Article Synopsis
  • - The study investigates the effects of mitochondrial transplantation (MT) after cardiopulmonary resuscitation (CPR) on microglia/macrophages (MG/MΦ) and neurological function in rats.
  • - The results indicated that the group receiving mitochondrial transplants (Mito) showed improved mitochondrial structure in MG/MΦ, reduced neuroinflammation, less neuronal apoptosis, and better neurological scores compared to other groups.
  • - The findings suggest that exogenous MT may enhance brain protection following CPR by facilitating the polarization of MG/MΦ towards a beneficial M2-type, indicating potential for therapeutic strategies in post-resuscitation care.

Monocyte/macrophage pyroptosis and C5b-9-induced cyst enlargement in Pkd1-/- mice.

Nephrol Dial Transplant

November 2024

Kidney Institution of the Chinese People's Liberation Army, Chang Zheng Hospital, the Navy Military Medical University, Shanghai, China.

Article Synopsis
  • C5b-9 levels are higher in autosomal dominant polycystic kidney disease (ADPKD), and its role in cyst growth is not fully understood.
  • Experimental methods involved treating different groups of chronic-onset Pkd1-/- mice with C5b-9, anti-C9 antibodies, or control substances and analyzing the effects on macrophages and cyst development.
  • Results indicated that C5b-9 induces the death of specific immune cells (Ly6C+ monocytes/macrophages), which promotes kidney cell growth and contributes to the worsening of cysts in the disease.

Background: Patients with chronic heart failure (CHF) and type 2 diabetes mellitus (DM) are prone to insulin resistance and malnutrition, both of which are significant prognostic factors for CHF. However, the combined effect of the triglyceride-glucose index (TyG index) and prognostic nutritional index (PNI) on the mortality risk in patients with CHF and type 2 DM has not yet been studied.

Methods: We enrolled 3,315 patients with CHF and type 2 DM.

COVID-19 may induce a state of hypercoagulability, particularly in critically ill patients, for reasons that remain unknown. Numerous studies have identified the presence of antiphospholipid antibodies in patients with COVID-19; however, the definitive diagnosis of antiphospholipid syndrome continues to pose challenges. Here, we present the case of a patient infected with SARS-CoV-2 who developed life-threatening severe thrombocytopenia, profound anaemia, acute pulmonary hypertension, right ventricular failure, and renal insufficiency.

Acute organ injuries, such as acute kidney injury (AKI) and acute liver injury (ALI), usually present high morbidity and mortality in patients. However, the current clinical treatments remain limited, especially the lack of effective drug-based treatment. Since these acute injuries are often associated with reactive oxygen species (ROS) overproduction, it is a promising strategy to develop therapeutic agents with potent ROS scavenging ability and excellent biocompatibility for efficient antioxidation therapy.

Background: Increasing evidence suggests that serum urea nitrogen may be a risk factor for prostate cancer (PCa) and influence serum prostate-specific antigen (PSA) concentrations, but direct evidence of a relationship between PSA and serum urea nitrogen levels in the general population is still lacking. The aim of this study was to demonstrate the relationship between serum urea nitrogen levels and prostate-specific antigen (PSA) and prostate cancer.

Methods: We conducted a cross-sectional study using the National Health and Nutrition Examination Survey (NHANES, 2003-2010) database.

Cisplatin, the pioneering heavy metal compound, stands out as a potent drug for the treatment of various solid tumors. However, its clinical utility is hampered by notable toxicity and adverse effects, particularly nephrotoxicity. The potency of rutecarpine, a phytochemical, in mitigating cisplatin-induced nephrotoxicity was assessed in the present study.

Objective: To study the role of transforming growth factor beta 1 (TGF-β1) and tumor necrosis factor receptor related factor 6 (TRAF6) in the progression of epithelial mesenchymal transformation (EMT) in allergic rhinitis (AR).

Methods: A total of 30 patients underwent nasal endoscopic surgery at our Hospital were selected for 15 patients in each group based on their allergy status. Inferior turbinate mucosa tissue was obtained and analyzed using immunohistochemical (IHC) tests, real-time quantitative PCR (qRT-PCR) detection, and Western blotting (WB) tests to measure TGF-β1, TRAF6, E-cadherin, Vimentin, and α-Smooth Muscle Actin (α-SMA) expression levels.

Background: First-pass successful reperfusion (FPSR), defined as a successful/complete reperfusion achieved after a single thrombectomy pass, is predictive of favorable outcome in patients with acute ischemic stroke with large-vessel occlusion. It is unknown whether intravenous tirofiban is effective in increasing the rate of FPSR in acute anterior large-vessel occlusion stroke.

Methods And Results: Patients who had acute large-vessel occlusion stroke presenting within 24 hours and underwent endovascular thrombectomy were analyzed from the RESCUE BT (Intravenous Tirofiban for Patients With Large Vessel Occlusion Stroke) clinical trial, of which the main analysis was neutral.

Atherosclerosis (AS) is a chronic vascular disease primarily affecting large and medium-sized arteries and involves various complex pathological mechanisms and factors. Previous studies have demonstrated a close association between atherosclerosis and inflammatory damage, metabolic disorders, and gut microbiota. It is also closely linked to several cellular processes, such as endothelial cell pyroptosis, ferroptosis, mitophagy, mitochondrial dynamics, and mitochondrial biogenesis.

Rationale: Kidney inflammation plays a crucial role in the pathogenesis of IgA nephropathy (IgAN), yet the specific phenotypes of immune cells involved in disease progression remain incompletely understood. Utilizing joint profiling through longitudinal single-cell RNA-sequencing (scRNAseq) and single-cell assay for transposase-accessible chromatin sequencing (scATACseq) can provide a comprehensive framework for elucidating the development of cell subset diversity and how chromatin accessibility regulates transcription.

Objective: We aimed to characterize the dynamic immune cellular landscape at a high resolution in an early IgAN mouse model with acute kidney injury (AKI).

Department of Nephrology, First Medical Center of Chinese PLA General Hospital, Nephrology Institute of the Chinese People's Liberation Army, State Key Laboratory of Kidney Diseases, National Clinical Research Center for Kidney Diseases Beijing Key Laboratory of Kidney Disease Research Beijing China.

IgA nephropathy (IgAN) is the most prevalent primary glomerulonephritis, resulting in end-stage renal disease and increased mortality rates. Prognostic biomarkers reflecting molecular mechanisms for effective IgAN management are urgently needed. Analysis of kidney single-cell transcriptomic sequencing data demonstrated that IgAN expressed high-expression levels of inflammatory cytokines TNFSF10, TNFSF12, CCL2, CXCL1, and CXCL12 than healthy controls (HCs).

Article Synopsis
  • - This study investigated the role of lncRNA XIST in chronic obstructive pulmonary disease (COPD) and chronic pulmonary heart disease (CPHD), identifying it as a potential biomarker for screening and prognosis.
  • - A total of 127 COPD patients were compared to 76 healthy individuals, revealing that XIST levels were significantly higher in COPD patients, especially those with acute exacerbations and CPHD.
  • - The findings suggest that elevated XIST levels correlate with greater pulmonary complications and can indicate a poor 3-year prognosis for COPD patients, particularly those also suffering from CPHD.
