The purpose of this study was to evaluate percutaneous penetration and arrhythmogenic effects of aconitine after transdermal administration, compared with the oral route. Skin penetration of aconitine was tested by a microdialysis technique in rats and in vivo recovery was determined by retrodialysis. After oral and transdermal administration of aconitine, dialysate was sampled at 20 min intervals until the end of the experiment for the determination of concentration of aconitine in skin.

Methods: Flow cytometry was used to examine apoptosis and a PCR ELISA kit was used to detect telomerase activity.

A reversed-phase HPLC method is described for determination of 5-fluorouracil in human serum. After being extracted with ethyl acetate 5-fluorouracil was analyzed by reversed-phase HPLC column (Zorbax ODS) and UV-detector (273nm) with tegaful as internal standard. The mobile phase was a mixture of methyl alcohol and water (20:80, V:V) at a flow rate of 1mL/min.

Its primary effect is the antitumor activity on transplanted mouse tumors and cultured tumor cells. 4-[4''-(2'',2'',6'',6''-Tetramethyl-1''-piperidinyloxy)amino]-4'- demethylepipodophyllotoxin (GP-7) 10-40 mg.kg-1 ip daily for 7 d reduced the specific antibody formation of splenocytes, serum agglutinin titer, and hemolysin HC50 in mice immunized with SRBC. GP-7 inhibited the footpad delayed hypersensitivity reaction and decreased the weights of spleen and thymus, but did not affect the phagocytic function of the peritoneal macrophages.
