Repeat-associated non-AUG translation (RAN translation) is observed in transcripts that are causative for polyglutamine (polyQ) diseases and generates proteins with mono amino acid tracts such as polyalanine (polyA), polyleucine (polyL) and polyserine (polyS) in neurons, astrocytes and microglia. We have previously shown that microglia with aggregated polyQ led to defective differentiation and degeneration of neuron-like cells. However, it has not been determined whether only microglia containing a specific RAN product, but not other RAN products, is harmful in vitro and in vivo.

Epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitors are standard therapeutic agents for non-small cell lung cancer (NSCLC) patients with major EGFR mutations such as exon 19 deletions and a L858R mutation, whereas treatment strategies for cases with uncommon EGFR mutations remain to be fully established. Here, we report a long-term (≥20 years from initial diagnosis) NSCLC survivor carrying EGFR L858R and L747V mutations. The patient received gefitinib monotherapy, systemic chemotherapy/chemoimmunotherapy, and local consolidative therapies for oligometastatic lesions, and responded to afatinib rechallenge with a progression-free survival of 12 months.

Objective: In this study, we focused on five microRNAs (miRNAs) that have been reported to regulate phosphatase and tensin homolog deleted on chromosome 10 (PTEN) gene expression, namely miR-182, miR-183, miR-200a, miR-200b, and miR-205, and examined their relationships with PTEN protein expression in endometrial cancer tissues.

Methods: By utilizing paraffin-embedded blocks of normal endometrium (NE) and endometrial carcinoma (EC) tissue (40 cases each), we measured the expression of miRNAs by real-time PCR. Conversely, we examined PTEN protein expression by immunohistochemistry and computer-assisted image analysis.

Article Synopsis
  • - The study investigates the relationship between sarcoidosis and tumor development, analyzing data from 312 cases of sarcoidosis and noting 25 patients who were diagnosed with malignant tumors after their sarcoidosis diagnosis.
  • - Analysts found that some markers related to sarcoidosis activity, such as serum angiotensin I-converting enzyme levels and mediastinal lymph node size, decreased at the time of malignant tumor diagnosis, suggesting a potential link between decreasing sarcoidosis activity and tumor development.
  • - Patients with a history of malignant tumors tended to be older and exhibited less severe cases of sarcoidosis, indicating that prior tumors may impact the progression and treatment of the disease, particularly regarding the use of corticosteroid therapy.

Article Synopsis
  • - Sarcoidosis, a disease causing granuloma formation in various organs like the lungs, is diagnosed mainly through clinicopathologic findings with traditional markers being ACE and sIL-2R, though new specific markers are needed.
  • - This study found that serum neuron-specific enolase (NSE) levels are elevated in sarcoidosis patients and correlate positively with ACE and sIL-2R levels, with the best diagnostic sensitivity achieved when combining NSE with these traditional markers.
  • - Elevated serum NSE not only differentiates sarcoidosis from small cell lung cancer (SCLC) and other benign conditions but also reflects disease activity and treatment response, showing potential as a marker for diagnosis and monitoring of sarcoidosis.

Ketone body β-hydroxybutyrate (βOHB) and fibroblast growth factor-21 (FGF21) have been proposed to mediate systemic metabolic response to fasting. However, it remains elusive about the signaling elicited by ketone and FGF21 in the heart. Stimulation of neonatal rat cardiomyocytes with βOHB and FGF21 induced peroxisome proliferator-activated receptor α (PPARα) and PGC1α expression along with the phosphorylation of LKB1 and AMPK.

Previous reports indicate that the right ventrolateral prefrontal cortex (VLPFC) is involved in emotional regulation. However, most such studies were performed under unphysiological conditions, like the administration of transcranial direct current or repetitive transcranial magnetic stimulation. We have shown that the right VLPFC is stimulated by an acute bout of daily activity, such as cleaning.

Two types of interleukin (IL)-5 antibody biologics, anti-IL-5 antibodies (mepolizumab) and anti-IL-5α receptor antibodies (benralizumab), are indicated for severe asthma. While high-dose mepolizumab is also indicated for EGPA, benralizumab is indicated only for severe asthma. Benralizumab is characterized by antibody-dependent cell-mediated cytotoxicity activity, giving them specific and rapid anti-IL-5α receptor binding abilities and the ability to target a high number of eosinophils in tissues as well as peripheral blood.
