Unraveling the hidden role of a uORF-encoded peptide as a kinase inhibitor of PKCs.

Proc Natl Acad Sci U S A

October 2021

The Shraga Segal Department of Microbiology, Immunology and Genetics, Faculty of Health Sciences, Ben-Gurion University of the Negev, Beer-Sheva 84105, Israel;

Approximately 40% of human messenger RNAs (mRNAs) contain upstream open reading frames (uORFs) in their 5' untranslated regions. Some of these uORF sequences, thought to attenuate scanning ribosomes or lead to mRNA degradation, were recently shown to be translated, although the function of the encoded peptides remains unknown. Here, we show a uORF-encoded peptide that exhibits kinase inhibitory functions.

In humans, exercise-induced thermogenesis is a markedly variable component of total energy expenditure, which had been acutely affected worldwide by COVID-19 pandemic-related lockdowns. We hypothesized that dietary macronutrient composition may affect metabolic adaptation/fuel selection in response to an acute decrease in voluntary activity. Using mice fed short-term high-fat diet (HFD) compared to low-fat diet (LFD)-fed mice, we evaluated whole-body fuel utilization by metabolic cages before and 3 days after omitting a voluntary running wheel in the cage.

Background: Basement membrane (BM) accumulation is a hallmark of micro-vessel disease in diabetes mellitus (DM). We previously reported marked upregulation of BM components in internal thoracic arteries (ITAs) from type 2 DM (T2DM) patients by mass spectrometry. Here, we first sought to determine if BM accumulation is a common feature of different arteries in T2DM, and second, to identify other effects of T2DM on the arterial proteome.

Human adipose tissue is a putative direct target of daytime orexin with favorable metabolic effects: A cross-sectional study.

Obesity (Silver Spring)

November 2021

Department of Clinical Biochemistry and Pharmacology and the National Institute of Biotechnology in the Negev, Ben-Gurion University, Beer-Sheva, Israel.

Objective: Orexin/hypocretin (Ox) and its receptors (OxR), a neuroendocrine system centrally regulating sleep/wakefulness, were implicated in the regulation of peripheral metabolism. It was hypothesized that human adipose tissue constitutes a direct target of the OxA/OxR system that associates with distinct metabolic profile(s).

Methods: Serum Ox levels and abdominal subcutaneous and visceral adipose tissue expression of Ox/HCRT, OxR1/HCRTR1, and OxR2/HCRTR2 were measured in n = 81 patients.

The Use of Tamoxifen as a Potential Treatment for Bipolar Disorder.

Psychiatry Clin Psychopharmacol

September 2021

Department of Clinical Biochemistry and Pharmacology, Ben-Gurion University of the Negev, Israel.

Bipolar disorder (BD) is a chronic mental disease that substantially affects the patient's quality of life and causes significant morbidity and mortality. Although there are a variety of available treatments, therapeutic intervention is not beneficial in many cases and recurrence rates remain high. Recent data suggested tamoxifen, a drug with a wide range of activities, as a potential treatment for reducing manic symptoms of BD.

Article Synopsis
  • The study investigates how frequencies of regulatory T cells (Tregs) and myeloid-derived suppressor cells (MDSCs) in peripheral blood can be potential biomarkers for certain diseases, focusing on the effects of cytomegalovirus (CMV) infection.
  • Findings show that middle-aged individuals with a CMV infection tend to have lower frequencies of certain Tregs and MDSCs, suggesting that chronic infections can influence immune cell populations.
  • The research indicates that historical immunological exposures, such as infections, are more influential in determining Treg and MDSC levels than genetic factors, as assessed through twin studies.

The gut metabolite composition determined by the microbiota has paramount impact on gastrointestinal physiology. However, the role that bacterial metabolites play in communicating with host cells during inflammatory diseases is poorly understood. Here, we aim to identify the microbiota-determined output of the pro-inflammatory metabolite, succinate, and to elucidate the pathways that control transepithelial succinate absorption and subsequent succinate delivery to macrophages.
