Proteins typically adopt a single fold to carry out their function, but metamorphic proteins, with multiple folding states, defy this norm. Deciphering the mechanism of conformational interconversion of metamorphic proteins is challenging. Herein, we employed nuclear magnetic resonance (NMR), circular dichroism (CD), and all-atom molecular dynamics (MD) simulations to elucidate the mechanism of fold switching in proteins GA95 and GB95, which share 95% sequence homology.

Two new stilbenes, denominated Cajanotone B (CAB) and Cajanotone C (CAC), were isolated from the leaves of Cajanus cajan. In this study, the structures of CAB and CAC were unambiguously elucidated by a combination of various spectral methods. Both compounds significantly inhibited the adipogenesis in 3T3-L1 adipocytes by reducing the lipid accumulation, triglyceride content and FFA secretion.

Previous studies in small samples have identified inconsistent cortical abnormalities in major depressive disorder (MDD). Despite genetic influences on MDD and the brain, it is unclear how genetic risk for MDD is translated into spatially patterned cortical vulnerability. Here, we initially examined voxel-wise differences in cortical function and structure using the largest multi-modal MRI data from 1660 MDD patients and 1341 controls.

Soil moisture (SM) is essential for sustaining services from Earth's critical zone, a thin-living skin spanning from the canopy to groundwater. In the Anthropocene epoch, intensive afforestation has remarkably contributed to global greening and certain service improvements, often at the cost of reduced SM. However, attributing the response of SM in deep soil to such human activities is a great challenge because of the scarcity of long-term observations.

Four novel new isocoumarins, cajanolactone B, C, D1 and D2 (1-4), were isolated from ethanolic extracts of the leaves of . The structural elucidation has been completed mainly depending on extensive spectroscopic analysis including UV, IR, NMR (1D and 2D), HRESIMS and chiral analysis. Notably, all these new isocoumarins were found to exist in racemic forms, among which compounds 3 and 4 share the same planar structure.

Various phosphorus (P)-inactivating materials with a strong capability of immobilizing P in sediment have been developed for lake geoengineering purposes to control internal P pollution. However, unsatisfactory applications have raised concerns about the reliability of the method. This study hypothesized that P migration from sediment to material is a key process regulating the immobilization, which is often neglected by common assessment procedures that assume that the material is closely in contact with sediment (e.

The importance of controlling internal phosphorus (P) pollution in lakes has been recognized by scientists, and the application of P-inactivating materials to immobilize sediment P is often considered. However, sediment resuspension, a typical physical process occurring in lakes, has been demonstrated to increase the uncertainty of immobilization. In this study, we explored the characteristics of P immobilization in the horizontal direction under the effects of resuspension using annular flume tests based on drinking water treatment residuals (DWTR).

Unlabelled: Flowering time is one of the most fundamental factors that determine the distribution and final yield of rice. Ehd1 (Early heading date 1) is a B-type response regulator which functions as a flowering time activator. Although diverse flowering time genes have been reported as regulatory factors of expression, the potential regulators of largely remain to be identified.

Orphan nuclear receptor Nurr1 plays important roles in the progression of various diseases, including Parkinson's disease, neuroinflammation, Alzheimer's disease, and multiple sclerosis. It can recognize DNA as a monomer or heterodimer with retinoid X receptor α (RXRα). But the molecular mechanism of its transcriptional activity regulation is still largely unknown.

In this study, the phytoremediation potentials and invasive risks of an exotic invasive plant (Xanthium strumarium, LT), a native plant (X. sibiricum, CR), and combinations of inoculations (EG, with CR as the scion and LT as the rootstock; SG, with CR as both the scion and rootstock) were evaluated on Cd/Cu/Ni tailings.

As a subgroup of sorting nexins (SNXs) that contain regulator of G protein signaling homology (RH) domain, SNX-RH proteins, including SNX13, SNX14 and SNX25, were proposed to play bifunctional roles in protein sorting and GPCR signaling regulation. However, mechanistic details of SNX-RH proteins functioning via RH domain remain to be illustrated. Here, we delineate crystal structures of the RH domains of SNX13 and SNX25, revealing a homodimer of SNX13 RH domain mediated by unique extended α4 and α5 helices, and a thiol modulated homodimer of SNX25-RH triggered by a unique cysteine on α6 helix.

α-Synuclein (α-syn) is the main protein component of Lewy bodies, the major pathological hallmarks of Parkinson's disease (PD). C-terminally truncated α-syn is found in the brain of PD patients, reduces cell viability and tends to form fibrils. Nevertheless, little is known about the mechanisms underlying the role of C-terminal truncation on the cytotoxicity and aggregation of α-syn.

Lake restoration (typically sediment dredging) commonly involves producing a new sediment-water interface (SWI). This study comprehensively investigated the migration and transformation of P during the formation of a new SWI under different dissolved oxygen (DO) levels in the overlying water, based on Fe/Al-rich sediment. The results suggest that DO had a profound effect on the 0-7 cm sediment layer properties and higher DO levels in the overlying water resulted in the diffusion of DO deeper into the sediments.

Autophagy-mediated mitochondrial degradation plays pivotal roles in both the acquisition and maintenance of pluripotency, but the molecular mechanisms that link autophagy-mediated mitochondrial homeostasis to pluripotency regulation are unclear. Here, we identified that the mitophagy receptor BNIP3 regulates pluripotency. In mouse ESCs, depletion of BNIP3 caused accumulation of aberrant mitochondria accompanied by decreased mitochondrial membrane potential, increased production of reactive oxygen species (ROS), and reduced ATP generation, which led to compromised self-renewal and differentiation.

As an important component of carbonaceous aerosols (CA), organic carbon (OC) exerts a strong, yet insufficiently constrained perturbation of the climate. In this study, we reported sources of OC based on its natural abundance radiocarbon (C) fingerprinting in aerosols and water-insoluble organic carbon (WIOC) in snowpits across the Tibetan Plateau (TP) - one of the remote regions in the world and a freshwater reservoir for billions of people. Overall, the proportions from C-based non-fossil fuel contribution (f) for OC in aerosols was 74 ± 10%, while for WIOC in snowpits was 81 ± 10%, both of which were significantly higher than that of elemental carbon (EC).

Lake geoengineering with phosphorus (P)-inactivating materials to reduce sediment P loading is often used for eutrophication control. The redistribution of materials in sediment, especially those induced by resuspension, is reportedly a common phenomenon during practical applications, which may interfere with the pollution control. Notably, a recent study by the authors initially found that the heterogeneous properties of materials and sediments varied the P immobilization in different sized sediments which exhibited diverse movement characteristics.
