Accelerometry: a technique for quantifying movement patterns during walking.

The popularity of using accelerometer-based systems to quantify human movement patterns has increased in recent years for clinicians and researchers alike. The benefits of using accelerometers compared to more traditional gait analysis instruments include low cost; testing is not restricted to a laboratory environment; accelerometers are small, therefore walking is relatively unrestricted; and direct measurement of 3D accelerations eliminate errors associated with differentiating displacement and velocity data. However, accelerometry is not without its disadvantages, an issue which is scarcely reported in gait analysis literature.

Strength training improves supramaximal cycling but not anaerobic capacity.

This study examined supramaximal cycling performed to exhaustion at 120% of peak O(2) uptake (120% VO(2)peak) before and after 8 weeks of strength training. Eight previously untrained men completed 8 weeks of leg-strength training 3 days week(-1) on a hack-squat machine; four sets, five repetitions at 85% of one repetition maximum each session. Anaerobic capacity was quantified by determining the maximal accumulated O(2) deficit during supramaximal cycling.

Threat interpretation bias as a vulnerability factor in childhood anxiety disorders.

The present study examined threat interpretation biases in children 7-12 years of age with separation, social and generalised anxiety disorders (N=15), non-anxious offspring at risk due to parental anxiety (N=16) and non-anxious controls of non-anxious parents (N=14). Children provided interpretations of ambiguous situations to assess cognitive, emotional and behavioural responses. In comparison with non-anxious control children and at-risk children who did not differ from each other, anxious children reported stronger negative emotion and less ability to influence ambiguous situations.

This study examined graduating Bachelor of Nursing (BN) students' perceptions of a Web-enhanced learning environment, their computer literacy skills, and use of technology, and how these influenced their satisfaction. This Australian survey produced a 64% (n = 170) response rate.

Energy turnover for Ca2+ cycling in skeletal muscle.

The majority of energy consumed by contracting muscle can be accounted for by two ATP-dependent processes, cross-bridge cycling and Ca(2+) cycling. The energy for Ca(2+) cycling is necessary for contraction but is an overhead cost, energy that cannot be converted into mechanical work. Measurement of the energy used for Ca(2+) cycling also provides a means of determining the total Ca(2+) released from the sarcoplasmic reticulum into the sarcoplasm during a contraction.

Matrix metalloproteinase localisation by in situ-RT-PCR in archival human breast biopsy material.

Utilising archival human breast cancer biopsy material we examined the stromal/epithelial interactions of several matrix metalloproteinases (MMPs) using in situ-RT-PCR (IS-RT-PCR). In breast cancer, the stromal/epithelial interactions that occur, and the site of production of these proteases, are central to understanding their role in invasive and metastatic processes. We examined MT1-MMP (MMP-14, membrane type-1-MMP), MMP-1 (interstitial collagenase) and MMP-3 (stromelysin-1) for their localisation profile in progressive breast cancer biopsy material (poorly differentiated invasive breast carcinoma (PDIBC), invasive breast carcinomas (IBC) and lymph node metastases (LNM)).

The terminal sugar sialic acid (Sia) plays a pivotal role in cell-cell interaction and recognition. A prerequisite for the biosynthesis of sialoglycoconjugates is the activation of Sia to cytidine monophosphate-Sia (CMP-Sia), by CMP-Sia synthetases (CMP-Sia-syn). CMP-Sia-syn are conserved from bacteria to man, and have been found to reside in the nucleus of all vertebrate species analysed to date.

We report an easy and direct application of 'Saturation Transfer Double Difference' (STDD) NMR spectroscopy to identify ligands that bind to a Sepharose-immobilised target protein. The model protein, cytidine 5'-monophosphate sialic acid (CMP-Sia) synthetase, was expressed as a Strep-Tag II fusion protein and immobilised on Strep-Tactin Sepharose. STD NMR experiments of the protein-enriched Sepharose matrix in the presence of a binding ligand (cytidine 5'-triphosphate, CTP) and a non-binding ligand (alpha/beta-glucose) clearly show that CTP binds to the immobilised enzyme, whereas glucose has no affinity.

Foot stamping, progressively loaded squats, and in-line dancing positively influence proximal femoral bone mineral density, lower extremity strength, and static and dynamic balance.

Introduction: Foot stamping, squats exercises, and in-line dancing together create a suitable activity program for sedentary, independent-living older women.

The effect of manipulating sediment pH on the porewater chemistry of copper- and zinc-spiked sediments.

Spiking of sediment with metal cations that readily hydrolyse causes the sediment pH to decrease. Displaced iron and manganese also oxidise and hydrolyse, further lowering sediment pH. The lower pH of metal-spiked sediments requires a subsequent sediment neutralisation.

As part of our research interest directed toward the development of antimycobacterial agents, we have investigated compounds based on galactofuranose (Galf), an essential cell wall component of mycobacteria. The objective of this study was to explore structure activity relationships of Galf thioglycosides with straight chain and branched aglycons. Acylated Galf 9-heptadecyl thioglycoside was prepared by Lewis acid-catalyzed thioglycosidation of 1,2,3,5,6-penta-O-acyl-D-galactofuranose with 9-heptadecanethiol, and subsequently converted to the corresponding sulfone using m-CPBA.

Inverse relations in the patterns of muscle and center of pressure dynamics during standing still and movement postures.

The aim of this study was to investigate the postural center of pressure (COP) and surface muscle (EMG) dynamics of young adult participants under conditions where they were required to voluntarily produce random and regular sway motions in contrast to that of standing still. Frequency, amplitude and regularity measures of the COP excursion and EMG activity were assessed, as were measures of the coupling relations between the COP and EMG outputs. The results demonstrated that, even when standing still, there was a high degree of regularity in the COP output, with little difference in the modal frequency dynamics between standing still and preferred motion.

Detection of intermittent sewage pollution in a subtropical, oligotrophic, semi-enclosed embayment system using sterol signatures in sediments.

A field study was conducted to investigate sewage inputs at popular anchorages in Moreton Bay, a sub-tropical, semienclosed embayment system in Southeast Queensland, Australia. Sterol biomarkers were quantified in sediments revealing low levels over a spatial and temporal scale consistent with a shallow, oligotrophic, highly dynamic, sand dominated system. Despite low concentrations (ng/g) and high variability, relevant sterol/stanol pairs remained well-correlated and were successful in identifying an unexpected once-off pollution event from a point source at Moreton Bay Island.

Galectin-3 is a multifunctional carbohydrate-binding protein that has roles in cancer progression. In addition to carbohydrate-dependent extracellular functions, galectin-3 participates in carbohydrate-independent intracellular signalling pathways, including apoptosis, via protein-protein interactions, some of which engage the carbohydrate-binding groove. When ligands bind within this site, conformational rearrangements are induced and information on unliganded galectin-3 is therefore valuable for structure-based drug design.

The recent emergence of clinically oppressive superbugs, some with resistance to nearly all frontline drug therapies, has challenged our ability to combat such infectious organisms as Mycobacterium tuberculosis, the causative agent of tuberculosis (TB). Our medicinal chemistry program targeting this pathogen has identified several potent galactofuranose-based in vitro inhibitors of mycobacterial growth. The most potent compound, the Galf N,N-didecyl sulfenamide 8d, displayed anti-mycobacterial activity (MIC) of 1 microg/mL in a cell based assay against a representative strain of Mycobacterium smegmatis.
