Purification and characterization of two grandiuvarones from leaves and their antimicrobial activities.

A novel aromatic compound, grandiuvarone B (5-acetoxy-3-benzoyloxymethyl-5H-oxepin-4-one), along with a known compound grandiuvarone A (5-acetoxy-6-benzoyloxymethyl-5H-oxepin-4-one) were isolated from methanol extracts of leaves. Their structures were determined by various spectroscopic techniques including nuclear magnetic resonance (NMR), high-resolution electrospray ionisation mass spectrometry (HR-ESI-MS) and circular dichroism (CD). Grandiuvarone A and grandiuvarone B are isomers and the configuration of grandiuvarone B was reported for the first time.

Pre-column dilution large volume injection (PD-LVI), a novel sample injection technique for reverse phase ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS), was developed in this study. The PD-LVI UHPLC-MS/MS system was designed by slightly modifying the commercial UHPLC-MS/MS equipment with a mixer chamber. During the procedure of PD-LVI, sample solution of 200μL was directly carried by the organic mobile phase to the mixer and diluted with the aqueous mobile phase.

Four spiroalkaloids, including a new compound shensongine A (1), were isolated from the anti-arrhythmic TCM formula Shensong Yangxin capsule. Their structures were determined on the basis of spectroscopic analysis. Compounds 1 and 3 displayed cardiovascular activities by shortened APD in rat myocardial cells.

In this study, we purified and characterized the procoagulant protein FV-2 from Daboia russelli siamensis (Myanmar) venom using ion-exchange chromatography on CM-Sephadex C-50 and gel filtration on Superdex G-75 column. The activation of factor X and prothrombin was determined, respectively, by specific chromogenic substrates. The fibrinogen-clotting activity, thermal stability, and pH stability were also determined.

Refractory chronic graft-versus-host disease (cGVHD) is a significant complication resulting from allogeneic hematopoietic stem cell transplantation (HSCT). Mesenchymal stromal cells (MSCs) have shown promise for treating refractory cGVHD, but the favorable effects of MSCs therapy in cGVHD are complex and not fully understood. In this prospective clinical study, 20 of 23 cGVHD patients had a complete response or partial response in a 12-month follow-up study.

Objective: To provide identification basis for distinguishing Elephantopi Herba and its adulterants.

Methods: Light microscopy (LM), scanning electron microscopy (SEM) and thin layer chromatography scanning (TLCS) methods were used to identify Didancao (Elephantopus scaber) and its adulterants Baihuadidancao (Elephantopus mollis) and Jiadidancao (Pseudelephantopus spicatus).

Results: Based on the microscopic features and TLC profiles, the commercial Didancao samples retailed in mainland China, Taiwan, Hong Kong and Macau markets were identified as Elephantopus scaber.

Contribution of serum IL-4 and IgE to the early prediction of allergic reactions induced by chlorogenic acid.

Chlorogenic acid (CA) is one of the active ingredients in some Chinese herbal injections, which may cause allergic reactions in clinic therapy. However, the criterion of test for allergen had not been employed in current Pharmacopeia of United States, European Pharmacopeia, Japanese Pharmacopeia and British Pharmacopeia. In order to find a new way to predict allergic reactions induced by CA earlier, the guinea pigs were sensitized successively by injecting CA intravenously once a day for three times, the results were compared that of Chinese Pharmacopeia by injecting CA intraperitoneally once every other day for three times, serum IL-4 and total IgE were detected by method of enzyme linked immunosorbent assay (ELISA) before guinea pigs were challenged once by injecting the same drug intravenously.
