) and irradiation fields (involved field vs. elective nodal) on survival outcomes for patients with locally advanced esophageal squamous cell carcinoma (ESCC).
  • Results indicated that there were no significant differences in overall survival (OS) or progression-free survival (PFS) between the different irradiation fields, but the high-dose treatment suggested an improvement in PFS compared to the standard dose.
  • The best prognostic outcomes were observed in the high-dose with involved field group (HD+IFI), while the standard dose with involved field group (SD+IFI) had the worst outcomes, and no substantial differences in severe adverse events were

Background: Current research separately analyzed the connection between postpartum depression, fatigue, sleep and infant development. However, depression, fatigue and sleep quality often coexisted as adverse symptoms in postpartum women and influenced infant development together. This study explored the maternal postpartum symptoms on infant growth.

The synergy of morphokinetic parameters and sHLA-G in cleavage embryo enhancing implantation rates.

Front Cell Dev Biol

July 2024

State Key Laboratory of Reproductive Medicine and Offspring Health, Clinical Center of Reproductive Medicine, The First Affiliated Hospital of Nanjing Medical University, Nanjing, China.

This study aimed to assess the relationship between implantation and soluble HLA-G (sHLA-G) expression in cleavage embryo culture medium (ECM) in conjunction with early developmental kinetics determined by time-lapse imaging (TLI). A retrospective, single-center study was conducted involving 238 embryos from 165 patients who underwent Frozen-thawed embryo transfer (FET) using autologous oocytes, with either single or double embryo transfer. TLI morphokinetic parameters (t2, t3, t4, t5, t6, t7, t8, cc2, s2, cc3, s3) of embryos were analyzed, and sHLA-G levels in D3 ECM were measured using an enzyme-linked immunosorbent assay (ELISA).

Background: Various electrocardiographic (ECG) abnormalities are associated with the severity of pulmonary thromboembolism (PTE). The utility of evaluating the clot burden of PTE based on ECG findings alone has yet to be thoroughly investigated in Chinese patients. The aim of this study was therefore to use ECG signs to establish novel models for quantitative and localization analysis of clot burden in patients with acute PTE.

Background: The band and loop space maintainer is used to maintain the missing space of deciduous molars which are lost early. When the second deciduous molar is lost prematurely, the stress on the first permanent molar during different degrees of development may vary when it is the abutment. The design and use of the space maintainer may also lead to damage of the loop.

Article Synopsis
  • The study aimed to explore whether there is a genetic connection between different sleep traits (like chronotype and insomnia) and periodontitis (a gum disease).
  • Using advanced statistical methods, the researchers analyzed extensive genetic data but found no significant influence of sleep traits on periodontitis or vice versa.
  • The findings indicate that there is no evidence of a two-way genetic relationship between sleep characteristics and the risk of developing periodontitis.

Key Laboratory of Neuroregeneration of Jiangsu and Ministry of Education, Co-Innovation Center of Neuroregeneration, NMPA Key Laboratory for Research and Evaluation of Tissue Engineering Technology Products, Nantong University, Nantong, 26001, China.

Skeletal muscular atrophy is a complex disease involving a large number of gene expression regulatory networks and various biological processes. Despite extensive research on this topic, its underlying mechanisms remain elusive, and effective therapeutic approaches are yet to be established. Recent studies have shown that epigenetics play an important role in regulating skeletal muscle atrophy, influencing the expression of numerous genes associated with this condition through the addition or removal of certain chemical modifications at the molecular level.
