Objectives: Taxane (paclitaxel or docetaxel) and platinum (cisplatin or carboplatin) chemotherapy is commonly used in the treatment of ovarian cancer. Despite an initial high response to therapy, the 5-year survival rate remains low. The identification of pharmacogenomic markers to identify patients unlikely to respond or at risk for severe toxicity will assist in the goal of individualizing ovarian cancer treatment.

Thiopurine methyltransferase (TPMT) activity shows significant interindividual variation, with approximately 90% of individuals having high (wild-type) activity, 10% with intermediate activity, and 0.3% with low activity. Low and intermediate TPMT activity leads to toxicity from mercaptopurine and the need for dose reduction.

Multiple combinations of chemotherapy regimens are available, including anthracyclines, taxanes, antimetabolites, alkylating agents, platinum drugs and vinca alkaloids. This review discusses the pharmacokinetic and pharmacogenomic information available for commonly used breast cancer chemotherapy drugs.
