Highly selective inhibitors of cyclin-dependent kinase 4 and 6 (CDK4/6is) have emerged as a standart of care for first- and second-line therapies in combination with endocrine therapy (ET) for HR+/HER2- metastatic breast cancer (MBC) patients. It has been reported that combination therapy is more effective than ET alone and is safe in elderly patients as well as young patients. Nevertheless, elderly and very old patients with HR+/HER2-MBC treated with CDK4/6 inhibitor (CDK4/6i) combinations are relatively underrepresented in randomized controlled trials.

GENCODE 2025: reference gene annotation for human and mouse.

Nucleic Acids Res

January 2025

European Molecular Biology Laboratory, European Bioinformatics Institute, Wellcome Genome Campus, Hinxton, Cambridge CB10 1SD, UK.

GENCODE produces comprehensive reference gene annotation for human and mouse. Entering its twentieth year, the project remains highly active as new technologies and methodologies allow us to catalog the genome at ever-increasing granularity. In particular, long-read transcriptome sequencing enables us to identify large numbers of missing transcripts and to substantially improve existing models, and our long non-coding RNA catalogs have undergone a dramatic expansion and reconfiguration as a result.

Article Synopsis
  • - Accurate gene annotations are essential for interpreting how genomes function, and the GENCODE consortium has spent twenty years creating reference annotations for human and mouse genomes, serving as a vital resource for researchers globally.
  • - Previous annotations of long non-coding RNAs (lncRNAs) were incomplete and poorly organized, hindering research, prompting GENCODE to launch a comprehensive effort that resulted in adding nearly 18,000 novel human genes and over 22,000 novel mouse genes, significantly increasing the catalog of transcripts.
  • - The new annotations not only show evolutionary patterns and link to genetic variants associated with traits but also improve understanding of previously unclear genomic functions, greatly advancing research into both human and mouse genetic diseases.

Article Synopsis
  • The study investigated the significance of metabolic tumor volume (tMTV) in patients with advanced non-small cell lung cancer (NSCLC) undergoing immune checkpoint blockade (ICB) therapy, using 18F-FDG-PET/CT scans.
  • It involved 518 patients across multiple institutions and found that those with high tMTV had poorer overall survival when treated with ICBs alone compared to those with low tMTV.
  • The research suggests that high tMTV is associated with increased systemic inflammation and genomic instability, making it a potential biomarker for determining treatment strategies in NSCLC patients with positive PD-L1 expression.

As screening strategies employ better biomarkers and genetics to identify individuals at an increased risk of prostate cancer, there are currently no chemotherapeutic prevention strategies. With any chemoprevention strategy, the population will be younger and healthier; therefore, they will be less tolerant of side effects. This study translated findings from screening a natural product library and pre-clinical evaluation of curcumin (CURC) in combination with ursolic acid (UA) in prostate cancer models.

Article Synopsis
  • Schizophrenia's complexity has made it difficult to understand its mechanisms and develop effective treatments; a new study addresses this by examining transcriptomic changes at the single-cell level in the human prefrontal cortex of 140 individuals.
  • The researchers found that excitatory neurons were the most affected, with changes related to neurodevelopment and synapse function, and identified both common and rare genetic risk factors influencing these neuronal alterations.
  • Their findings reveal two distinct groups of individuals with schizophrenia based on specific excitatory and inhibitory neuron states, linking genetic risk to cellular changes and enhancing our understanding of schizophrenia's underlying biology.
