Background And Aims: The tumor microenvironment (TME) exerts an important role in carcinogenesis and progression. Several investigations have suggested that immune cell infiltration (ICI) is of high prognostic importance for tumor progression and patient survival in many tumors, particularly prostate cancer. The pattern of immune infiltration of PCa, on the other hand, has not been thoroughly understood.

Sci Rep

July 2024

Department of Neurosurgery, Guizhou Provincial People's Hospital, Nanming District, No.83, Zhongshan East Road, Guiyang, 550002, China.

This study aimed to assess the current status and changing trends of the disease burden of stroke and its subtypes due to low dietary fiber intake in China from 1990 to 2019. In cases of stroke and its subtypes attributable to low dietary fiber, deaths, disability-adjusted life-years (DALYs), age-standardized mortality rates (ASMR), age-standardized DALYs rates (ASDR), and percentage change were used to assess disease burden. Data were obtained from the 2019 global burden of disease study.

A novel variant c.902C>A (p. A301D) in KCNQ4 associated with non-syndromic deafness 2A in a Chinese family.

Mol Genet Genomic Med

July 2024

Department of Medical Genetics/Prenatal Diagnosis Center, Guizhou Provincial People's Hospital, Guiyang, Guizhou Province, China.

Background: Deafness autosomal dominant 2A (DFNA2A) is related to non-syndromic genetic hearing impairment. The KCNQ4 (Potassium Voltage-Gated Channel Subfamily Q Member 4) can lead to DFNA2A. In this study, we report a case of autosomal dominant non-syndromic hearing loss with six family members as caused by a novel variant in the KCNQ4 gene.

Cisplatin resistance is a major challenge for systemic therapy against advanced bladder cancer (BC). Little information is available on the regulation of cisplatin resistance and the underlying mechanisms require elucidation. Here, we detected that downregulation of the tumor suppressor, PPP2R2B (a serine/threonine protein phosphatase 2 A regulatory subunit), in BC promoted cell proliferation and migration.

Department of Human Anatomy, School of Basic Medicine, Guizhou Medical University, Gui'an New District, PR China; Key Laboratory of Human Brain Bank for Functions and Diseases of Department of Education of Guizhou Province, College of Basic Medical, Guizhou Medical University, Gui'an New District, PR China. Electronic address:

Article Synopsis
  • Spinal cord injury (SCI) presents global treatment challenges, and repetitive trans-spinal magnetic stimulation (rTSMS) is a promising therapy, though its exact recovery mechanisms are not well understood.
  • In a study using 8-week-old female mice with SCI models, researchers compared the effects of rTSMS, LY364947 (a drug), and a combination of both on functional recovery and tissue modulation.
  • Results indicated that both treatments improved recovery, reduced scar formation and inflammation, and suggested that rTSMS works by inhibiting the TGFβ1/Smad2/3 signaling pathway, highlighting its potential as a clinical treatment strategy.

Article Synopsis
  • Postoperative delirium remains common after surgery, prompting ongoing research to find effective interventions despite mixed results from trials.
  • A total of 201 trials were analyzed, revealing that trials conducted in the USA/Canada and Europe had higher odds ratios, indicating lower effectiveness compared to those from China.
  • The study emphasizes that the incidence of delirium, regional differences, and potential biases significantly influence trial outcomes, suggesting a need for better-quality global research on this issue.

Percutaneous Ultrasound-Guided Medication Injection: A Potential Technique for Subglottic Stenosis.

Respiration

September 2024

National Center for Respiratory Medicine, State Key Laboratory of Respiratory Health and Multimorbidity, National Clinical Research Center for Respiratory Diseases, Institute of Respiratory Medicine, Chinese Academy of Medical Sciences, Department of Pulmonary and Critical Care Medicine, Center of Respiratory Medicine, China-Japan Friendship Hospital, Beijing, China.

Introduction: Subglottic stenosis, manifested by granulation tissue hyperplasia, is challenging and requires multiple repeated treatments and stent maintenance at times. Corticosteroids prevent severe subglottic stenosis development owing to their antifibrotic and anti-inflammatory properties. Submucosal injection of glucocorticoids, a useful adjuvant therapeutic method, improves the mean interval between endoscopic procedures and reduces airway restenosis risks.

Although fangchinoline has been widely used as an adjunct therapy for a variety of inflammatory and cancerous diseases, its mechanism of action on tumor cells remains unclear. Fangchinoline derivative LYY-35 reduced the number of A549 cells, deformed cell morphology and increased cell debris. Cell viability was significantly reduced, while the same concentration of LYY-35 had little effect on BEAS-2B viability of normal lung epithelial cells.

Glioblastoma (GBM) is a primary intracranial malignant tumor with the highest mortality and morbidity among all malignant central nervous system tumors. Tanshinone IIA is a fat-soluble active ingredient obtained from Salvia miltiorrhiza, which has an inhibitory effect against various cancers. We designed and synthesized a novel L-shaped ortho-quinone analog TE5 with tanshinone IIA as the lead compound and tested its antitumor activity against GBM.

Background: Growing evidence highlights the role of the spleen-brain axis in inflammation-associated depression. The α7-subtype of nicotinic acetylcholine receptor (α7 nAChR, encoded by the Chrna7 gene) is implicated in systemic inflammation, with Chrna7 knock-out (KO) mice displaying depression-like behaviors. Yet, the influence of spleen nerve on depression-like behaviors in these KO mice remains to be elucidated.

Multiple myeloma (MM) is a form of clonal plasma cell malignancy that associates with clinical manifestations such as anemia, hypercalcemia, bone pain, and renal impairment. Approximately 20-50% of MM patients at initial diagnosis experience renal injury, a vital complication that significantly influences prognosis and quality of life. This review seeks to clarify the multifaceted mechanisms of renal injury in MM, scrutinizing the pathogenic role of monoclonal proteins, the impact of hypercalcemia, and direct renal infiltration by plasma cells.

December 2024

State Key Laboratory of Experimental Hematology, National Clinical Research Center for Blood Diseases, Haihe Laboratory of Cell Ecosystem, Institute of Hematology and Blood Diseases Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College, Tianjin 300020.

Article Synopsis
  • The study assessed flumatinib for treating Chinese patients with Philadelphia chromosome-positive chronic-phase chronic myeloid leukemia (CP-CML) who previously failed tyrosine kinase inhibitors (TKIs).
  • Among 336 patients, flumatinib showed high response rates, with 86.4% achieving complete hematologic response and significant cytogenetic and molecular responses, particularly in those without prior TKI resistance.
  • Adverse events were manageable and similar to first-line therapies, suggesting flumatinib is a promising option for TKI-resistant or intolerant CP-CML patients, especially those without prior resistance to second-generation TKIs.

Tissue engineering of small-diameter vessels remains challenging due to the inadequate ability to promote endothelialization and infiltration of smooth muscle cells (SMCs). Ideal vascular graft is expected to provide the ability to support endothelial monolayer formation and SMCs infiltration. To achieve this, vascular scaffolds with both orientation and dimension hierarchies were prepared, including hierarchically random vascular scaffold (RVS) and aligned vascular scaffold (AVS), by utilizing degradable poly(ε-caprolactone)-co-poly(ethylene glycol) (PCE) and the blend of PCE/gelatin (PCEG) as raw materials.
