The expression dysregulation of microRNAs (miRNA) has been widely reported during cancer development, however, the underling mechanism remains largely unanswered. In the present work, we performed a systematic integrative study for genome-wide DNA methylation, copy number variation and miRNA expression data to identify mechanisms underlying miRNA dysregulation in lower grade glioma. We identify 719 miRNAs whose expression was associated with alterations of copy number variation or promoter methylation.

Kinesin motor KIF16A regulates microtubule stability and actin-dependent spindle migration in mouse oocyte meiosis.

FASEB J

July 2024

College of Animal Science and Technology, Anhui Agricultural University, Key Laboratory of Local Livestock and Poultry Genetical Resource Conservation and Breeding of Anhui Province, Hefei, China.

Article Synopsis
  • Kif16A is a kinesin-3 motor protein that is crucial for mouse oocyte meiotic maturation, affecting key processes like spindle assembly, chromosome alignment, and actin filament distribution.
  • Targeted depletion of Kif16A disrupts the meiotic cell cycle, causing spindle assembly errors, misaligned chromosomes, and reduced microtubule stability.
  • The study also highlights that Kif16A is essential for proper spindle assembly checkpoint activity, as its absence increases defective kinetochore-microtubule attachments and inhibits vital protein localization.

Background: Proline dehydrogenase () encodes a mitochondrial protein that catalyzes the first step of proline degradation and is related to angiogenesis. Angiogenesis is a critical process in the development and progression of tumors, including lung adenocarcinoma (LUAD), as tumor growth and metastasis are dependent on angiogenesis. The mitochondria and their associated genes thus play a vital role in tumor therapy.

Base-Mediated Cascade Lactonization/1,3-Dipolar Cycloaddition Pathway for the One-Pot Assembly of Coumarin-Functionalized Pyrrolo[2,1-]isoquinolines.

J Org Chem

June 2024

Key Laboratory of Functional Molecular Solids, Ministry of Education, Anhui Laboratory of Molecule-Based Materials (State Key Laboratory Cultivation Base), College of Chemistry and Materials Science, Anhui Normal University, Wuhu 241002, P. R. China.

An elegant and highly concise strategy for the construction of coumarin-functionalized pyrrolo[2,1-]isoquinolines from available propargylamines and isoquinolinium -ylides has been disclosed. In this reaction, isoquinolinium -ylides acted as a C2 synthon to form a coumarin ring as well as a 1,3-dipole to construct a pyrrole ring in a single pot. This cascade process involves 1,4-conjugate addition/lactonization/1,3-dipolar cycloaddition to construct four chemical bonds (one C-O bond and three C-C bonds) and two new heterocyclic skeletons.

Article Synopsis
  • This study investigates the role of white adipose tissue (WAT) in rheumatoid arthritis (RA), suggesting it could be a potential target for treatment.
  • Researchers compared the WAT of healthy rats to those with adjuvant-induced arthritis (AIA) and found that AIA rats had smaller WAT but higher secretion levels of inflammatory proteins.
  • The study concludes that specific hepatokines enhance inflammatory responses in pre-adipocytes, which may worsen RA, indicating that targeting this dysfunction could help reduce the severity of the disease.

Ferroptosis and ferritinophagy play critical roles in various disease contexts. Herein, we observed that ferroptosis and ferritinophagy were induced both in the brains of mice with diabetes mellitus (DM) and neuronal cells after high glucose (HG) treatment, as evidenced by decreases in GPX4, SLC7A11, and ferritin levels, but increases in NCOA4 levels. Interestingly, melatonin administration ameliorated neuronal damage by inhibiting ferroptosis and ferritinophagy both and .
