Inherited genetics represents an important contributor to risk of esophageal adenocarcinoma (EAC), and its precursor Barrett's esophagus (BE). Genome-wide association studies have identified ∼30 susceptibility variants for BE/EAC, yet genetic interactions remain unexamined. To address challenges in large-scale G×G scans, we combined knowledge-guided filtering and machine learning approaches, focusing on genes with (A) known/plausible links to BE/EAC pathogenesis (n=493) or (B) prior evidence of biological interactions (n=4,196).

Purpose: To evaluate the efficacy of 25 Gy in 5 fractions (25 Gy/5#) prophylactic pelvic nodal irradiation for regional control during stereotactic radiation therapy (SBRT) for high-risk prostate cancer.
Methods And Materials: The multinational SHARP consortium database of patients treated with curative-intent prostate SBRT for high-risk prostate cancer was queried for prophylactic radiation therapy 25 Gy/5# to the pelvic lymph nodes. Details of Phoenix-defined biochemical failure and location of recurrence (local, regional, or distant) were extracted.

The fabrication of objects with complex shape and geometry has been greatly facilitated with the advancements in additive manufacturing. While synthetic polymers like ABS and PLA have found widespread use in extrusion 3D printing, other biobased thermoplastics that are both biodegradable and biocompatible could offer strategic advantages over traditional synthetic materials. In this work dextran of low (20 kDa) and medium (40 kDa) molecular weight (MW) was modified with palmitic acid to obtain meltable polymers for extrusion 3D printing/fused deposition modeling additive manufacturing.

Molecular subtypes, such as defined by The Cancer Genome Atlas (TCGA), delineate a cancer's underlying biology, bringing hope to inform a patient's prognosis and treatment plan. However, most approaches used in the discovery of subtypes are not suitable for assigning subtype labels to new cancer specimens from other studies or clinical trials. Here, we address this barrier by applying five different machine learning approaches to multi-omic data from 8,791 TCGA tumor samples comprising 106 subtypes from 26 different cancer cohorts to build models based upon small numbers of features that can classify new samples into previously defined TCGA molecular subtypes-a step toward molecular subtype application in the clinic.

The metabolites gluconic acid, 5-ketogluconic acid, proline, and glutamic acid, produced by Pseudomonas reptilivora B-6bs, are industrially important, particularly in food and pharmaceutical sectors. However, producing these metabolites involves biotin supplementation to enhance yields, which is an expensive additive, and reducing its use can significantly lower production costs. Thus, This study aimed to enhance the production of gluconic acid, 5-ketogluconic acid, proline, and glutamic acid without biotin supplementation.

Background: Psychiatric disease burden in patients with Inflammatory bowel disease (IBD) has risen substantially over the past few decades. However, there is limited data on the relationship between IBD disease activity and the incidence of psychiatric comorbidities. We sought to conduct a population-based study to investigate the impact of early onset disease activity in newly diagnosed IBD patients on psychiatric disease diagnoses and medication usage.

Background And Objectives: Perhaps stemming from the central role of detailed examinations and a focus on the subjective sphere that grounds their clinical practice, neurologists have frequently opined on experiences traditionally a province of humanities. The increasingly technological focus on medical education and care can be seen to devalue the subjective aspects of medicine. As a counter to this, we report on the existence of neurohumanities curricula within neurology residency training.

Background: The complex relationship among sleep, exercise, and the gut microbiome presents a unique opportunity to improve health and wellness. Here, we conducted the first large-scale investigation into the influence of a novel elite athlete-derived probiotic, consisting of a multi-strain Lactobacillus consortium, on sleep quality, exercise recovery, and gut microbiome composition in both elite athletes (n = 11) and the general population (n = 257).
Results: Our two-phase study design, which included an open-label study followed by a controlled longitudinal study in a professional soccer team, allowed us to identify key interactions between probiotics, the gut microbiome, and the host.

Dysregulation of integral membrane proteins (IMPs) has been linked to a myriad of diseases, making these proteins an attractive target in drug research. Whilst PROTAC technology has had a significant impact in scientific research, its application to IMPs is still limited. Limitations of the traditional approach of immunoblotting in PROTAC research include the low throughput compared to other methods, as well as a lack of spatial information for the target.

Hydrogel-based soft machines are promising in diverse applications, such as biomedical electronics and soft robotics. However, current fabrication techniques generally struggle to construct multimaterial three-dimensional hydrogel architectures for soft machines and robots, owing to the inherent hydrogel softness from the low-density polymer network nature. Herein, we present a multimaterial cryogenic printing (MCP) technique that can fabricate sophisticated soft hydrogel machines with accurate yet complex architectures and robust multimaterial interfaces.

This study aims to improve our understanding of acute ischemic stroke clot imaging by integrating CT attenuation information with MRI susceptibility signal of thrombi. For this proof-of-principle experimental study, fifty-seven clot analogs were produced using ovine venous blood with a broad histological spectrum. Each clot analog was analyzed to determine its RBC content and chemical composition, including water, Fe III, sodium, pH, and pO2.

Brucellosis has therapeutic challenges due to 3%-15% relapses/therapeutic failures (R/TF) after antibiotic treatment. Therefore, determining the antibiotic concentration in tissues, the physiopathological parameters, and the R/TF after treatment is relevant. After exploring different antibiotic quantities, we found that a combined dose of 100 µg/g of doxycycline (for 45 days) and 7.

Neuroblastoma is the most prevalent extracranial solid tumor among children and exhibits remarkable heterogeneity. The methylation of cytosine to form 5-methylcytosine (m5C) is the primary type of modification found in DNA and RNA. The NOL1/NOP2/sun (NSUN) family, specifically NSUN1, is responsible for the methylation process and has been shown to play a key role in cell differentiation and cancer development.

The microencapsulation of Lactocaseibacillus rhamnosus GG in a matrix of sodium alginate, xanthan gum, gum arabic and chitosan hydrochloride is a promising strategy for protecting this probiotic during passage through the gastrointestinal tract. This study evaluated the influence on the viability of Lactocaseibacillus rhamnosus GG encapsulated with these polymers by external ionic gelation with vibratory extrusion and the microcapsules that showed the best results of capsulation efficiency, viability, size and morphology were analyzed by Fourier transform infrared spectroscopy (FTIR), thermal analysis (TGA) and exposure to environmental stress conditions and gastrointestinal simulation. The result revealed encapsulation efficiency values above 95 % for all formulations and survival rate higher than 6 log CFU/mL for most analyzed groups.

RNA exhibits remarkable capacity as a functional polymer, with broader catalytic and ligand-binding capability than previously thought. Despite this, the low side chain diversity present in nucleic acids (two purines and two pyrimidines) relative to proteins (20+ side chains of varied charge, polarity, and chemical functionality) limits the capacity of functional RNAs to act as environmentally responsive polymers, as is possible for peptide-based receptors and catalysts. Here we show that incorporation of the modified nucleobase 2-thiouridine (2sU) into functional (aptamer and ribozyme) RNAs produces functionally inactivated polymers that can be activated by oxidative treatment.

Background: Polymorphisms of the folate-associated one-carbon metabolism (OCM) pathway genes may regulate certain susceptibilities to cancer. G80A, a polymorphism in the reduced folate carrier (RFC) gene, may be associated with cancer risk, although the results obtained from previous studies have been inconsistent.
Objectives: This study aimed to evaluate the association of G80A with lung cancer among a Chinese population and to examine the potential effect modifiers.
