Background The currently used atherosclerotic cardiovascular disease risk calculator relies on several measured variables and does not incorporate some well-established risk factors such as family history of premature myocardial infarction and other nontraditional risk factors. Our study aimed to develop and validate a simple risk score to predict 10-year risk of incident cardiovascular events using patient-reported information. Methods and Results Using data from the Atherosclerosis Risk in Communities cohort, we identified adults with no previous history of cardiovascular disease and randomly divided the cohort into "development" (70%) and "validation" (30%) subgroups.

To identify the genetic determinants of this variability, we performed a pharmacogenomic genome-wide meta-analysis of genetic variants influencing β-blocker BP response.Methods and ResultsGenome-wide association analysis for systolic BP and diastolic BP response to β-blockers from 5 randomized clinical trials consisting of 1254 patients with hypertension of European ancestry were combined in meta-analysis and single nucleotide polymorphisms (SNPs) with P<10 were tested for replication in 2 independent randomized clinical trials of β-blocker-treated patients of European ancestry (n=1552).
