Organ-on-a-chip (OOC, organ chip) technology can closely simulate the human microenvironment, synthesize organ-like functional units on a fluidic chip substrate, and simulate the physiology of tissues and organs. It will become an increasingly important platform for in vitro drug development and screening. Most importantly, organ-on-a-chip technology, incorporating 3D cell cultures, overcomes the traditional drawbacks of 2D (flat) cell-culture technology in vitro and in vivo animal trials, neither of which generate completely reliable results when it comes to the actual human subject.

Zoledronic acid (ZA) is one of the most important and effective class of anti-resorptive drug available among bisphosphonate (BP), which could effectively reduce the risk of skeletal-related events, and lead to a treatment paradigm for patients with skeletal involvement from advanced cancers. However, the exact molecular mechanisms of its anticancer effects have only recently been identified. In this review, we elaborate the detail mechanisms of ZA through inhibiting osteoclasts and cancer cells, which include the inhibition of differentiation of osteoclasts via suppressing receptor activator of nuclear factor κB ligand (RANKL)/receptor activator of nuclear factor κB (RANK) pathway, non-canonical Wnt/Ca2+/calmodulin dependent protein kinase II (CaMKII) pathway, and preventing of macrophage differentiation into osteoclasts, in addition, induction of apoptosis of osteoclasts through inhibiting farnesyl pyrophosphate synthase (FPPS)-mediated mevalonate pathway, and activation of reactive oxygen species (ROS)-induced pathway.

Backgrounds The aim of this study was to evaluate the utility of unilateral single injection thoracic paravertebral block (TPVB) with and without the addition of betamethasone for the acute pain management of patient's undergoing laparoscopic cholecystectomy (LC). Methods Eligible patients were allocated randomly to three groups: (A) general anesthesia followed by surgeon infiltration at port sites with ropivacaine (n = 48), (B) general anesthesia after single injection TPVB at right T7-8 level with ropivacaine only, Ropi_TPVB (n = 43), and (C) general anesthesia after single injection TPVB with ropivacaine plus betamethasone, Ropi_Betamet_TPVB (n = 45). Primary outcome was TPVB duration assessed by the number of dermatomes at regular intervals up to 72 hours (h).
