Inflammation, a central process in numerous diseases, plays a crucial role in hepatic disorders, arthritis, cardiac conditions, and neurodegenerative ailments. Given the lack of effective anti-inflammatory drugs, it is imperative to assess inflammation severity and explore novel therapeutics. Selenocysteine (Sec), a key mediator of selenium's biological function, is closely involved in anti-inflammatory responses.

Epigenetics in autosomal dominant polycystic kidney disease.

Biochim Biophys Acta Mol Basis Dis

January 2025

Department of Nephrology, Shanghai East Hospital, Tongji University School of Medicine, Shanghai, China; Department of Medicine, Rhode Island Hospital and Alpert Medical School, Brown University, Providence, RI, USA. Electronic address:

Autosomal dominant polycystic kidney disease (ADPKD) is the fourth leading cause of end-stage renal disease, contributing substantially to patient morbidity, mortality, and healthcare system strain. Emerging research highlights a pivotal role of epigenetics in ADPKD's pathophysiology, where mechanisms like DNA methylation, histone modifications, and non-coding RNA regulation significantly impact disease onset and progression. These epigenetic factors influence gene expression and regulate key processes involved in cyst formation and expansion, fibrosis, and inflammatory infiltration, thus accelerating ADPKD progression.

As combination therapy becomes more common in clinical applications, predicting adverse effects of combination medications is a challenging task. However, there are three limitations of the existing prediction models. First, they rely on a single view of the drug and cannot fully utilize multiview information, resulting in limited performance when capturing complex structures.

Emerging tick-borne orthonairovirus infections pose a growing global concern, with limited understanding of the viral ovarian tumor-like cysteine proteases (vOTUs) encoded by novel orthonairoviruses. These vOTUs, a group of deubiquinylases (DUBs), disrupt the innate immune response. Yezo virus (YEZV), a recently discovered pathogenic orthonairovirus, was first reported in Japan in 2021.

The Role of AIM2 in Cancer Development: Inflammasomes and Beyond.

J Cancer

January 2025

Center for Immunology, Key Laboratory of Prevention and Treatment of Cardiovascular and Cerebrovascular Diseases, Ministry of Education, Gannan Medical University, Ganzhou, Jiangxi 341000, China.

Absence in melanoma 2 (AIM2) protein functions as a double-stranded DNA sensor and is critical for host defense against intracellular bacterial and viral pathogens. Recent research has highlighted the significance of AIM2 in the pathogenesis of diverse malignancies. Through its recognition of foreign or intracellular dsDNA, AIM2 triggers inflammasome activation, resulting in the release of pro-inflammatory cytokines such as IL-1β, IL-18, and induction of pyroptosis.

Warfarin is the most widely used oral anticoagulant in clinical practice. The cytochrome P450 2C9 (CYP2C9), vitamin K epoxide reductase complex 1 (VKORC1), and cytochrome P450 4F2 (CYP4F2) genotypes are associated with warfarin dose requirements in China. Accurate genotyping is vital for obtaining reliable genotype-guided warfarin dosing information.

Haplotype-Resolved Genotyping and Association Analysis of 1,020 β-Thalassemia Patients by Targeted Long-Read Sequencing.

Adv Sci (Weinh)

December 2024

Innovation Center for Diagnostics and Treatment of Thalassemia, Nanfang Hospital, Southern Medical University, Guangzhou, Guangdong, 510515, China.

Despite the well-documented mutation spectra of β-thalassemia, the genetic variants and haplotypes of globin gene clusters modulating its clinical heterogeneity remain incompletely illustrated. Here, a targeted long-read sequencing (T-LRS) is demonstrated to capture 20 genes/loci in 1,020 β-thalassemia patients. This panel permits not only identification of thalassemia mutations at 100% of sensitivity and specificity, but also detection of rare structural variants (SVs) and single nucleotide variants (SNVs) in modifier genes/loci.

Targeted protein degradation (TPD) represents a promising therapeutic approach, encompassing several innovative strategies, including but not limited to proteolysis targeting chimeras (PROTACs), molecular glues, hydrophobic tag tethering degraders (HyTTD), and lysosome-targeted chimeras (LYTACs). Central to TPD are small molecule ligands, which play a critical role in mediating the degradation of target proteins. This review summarizes the current landscape of small molecule ligands for TPD molecules.

PeerJ

December 2024

Key Laboratory of Prevention and Treatment of Cardiovascular and Cerebrovascular Diseases of Ministry of Education, Gannan Medical University, Ganzhou, China.

Ischemic stroke (IS) remains a leading cause of disability and mortality worldwide, and inflammation and oxidative stress play significant roles in its pathogenesis. This study investigates the effects of dihydromyricetin (DHM) on IS using RT-qPCR and western blot with SH-SY5Y cells, focusing on its effects on the small nucleolar RNA host gene 10 (SNHG10)/microRNA (miR)-665/Ras association domain family member 5 (RASSF5) axis and nuclear factor-kappa B (NF-κB) signaling. In addition, the effects of the SNHG10/miR-665/RASSF5 axis on SH-SY5Y cell activity, apoptosis, oxidative stress, and inflammatory markers were analyzed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, flow cytometry, and enzyme-linked immunosorbent assays.

Photoinduced Concerted Fragmentation of Alkyl -Phthalimidoyl Oxalates: Mechanisms and Applications.

Org Lett

December 2024

School of Pharmaceutical and Chemical Engineering and Institute for Advanced Studies, Taizhou University, Taizhou 318000, China.

In this work, we used experiments and density functional theory calculations to investigate the mechanism and driving forces of the reductive fragmentation of NHPI esters. Mechanistic studies suggest that the fragmentation behavior of the NHPI ester is influenced not only by the electronic nature of the substituent group but also by the stability of the radical intermediate. To further investigate this transformation, we next examined the aminoalkoxycarbonylation of alkenes using alkyl -phthalimidoyl oxalates.

Benzothiadiazole (BT) has shown promising applications in fullerene solar cells. However, few BT-based polymer donors exhibited a noticeable power conversion efficiency (PCE) for the fused-ring small molecular acceptor-based polymer solar cells (PSCs). Herein, we developed a D-A (D: donor, A: acceptor) polymer donor F-1 based on fluorinated BT (ffBT) as A unit and chlorinated benzo [1,2-b:4,5-b'] dithiophene (BDT-2Cl) as D unit.
