Department of Cell Physiology, Graduate School of Medicine, Nagoya University, Nagoya 466-8550, Japan. Electronic address:

Sensorineural hearing loss causes cell death in central auditory neurons, but molecular mechanisms of triggering this process are not fully understood. We report here that loss of afferent activity promotes cell death by facilitating proBDNF-p75NTR signals in cochlear nucleus of chicks around hatch. RNA-seq analyses revealed up-regulation of genes related to proBDNF-p75NTR-JNK signals as well as apoptosis at the nucleus within 24hours after unilateral cochlea deprivation.

Alexithymia is a psychological trait characterized by difficulty expressing emotions. Previous studies reported that individuals with higher alexithymia have a decreased sense of interoception, which is the sense of monitoring and controlling internal organs. Thus, we hypothesized that internal organ activity (cardiac activities in the present study) was easily affected by false feedback in individuals with severe alexithymia.

Nonapoptotic caspase-3 guides C1q-dependent synaptic phagocytosis by microglia.

Nat Commun

January 2025

Department of Translational Neurobiology, National Institute of Neuroscience, National Center of Neurology and Psychiatry, Kodaira, Tokyo, 187-8502, Japan.

Caspases are known to mediate neuronal apoptosis during brain development. However, here we show that nonapoptotic activation of caspase-3 at presynapses drives microglial synaptic phagocytosis. Real-time observation and spatiotemporal manipulation of synaptic caspase-3 in the newly established, mouse-derived culture system demonstrate that increased neuronal activity triggers localized presynaptic caspase-3 activation, facilitating synaptic tagging by complements.

Neuronal processing of external sensory input is shaped by internally generated top-down information. In the neocortex, top-down projections primarily target layer 1, which contains NDNF (neuron-derived neurotrophic factor)-expressing interneurons and the dendrites of pyramidal cells. Here, we investigate the hypothesis that NDNF interneurons shape cortical computations in an unconventional, layer-specific way, by exerting presynaptic inhibition on synapses in layer 1 while leaving synapses in deeper layers unaffected.

Background: F-8-coil repetitive transcranial magnetic stimulation (rTMS) and H-1-coil deep repetitive transcranial magnetic stimulation (dTMS) have been indicated for the treatment of major depressive disorder (MDD) in adult patients by applying different treatment protocols. Nevertheless, the evidence for long-term electrophysiological alterations in the cortex following prolonged TMS interventions, as assessed by quantitative electroencephalography (qEEG), remains insufficiently explored. This study aims to demonstrate the qEEG-based distinctions between rTMS and dTMS in the management of depression and to evaluate the potential correlation between the electrophysiological changes induced by these two distinct TMS interventions and the clinical improvement in depressive and anxiety symptoms.
