Tuning the critical solution temperature (CST) of thermoresponsive polymers is essential to exploit their immense potential in various applications. In the present study, the effect of PEG-methyl ether methacrylate with a higher molecular weight of 1100 g/mol (mPEGMA) as a comonomer was investigated for its suitability for the CST adjustment of LCST-type polymers. The properties of the large unilamellar vesicles (LUVs) from 1,2-dimyristoyl-sn-glycero-3-phosphatidylcholine (DMPC), modified by lipopolysaccharides (LPS) from sv. Enteritidis, which mimics Gram-negative bacteria, were studied by various physical methods. LPS, in the range of 0/20/50 % / relative to the lipid, had a regulatory role in the structure of the LUVs toward the lower size, low polydispersity, and over-a-month size stability due to the lower negative zeta potential.

Multigene panel tests (MGPTs) revolutionized the diagnosis of Lynch syndrome (LS), however noncoding pathogenic variants (PVs) can only be detected by complementary methods including whole genome sequencing (WGS). Here we present a DNA-, RNA- and tumor tissue-based WGS prioritization workflow for patients with a suspicion of LS where MGPT detected no LS-related PV. Among the 100 enrolled patients, MGPT detected 28 simple PVs and an additional 3 complex PVs.

The development of stable biopharmaceutical formulations, such as monoclonal antibodies, poses a great challenge in the pharmaceutical industry. This study investigated the stabilizing effect of 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) in liquid and solid formulations of infliximab during processing and storage. After a long-distance migration, Avars with Eastern Asian ancestry arrived in Eastern Central Europe in 567 to 568 CE and encountered groups with very different European ancestry. We used ancient genome-wide data of 722 individuals and fine-grained interdisciplinary analysis of large seventh- to eighth-century CE neighbouring cemeteries south of Vienna (Austria) to address the centuries-long impact of this encounter. We found that even 200 years after immigration, the ancestry at one site (Leobersdorf) remained dominantly East Asian-like, whereas the other site (Mödling) shows local, European-like ancestry.

ESKAPE pathogens rapidly develop resistance against antibiotics in development in vitro.

Despite ongoing antibiotic development, evolution of resistance may render candidate antibiotics ineffective. Here we studied in vitro emergence of resistance to 13 antibiotics introduced after 2017 or currently in development, compared with in-use antibiotics. Laboratory evolution showed that clinically relevant resistance arises within 60 days of antibiotic exposure in Escherichia coli, Klebsiella pneumoniae, Acinetobacter baumannii and Pseudomonas aeruginosa, priority Gram-negative ESKAPE pathogens.

The primary method of treatment for patients suffering from drug-resistant focal-onset epilepsy is resective surgery, which adversely impacts neurocognitive function. Radio frequency (RF) ablation and laser ablation are the methods with the most promise, achieving seizure-free rates similar to resection but with less negative impact on neurocognitive function. By comparing infant-directed speech to spouse- and dog-directed talk, we aimed to investigate how pitch and utterance length are modulated by speakers considering the speech context and the partner's expected needs and capabilities. We found that mean pitch was modulated in line with the partner's attentional needs, while pitch range and utterance length were modulated according to the partner's expected linguistic competence. In a situation with a nursery rhyme, speakers used the highest pitch and widest pitch range with all partners suggesting that infant-directed context greatly influences these acoustic features.
