Evidence of a complex formation is a crucial step in the structural studies of ligand-receptor interactions. Here we presented a simple and fast approach for qualitative screening of the complex formation between the chimeric extracellular domain of the nicotinic acetylcholine receptor (α7-ECD) and three-finger proteins. Complex formation of snake toxins α-Bgtx and WTX, as well as of recombinant analogs of human proteins Lynx1 and SLURP-1, with α7-ECD was confirmed using fluorescently labeled ligands and size-exclusion chromatography with simultaneous absorbance and fluorescence detection.

Mutations in the glucocerebrosidase gene (GBA) cause Gaucher disease (GD), the lysosomal storage disorder (LSD), and are the most common genetic risk factor of Parkinson's disease (PD). Lysosome functionality plays a critical role for secretion of extracellular vesicles (EVs) and their content. Here we compared EVs from the blood plasma of 8 GD patients and 8 controls in terms of amounts, size distribution, and composition of their protein cargo.

Instrumented indentation has become an indispensable tool for quantitative analysis of the mechanical properties of soft polymers and biological samples at different length scales. These types of samples are known for their prominent viscoelastic behavior, and attempts to calculate such properties from the indentation data are constantly made. The simplest indentation experiment presents a cycle of approach (deepening into the sample) and retraction of the indenter, with the output of the force and indentation depth as functions of time and a force versus indentation dependency (force curve).

To study the structure and function of the pH-regulated receptor tyrosine kinase insulin receptor-related receptor (IRR), а member of the insulin receptor family, we obtained six mouse monoclonal antibodies against the recombinant IRR ectodomain. These antibodies were characterized in experiments with exogenously expressed full-length IRR by Western blotting, immunoprecipitation, and immunocytochemistry analyses. Utilizing a previously obtained set of IRR/IR chimeras with swapped small structural domains and point amino acid substitutions, we mapped the binding sites of the obtained antibodies in IRR.

Extracellular vesicles (EV) are involved in important processes of glioblastoma multiforme (GBM), including malignancy and invasion. EV secreted by glioblastoma cells may cross the hematoencephalic barrier and carry molecular cargo derived from the tumor into the peripheral circulation. Therefore, the determination of the molecular composition of exosomes released by glioblastoma cells seems to be a promising approach for the development of non-invasive methods of the detection of the specific exosomal protein markers in the peripheral blood.

The integration of copper nanoparticles as antifungal agents in polymeric matrices to produce copper polymer nanocomposites has shown excellent results in preventing the growth of a wide variety of toxigenic fungi. Copper-chitosan nanocomposite-based chitosan hydrogels (Cu-Chit/NCs hydrogel) were prepared using a metal vapor synthesis (MVS) and the resulting samples were described by transmission electron microscopy (TEM), X-ray fluorescence analysis (XRF), and small-angle X-ray scattering (SAXS). Aflatoxin-producing medium and VICAM aflatoxins tests were applied to evaluate their ability to produce aflatoxins through various strains of associated with peanut meal and cotton seeds.

In this study, a configuration of a compact imaging objective based on a reflecting annular harmonic lens was proposed. Light propagation through the proposed optical system was comprehensively modeled using a dedicated special program and the ZEMAX software, with the latter used to derive the point spread function (PSF). Several relationships were used to describe the connection between key parameters of the objective, including its focal length, field of view, and thickness.

We propose a new layout for the Offner imaging hyperspectrometer that is utilized onboard small space vehicles. The layout is based on a method of adjusting the adaptive temperature-dependent optical scheme by moving just two coaxial optical components located inside the hyperspectrometer. We present the results of modeling for a temperature range of -40 to +45 °C and an optical experiment using a heat and cold chamber for the range of 12 to 40 °C, proving the basic functionality of the proposed layout.

Electrospinning of polyacrylonitrile/DMF dopes containing salts of nickel, cobalt, zirconium, cerium, gadolinium, and samarium, makes it possible to obtain precursor nanofiber mats which can be subsequently converted into carbon nanofiber (CNF) composites by pyrolysis at 1000-1200 °C. Biodegradable poly(l-lactide)/calcium phosphate composites are promising materials for fabrication of bone fixation implants with improved properties. Multistage compounding was proposed as an efficient method for the preparation of biodegradable poly(l-lactide)/calcium phosphate composites with submicron filler dispersion and mechanical characteristics similar to native bone. The improvement of the characteristics is caused both by the filler itself and by the increase of polymer crystallinity due to the nucleation effect.

Interaction between proteins and synthetic polymers that represent a perspective potential in drug delivery or/and already used in medicine plays a key role in biological functioning of both molecules along with a system as a whole. In present study association between hen egg white lysozyme and Pluronic triblock-copolymers (L121, P123 and F127) in the bulk of the solution as well as at the aqueous-air and liquid-liquid interfaces was analyzed by means of spectroscopic and radiochemical assay. In protein-Pluronic complexes lysozyme keeps the secondary structure (CD and SAXS data results), while fluorescence and UV-analysis indicates changes in the local surrounding of fluorophoric amino acid residues.

The experimental evidence on depressurization foaming of the amorphous D,L-polylactide, which is plasticized by subcritical (initial pressures below the critical value) or supercritical (initial pressures above the critical value) carbon dioxide at a temperature above the critical value, relates to two extreme cases: a slow quasi-isothermal foam expansion, and a rapid quasi-adiabatic expansion. Under certain conditions, the quasi-isothermal mode is characterized by the non-monotonic dependencies of the foam volume on the external pressure that are associated with the expansion-to-shrinkage transition. The quasi-adiabatic and quasi-isothermal expansions are characterized by a significant increase in the degree of foam expansion under conditions where the CO initial pressure approaches the critical value.

TRAIL is considered a promising antitumor agent because it causes apoptosis of transformed cells without affecting normal cells. However, many types of tumors are cytokine resistant, and combination therapy with various chemotherapeutic drugs is being developed to overcome the resistance. We have demonstrated that the combination of TRAIL with doxorubicin, bortezomib, and panobinostat dramatically reduced the viability of TRAIL-resistant A549 and HT-29 cells.

We demonstrated and investigated, both theoretically and experimentally, the transformation of cylindrical vector beams with an embedded phase singularity under the condition of focusing perpendicularly to the axis of the anisotropic calcite crystal. Theoretical and numerical analysis, performed on the basis of decomposing the light field into a set of plane waves for an anisotropic medium, allowed us to study the dependence of the structural transformation of the initial laser beam on the polarisation and phase state in detail. The proposed approach allows one to perform the visual recognition of cylindrically-polarised vector beams of various orders and can be used for the demultiplexing of information channels in the case of polarisation-division multiplexing.

The lens capsule, a thin specialized basement membrane that encloses the crystalline lens, is essential for both the structural and biomechanical integrity of the lens. In this review, we have described the techniques used for the lens capsule biomechanical testing on the macro- and microscale and summarized the current knowledge about its mechanical properties.

Formulated forms of cancer therapeutics enhance the efficacy of treatment by more precise targeting, increased bioavailability of drugs, and an aptitude of some delivery systems to overcome multiple drug resistance of tumors. Drug carriers acquire importance for anti-cancer interventions via targeting tumor-associated macrophages with active molecules capable to either eliminate them or change their polarity. Although several packaged drug forms have reached the market, there is still a high demand for novel carrier systems to hurdle limitations of existing drugs on active molecules, toxicity, bioeffect, and stability.
