A monoclonal antibody against GPNMB.

As a melanosome-associated transmembrane glycoprotein, GPNMB plays an important role in numerous cell types, as well as in tumors. Producing a high specificity and affinity monoclonal antibody against human GPNMB provides an important tool to study the function of GPNMB protein. In this study, monoclonal antibodies to GPNMB were obtained by immunizing BALB/c mice with purified GST-GPNMB emulsified in Freund's adjuvant.

Reduction in nerve root compression by the nucleus pulposus after Feng's Spinal Manipulation.

Ninety-four patients with lumbar intervertebral disc herniation were enrolled in this study. Of these, 48 were treated with Feng's Spinal Manipulation, hot fomentation, and bed rest (treatment group). The remaining 46 patients were treated with hot fomentation and bed rest only (control group).

Effects of zishentongluo in patients with early-stage diabetic nephropathy.

The purpose of this study was to assess the efficacy of zishentongluo (ZSTL) for the treatment of diabetic nephropathy (DN) and its related mechanisms. Forty-five patients with DN were randomized to receive either ZSTL (n = 25) or benazepril (n = 20), an angiotensin converting enzyme inhibitor, for 12 weeks. Conventional biochemical tests were performed to determine fasting blood glucose (FBG), glycated hemoglobin (HbA1c), serum creatinine (SCr), endogenous creatinine clearance rate (Ccr), total cholesterol (TC), and triglyceride (TG) levels.

Characterization of cancer stem-like cells in the side population cells of human gastric cancer cell line MKN-45.

Objective: Side population (SP) cells may play a crucial role in tumorigenesis and the recurrence of cancer. Many kinds of cell lines and tissues have demonstrated the presence of SP cells, including several gastric cancer cell lines. This study is aimed to identify the cancer stem-like cells in the SP of gastric cancer cell line MKN-45.

Clinical analysis of the treatment of spinal cord injury with umbilical cord mesenchymal stem cells.

Background Aims: The purpose of this study was to observe the clinical effect and safety of umbilical cord mesenchymal stem cells (UC-MSCs) in treating spinal cord injury (SCI) by intrathecal injection.

Methods: From January 2008 to October 2010, we treated 22 patients with SCI with UC-MSCs by intrathecal injection; dosage was 1 × 10(6) cells/kg body weight once a week given four times as a course. Four patients received two courses, one patient received three courses and all other patients received one course.

Silencing of GPNMB by siRNA inhibits the formation of melanosomes in melanocytes in a MITF-independent fashion.

Background: Melanosomes are specialized membrane-surrounded organelles, which are involved in the synthesis, storage and transport of melanin. Glycoprotein (transmembrane) non-metastatic melanoma protein b (GPNMB), a melanosome-specific structural protein, shares significant amino acid sequence homology with Pmel-17. Proteomic analysis demonstrated that GPNMB is present in all stages (I-IV) of melanosomes.

Cotransplantation of allogeneic mesenchymal and hematopoietic stem cells in children with aplastic anemia.

We report here the preliminary results of allogeneic hematopoietic stem cell transplantation with mesenchymal stem cells (MSCs) for 6 cases of severe aplastic anemia. The patients ranged in age from 3 to 16 years, and the median time from diagnosis to transplantation was 32 months (range: 3-156 months). The conditioning regimens consisted of fludarabine, cyclophosphamide, and antithymocyte globulin with or without busulfan.

Objective: The purpose of this study was to investigate whether grape seed proanthocyanidin extract (GSPE) can provide photoprotection against ultraviolet (UV) irradiation.

Background Data: Study has shown that GSPE is a natural oxidant, and is used in many fields such as ischemia-reperfusion injury, chronic pancreatitis, and even cancer. However, the effect of GSPE on UV irradiation is as yet unknown.

Topical application of aloperine improves 2,4-dinitrofluorobenzene-induced atopic dermatitis-like skin lesions in NC/Nga mice.

Aloperine has been shown to inhibit 2,4-dinitrofluorobenzene (DNFB) induced allergic contact dermatitis in BALB/c mice. In the present study, we further investigated the effect of aloperine on DNFB-induced atopic dermatitis-like skin lesions in NC/Nga mice. NC/Nga mice elicited atopic dermatitis-like skin lesions after the topical application of DNFB.

[Pharmacokinetics of epirubicin hydrochloride long-circulating thermosensitive liposomes in rat plasma].

To develop and validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of epirubicin hydrochloride (EPI) in rat plasma, daunorubicin hydrochloride was used as internal standard. The plasma samples were deproteinated with methanol, and separation was performed on a reversed-phase CAPCELL PAK C18 column (3.0 mm x 50 mm, 3 microm).

A novel missense mutation of the ATP2C1 gene in a Chinese patient with Hailey-Hailey disease.

Benign familial chronic pemphigus (Hailey-Hailey disease, HHD; MIM 169600) is a rare autosomal dominant hereditary disorder characterized by pruritic vesicles, painful erosions and scaly erythematous plaques at the sites of friction and flexures. Mutations in ATP2C1, which encoding the human secretory pathway Ca²(+)/Mn²(+)-ATPase protein 1 (hSPCA1), have been identified as the pathogenic gene of HHD. We found a novel, distinct, heterozygous mutation during study of a Chinese patient with HHD.

Epirubicin-encapsulated long-circulating thermosensitive liposome improves pharmacokinetics and antitumor therapeutic efficacy in animals.

In the present work, a long-circulating epirubicin hydrochloride (EPI)-containing thermosensitive liposome aiming at antitumor therapy, DPPC/MSPC/DSPG/DSPE-mPEG(2000) (EPI-LTSL), was developed and evaluated. Nonthermosensitive and traditional liposomes, HSPC/cholesterol/DSPG/DSPE-mPEG(2000) (EPI-NTSL) and HSPC/cholesterol (EPI-LIP), were also prepared at the same time for comparison. Temperature-dependent EPI release from loaded liposomes in vitro was characterized by the fluorescence method.

Effects and mechanisms of aloperine on 2, 4-dinitrofluorobenzene-induced allergic contact dermatitis in BALB/c mice.

Allergic contact dermatitis is a prototypic T-cell-mediated cutaneous inflammatory response. Multiple cell types, inflammatory mediators and cytokines are involved in the regulation of immunologic and inflammatory processes in allergic contact dermatitis. Aloperine is an isolated alkaloid found in the plant of Sophora alopecuroides L.

[Clinical observation for the management of chronic hypertrophic rhinitis by endoscopic ethmoid and maxillary surgery].

Objective: To evaluate the therapeutic effect of endoscopic ethmoid and maxillary surgery on chronic hypertrophic rhinitis.

Method: A total of 54 cases of chronic hypertrophic rhinitis were treated by endoscopic ethmoid and maxillary surgery between 2003 and 2004, undergoing postoperative follow-up of more than one year. Age of patients ranged from 17 to 60 years, with a mean of 35 years.

Haploidentical hematopoietic stem cell transplantation in child hematologic malignancies with G-CSF-mobilized marrow grafts without T-cell depletion: a single-center report of 45 cases.

In this report, the authors describe a protocol for haploidentical bone marrow transplantation in children who received G-CSF-mobilized bone marrow grafts without T-cell depletion from HLA-mismatched parents. Forty-two of 45 patients achieved complete donor hematopoietic engraftment; the medium time for neutrophil and platelet recovery was 17 and 19 days, respectively. Three died of early transplantation-associated complications; other causes of death included relapse (11 cases), fungal pneumonia (5), and acute graft-versus-host disease (2).

A retropharyngeal lipoma causing obstructive sleep apnea in a child.

Background: Lipomas are common benign tumors, but they rarely occur in the retropharyngeal space and even more rarely occur in children.

Case Report: We report the case of an 11-year-old girl with a 5-year history of progressively worsening nasal obstruction, snoring, and excessive daytime sleepiness. Physical examination revealed a child with a body mass index of 16.

Upregulation of dentin matrix protein 1 promoter activities by core binding factor alpha1 in human dental pulp stem cells.

An osteoblast-specific transcription factor, core binding factor alpha1 (Cbfalpha1), is critical for osteoblast and odontoblast differentiation. In this study, the role of Cbfalpha1 in human dentin matrix protein1 (DMP1) gene expression in human dental pulp stem cells (HDPSCs) was investigated. The desired promoter fragments were obtained and cloned into the pGL3-basic vector.

Method: Case studies of Meniere's disease were carried out in 22 pilots.

Result: There were vertigo (n=22), tinnitus (n=21), abnormal hearing (n=19) and abnormal vestibular function (n=11) in 22 pilots.
