Dysregulation of long non-coding RNAs (lncRNAs) is common in colorectal cancer liver metastasis (CRLM). Emerging evidence links lncRNAs to multiple stages of metastasis from initial migration to colonization of distant organs. In this study we investigated the role of lncRNAs in metabolic reprogramming during CRLM using patient-derived organoid (PDO) models.

Inspired by biological processes, feature learning techniques, such as deep learning, have achieved great success in various fields. However, since biological organs may operate differently from semiconductor devices, deep models usually require dedicated hardware and are computation-complex. High energy consumption has made deep model growth unsustainable.

Serine arginine-rich splicing factor 1 (SRSF1) is a key oncogenic splicing factor in various cancers, promoting abnormal gene expression through post-translational regulation. Although the protumoral function of SRSF1 is well-established, the effects of inhibiting tumor-intrinsic SRSF1 on the tumor microenvironment and its impact on CD8 T cell-mediated antitumor immunity remain unclear. Our findings indicate that depleting SRSF1 in CD8 T cells improve antitumor immune function, glycolytic metabolism, and the efficacy of adoptive T cell therapy.

Adoptive transfer of genetically or nanoparticle-engineered macrophages represents a promising cell therapy modality for treatment of solid tumor. However, the therapeutic efficacy is suboptimal without achieving a complete tumor regression, and the underlying mechanism remains elusive. Here, we discover a subpopulation of cancer cells with upregulated CD133 and programmed death-ligand 1 in mouse melanoma, resistant to the phagocytosis by the transferred macrophages.

Objectives: Fasting plasma glucose (FPG), glycated hemoglobin A (HbA), and 2-hour post-load plasma glucose (2h PG) are all currently used to define prediabetes. We aimed to determine whether a higher number of prediabetes defects corresponds to an increased all-cause and cardiovascular disease (CVD) mortality.
Methods: Individuals with prediabetes and available information on FPG, HbA, 2h PG, and mortality data were derived from the 2005 - 2016 National Health and Nutrition Examination Survey.

Gene editing technologies, particularly clustered regularly interspersed short palindromic repeats (CRISPR) and CRISPR-associated (Cas) proteins, have revolutionized the ability to modify gene sequences in living cells for therapeutic purposes. Delivery of CRISPR/Cas ribonucleoprotein (RNP) is preferred over its DNA and RNA formats in terms of gene editing effectiveness and low risk of off-target events. However, the intracellular delivery of RNP poses significant challenges and necessitates the development of non-viral vectors.

Two series of polycyclic aromatic hydrocarbon isomers ( and , and and ) were designed and synthesized by isomerically fusing phenanthrene with thiophene and thieno[3,2-]thiophene, respectively. All of the new target molecules were confirmed by single-crystal X-ray analysis, and it was found that the solid-state packing can be effectively modulated through a combination of π-extended and isomeric fused strategies. Meanwhile, compared with thiophene ring-terminated isomers and , both having a V-shaped geometry and showing no obvious self-assembly behavior, π-extended unit thieno[3,2-]thiophene-terminated isomer displays a V-shaped structure with moderate self-assembly properties and isomer exhibits a C-shaped configuration with further enhanced self-assembly properties.

The design and development of advanced surgical sutures with appropriate structure and abundant bio-functions are urgently required for the chronic wound closure and treatment. In this study, an integrated technique routine combining modified electrospinning with hot stretching process was proposed and implemented to fabricate poly(L-lactic acid) (PLLA) nanofiber sutures, and the Salvia miltiorrhiza Bunge-Radix Puerariae herbal compound (SRHC) was encapsulated into PLLA nanofibers during the electrospinning process to enrich the biofunction of as-generated sutures. All the PLLA sutures loading without or with SRHC were found to exhibit bead-free and highly-aligned nanofiber structure.

Neurodegenerative diseases are a group of disorders characterized by progressive degeneration or death of neurons. The complexity of clinical symptoms and irreversibility of disease progression significantly affects individual lives, leading to premature mortality. The prevalence of neurodegenerative diseases keeps increasing, yet the specific pathogenic mechanisms remain incompletely understood and effective treatment strategies are lacking.

Triple-negative breast cancer (TNBC) is a difficulty and bottleneck in the clinical treatment of breast cancer due to a lack of effective therapeutic targets. Herein, we first report that secernin 2 (SCRN2), an uncharacterized gene in human cancer, acts as a novel tumor suppressor in TNBC to inhibit cancer progression and enhance therapeutic sensitivity to poly(ADP-ribose) polymerase (PARP) inhibition both in vitro and in vivo. SCRN2 is downregulated in TNBC through chaperone-mediated autophagic degradation, and its downregulation is associated with poor patient prognosis.

The cultivation and differentiation of human embryonic stem cells (hESCs) into organoids are crucial for advancing of new drug development and personalized cell therapies. Despite establishing of chemically defined hESC culture media over the past decade, these media's reliance on growth factors, which are costly and prone to degradation, poses a challenge for sustained and stable cell culture. Here, we introduce an hESC culture system(E6Bs) that facilitates the long-term, genetically stable expansion of hESCs, enabling cells to consistently sustain high levels of pluripotency markers, including NANOG, SOX2, TRA-1-60, and SSEA4, across extended periods.

Extracellular vesicles (EVs) have been demonstrated to own the advantages in evading phagocytosis, crossing biological barriers, and possessing excellent biocompatibility and intrinsic stability. Based on these characteristics, EVs have been used as effective therapeutic carriers for drug delivery, but the low drug loading capacity greatly limits further applications. Herein, we developed a drug loading method based on cell-penetrating peptide (CPP) to enhance the encapsulation of therapeutic reagents in EVs, and EVs-based drug delivery system achieved higher killing efficacy to tumor cells.

Purpose: This study evaluated the differences in amyloid-β (Aβ), tau deposition, and longitudinal tau deposition between subjective cognitive decline (SCD) and objective subtle cognitive difficulties (Obj-SCD).
Methods: Participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI) cohort (n = 234) and the Huashan cohort (n = 267) included individuals with Obj-SCD, SCD, subjective memory concern (SMC), and healthy controls (HC). General linear models (GLM) were used to compare baseline and longitudinal differences in Aβ and tau among the groups, and to examine the associations between these biomarkers.

In plants, sperm cell formation involves two rounds of pollen mitoses, in which the microspore initiates the first pollen mitosis (PMI) to produce a vegetative cell and a generative cell, then the generative cell continues the second mitosis (PMII) to produce two sperm cells. DUO1, a R2R3 Myb transcription factor, is activated in the generative cell to promote S-G2/M transition during PMII. Loss-of-function of DUO1 caused a complete arrest of PMII.

Background: Despite the importance of lower limb sensation in walking highlighted in systematic reviews, there is limited research investigating the effect of proprioceptive deficits after stroke and any relationship with walking ability.
Objectives: With stroke survivors of different walking ability, this study aimed to (1) explore side (affected/unaffected) and movement direction (inversion/plantar flexion) effects in ankle joint position sense (JPS) acuity, and (2) compare ankle JPS acuity between groups of stroke survivors with different walking ability.
Methods: Seventy subacute stroke survivors were recruited and divided into three groups based on walking ability, as determined by their gait speed on the 10-Meter Walking Test: household (<0.
