Int J Mol Sci

November 2024

Unité de Génétique Moléculaire des Maladies Métaboliques et de la Reproduction, Hôpital Bicêtre, Faculté de Médecine Paris Saclay, INSERM U1193, 94275 Le Kremlin-Bicêtre, France.

Article Synopsis
  • Diminished ovarian reserve (DOR) and primary ovarian insufficiency (POI) significantly contribute to female infertility, with a recent study finding a genetic cause in 29.3% of POI cases.
  • A prospective study of 120 patients with unexplained DOR revealed a 24.2% diagnostic yield using advanced sequencing technology to identify genes related to various biological pathways.
  • Findings indicated that impaired oocyte quality is linked to meiosis/DNA repair genes, and the study supports the need for genetic testing as a routine part of infertility diagnosis and a potential predictor for progression to POI.

Article Synopsis
  • The study compares three bacterial strains from the same patient to understand how antimicrobial resistance affects evolution and fitness, specifically focusing on a resistant strain's genetic changes.
  • Whole-genome sequencing revealed that resistance to antibiotics like piperacillin-tazobactam and extended-spectrum cephalosporins was due to specific genetic mutations and loss of genes, but all strains had similar virulence levels in a mouse model.
  • The research suggests that while resistance mutations can arise, they may come with fitness costs that limit their successful spread in clinical settings, potentially explaining why certain resistant types are rarely observed in practice.

Objective: Guidelines for the management of pediatric severe traumatic brain injury (TBI) recommend external ventricular drainage for CSF drainage as a first-tier treatment in the intracranial pressure (ICP) pathway. However, ventriculostomy in children can sometimes be challenging because of the small size of the lateral ventricles. External lumbar drainage (ELD) may be a useful alternative; therefore, the authors analyzed the outcome of a cohort of pediatric patients who underwent ELD to manage intracranial hypertension (ICH).

Eur J Clin Microbiol Infect Dis

November 2024

Service de Bactériologie, Assistance Publique Hôpitaux de Paris, Hôpitaux Universitaires Paris Centre, Site Cochin, 27 rue du Faubourg Saint-Jacques, Paris, 75014, France.

Article Synopsis
  • Two clinical isolates of Klebsiella michiganensis from the same patient were genetically compared, revealing differing resistance profiles: one was resistant to piperacillin-tazobactam and intermediate to cefotaxime, while the other was resistant to ceftazidime but susceptible to piperacillin-tazobactam.
  • * A mutation in the bla gene, resulting in a L169M change in the Ω loop, was identified as a key factor in the resistance differences.
  • * Cloning experiments in E. coli demonstrated that this mutation allowed for increased resistance to ceftazidime and cefepime but reduced resistance to piperacillin-tazobactam, marking a novel discovery of how

Article Synopsis
  • - The study examines the role of calcitonin gene-related peptide (CGRP) in critical COVID-19 patients, finding that levels of CGRP are elevated in both the plasma and lungs during the early stages of infection with SARS-CoV-2.
  • - CGRP and its stable analog, SAX, were found to inhibit the infection of bronchial cells by SARS-CoV-2 variants in a dose-dependent manner, suggesting that CGRP plays a protective role against the virus.
  • - The research indicates that increased CGRP levels may help in viral clearance by directly inhibiting SARS-CoV-2 replication, which opens potential avenues for CGRP-based therapies in managing COVID-19.

Background: Brain injury is one of the most serious complications after cardiac arrest (CA). To prevent this phenomenon, rapid cooling with total liquid ventilation (TLV) has been proposed in small animal models of CA (rabbits and piglets). Here, we aimed to determine whether hypothermic TLV can also offer neuroprotection and mitigate cerebral inflammatory response in large animals.

Targeting ferritinophagy impairs quiescent cancer stem cells in acute myeloid leukemia in vitro and in vivo models.

Sci Transl Med

July 2024

Centre for Translational Research in Onco-Hematology, Faculty of Medicine, University of Geneva, and Swiss Cancer Centre Leman, 1206 Geneva, Switzerland.

Article Synopsis
  • Acute myeloid leukemia (AML) is a difficult-to-treat blood cancer, mainly due to the presence of leukemic stem cells (LSCs) that lead to treatment resistance and relapse.
  • This research focuses on how the quiescence (dormancy) of LSCs and related molecular mechanisms impact AML development, revealing that quiescent LSCs have a distinct gene signature and increased autophagic activity that helps them survive.
  • The study identifies nuclear receptor coactivator 4 (NCOA4) as a key player in iron metabolism for quiescent LSCs, showing that inhibiting NCOA4 can effectively target these cells without harming normal blood progenitors, highlighting its potential as

is among the few pathogens that have not developed resistance to ß-lactam antibiotics despite decades of clinical use. The molecular basis of this long-lasting susceptibility has not been investigated, and it is not known whether specific mechanisms constrain the emergence of resistance. In this study, we first report ß-lactam tolerance due to the inactivation of the c-di-AMP phosphodiesterase GdpP.

Background: A sepsis-like syndrome is known to occur after cardiac arrest, leading to cerebral infiltration by white blood cells (WBC). We hypothesized that pharmacological sequestration of WBC, and more specifically lymphocytes within lymphoid tissues, could reduce the cerebral infiltration by these inflammatory cells and subsequent acute brain injury in a porcine model of cardiac arrest. Lymphocyte sequestration was induced by the sphingosine-1 phosphate receptors agonist fingolimod.

Article Synopsis
  • Group B Streptococcus (GBS), particularly the capsular serotype III called CC17, is a major cause of serious infections like bacteremia and meningitis in newborns, being labeled as a hypervirulent clone.
  • The study found that CC17 strains are engulfed (phagocytosed) more effectively by human monocytes and macrophages compared to non-CC17 strains, due to enhanced attachment facilitated by specific proteins (HvgA and PI-2b pilus).
  • Despite being phagocytosed more efficiently, both CC17 and non-CC17 strains survive similarly within immune cells, suggesting that CC17's enhanced phagocytosis might help it evade the immune system and persist in the

Thermodynamic consequences of a three-hour long anoxia were investigated on the isolated mammalian rat myocardium. The anoxic heart operated in a far-from-equilibrium manner as attested by the non-linearity between the thermodynamic force and the thermodynamic flow. When subjected to slight fluctuations due to anoxia, the open far-from-equilibrium cardiac system presented a thermodynamic bifurcation at ~ 60 minutes of anoxia.
