Methods: Patients who completed any of three parent studies (NCT02889796: inadequate response [IR] to methotrexate [MTX]; NCT02873936: IR to biologic disease-modifying antirheumatic drugs; NCT02886728: MTX-naïve) without rescue therapy could enter the LTE; patients taking FIL continued their dosage, and those who received comparators were rerandomised to FIL200 or FIL100. This analysis includes week 156 interim results.

Article Synopsis
  • This study explored the adverse events (AEs) related to COVID-19 vaccination in patients with rheumatoid arthritis (RA) as part of the COVAD-2 study, which surveyed participants from February to June 2022.
  • It included 7203 participants, including 1423 RA patients, and found that RA patients reported higher rates of both major and minor AEs compared to healthy controls, and even more when compared to those with other autoimmune diseases.
  • Active RA significantly increased the risk of major AEs and hospitalization, while patients on certain medications like methotrexate or TNF inhibitors experienced fewer AEs than those not on these treatments.

Objectives: This study aimed to elucidate which bio-naïve patients with rheumatoid arthritis (RA) are suitable for treatment with CTLA4-Ig.

Methods: This study enrolled 953 patients with RA who were administered their first biological disease-modifying antirheumatic drug (CTLA4-Ig, n = 328; tumour necrosis factor inhibitor [TNFi], n = 625) from July 2013 to August 2022. The primary outcome was the Clinical Disease Activity Index (CDAI) remission rate at week 24 in each group, adjusted using Propensity Score-based Inverse Probability of Treatment Weighting (PS-IPTW).

Symmetric inflammatory polyarthritis is the most prominent manifestation of rheumatoid arthritis (RA). However, RA can practically affect any organ system, whether hematologic, neurological, cardiac, lung, skin, eyes, or kidneys. Systemic involvement in RA can be severe when there is interstitial lung disease, scleritis, amyloidosis, pure red cell aplasia, or myelodysplasia.

Purpose Of Review: Common variable immunodeficiency enteropathy (CVID-E) is a noninfectious complication of CVID caused by chronic inflammation of the gastrointestinal (GI) tract. Based on literature, a paucity or lack of plasma cells, although not obligatory for diagnosis, is a pathognomonic feature of CVID and more frequent in CVID-E. However, there is no consensus on standardized histopathological analysis of this feature in biopsies.

Department of Pharmacy Practice, JSS College of Pharmacy, JSS Academy of Higher Education and Research, Sri Shivarathreeshwara Nagar, Mysuru, Karnataka, India.

Article Synopsis
  • * Rarely, SSZ can cause agranulocytosis, a serious condition where white blood cell counts drop dangerously low, as seen in two reported cases after six weeks of SSZ treatment.
  • * Monitoring blood counts regularly after starting SSZ is crucial, as patients showed recovery after stopping the drug and receiving additional treatments like colony-stimulating factors and antibiotics.

  • - Juvenile idiopathic arthritis (JIA) encompasses various types of arthritis in children under 16, with treatments aiming to manage the disease, support growth, and improve quality of life using medications like NSAIDs and DMARDs.
  • - A study compared intraarticular corticosteroid injection (IACI) practices among pediatric rheumatologists in Turkey and India, gathering data through a questionnaire, which revealed that all clinicians used IACIs, mainly for oligoarticular JIA, with varying preferences in agents, techniques, and follow-up care.
  • - Despite the widespread use of IACIs, there are notable differences in practices and outcomes; thus, the authors suggest the need for standardized protocols through randomized studies to improve effectiveness and patient

Article Synopsis
  • The study aimed to determine how methotrexate-polyglutamates (MTX-PGs) concentrations relate to treatment effectiveness and safety in patients new to methotrexate for rheumatoid arthritis.
  • In a clinical trial involving 300 patients, researchers measured MTX-PGs levels as they adjusted methotrexate dosages over 24 weeks, finding that higher MTX-PGs were linked to lower disease activity but were also associated with increased risk of liver damage.
  • Results indicated that while higher MTX-PGs initially correlated with better treatment outcomes, this link diminished when patients also received another medication, adalimumab, highlighting the complex interplay between drug dosages and patient responses.
