Background: Umbilical outpouchings (UOs) in pigs are a multifactorial disease and little is known about effective prevention strategies and risk factors for UO development. UOs are common in Danish pigs and legislation complicates and increases the cost of keeping and raising pigs with UO. Recommendations for preventive measures exist but the scientific evidence behind the recommendations is often lacking.

Automated stenosis estimation of coronary angiographies using end-to-end learning.

The initial evaluation of stenosis during coronary angiography is typically performed by visual assessment. Visual assessment has limited accuracy compared to fractional flow reserve and quantitative coronary angiography, which are more time-consuming and costly. Lessons learned from spatial transcriptomic analyses in clear-cell renal cell carcinoma.

Spatial transcriptomics has emerged as a powerful tool for discerning the heterogeneity of the tumour microenvironment across various cancers, including renal cell carcinoma (RCC). Spatial transcriptomics-based studies conducted in clear-cell RCC (the only RCC subtype studied using this technique to date) have given insights into spatial interactions within this disease. These insights include the role of epithelial-to-mesenchymal transitioning, revealing proximity-dependent interactions between tumour cells, fibroblasts, interleukin-2-expressing macrophages and hyalinized regions.

Prospective validation of ORACLE, a clonal expression biomarker associated with survival of patients with lung adenocarcinoma.

Human tumors are diverse in their natural history and response to treatment, which in part results from genetic and transcriptomic heterogeneity. In clinical practice, single-site needle biopsies are used to sample this diversity, but cancer biomarkers may be confounded by spatiogenomic heterogeneity within individual tumors. Here we investigate clonally expressed genes as a solution to the sampling bias problem by analyzing multiregion whole-exome and RNA sequencing data for 450 tumor regions from 184 patients with lung adenocarcinoma in the TRACERx study.

A reform of value-added taxes on foods can have health, environmental and economic benefits in Europe.

Fiscal policies can provide important incentives for encouraging the dietary changes needed to achieve global policy targets. Across Europe, the foods relevant to health and the environment often incur reduced but non-zero value-added tax (VAT) rates at about half the maximum rates, which allows for providing both incentives and disincentives. Integrating economic, health and environmental modelling, we show that reforming VAT rates on foods, including increasing rates on meat and dairy, and reducing VAT rates on fruits and vegetables can improve diets and result in health, environmental and economic benefits in most European countries.

Alkaline phosphatase decline and pain response as predictors of overall survival benefit in patients treated with radium-223: a post hoc analysis of the REASSURE study.

Background: Alkaline phosphatase (ALP) declines and pain responses can occur during radium-223 (Ra) treatment, but their association with treatment outcomes is unclear.

Methods: For patients with metastatic castration-resistant prostate cancer treated with Ra in the REASSURE study, we investigated whether ALP decline (Week 12) and/or pain response (during treatment) are associated with improved overall survival (OS). The Brief Pain Inventory-Short Form (BPI-SF) was used to assess pain at baseline and pain response (in patients with baseline BPI-SF score ≥2).

The therapeutic potential of circular RNAs.

Over the past decade, research into circular RNA (circRNA) has increased rapidly, and over the past few years, circRNA has emerged as a promising therapeutic platform. The regulatory functions of circRNAs, including their roles in templating protein translation and regulating protein and RNA functions, as well as their unique characteristics, such as increased stability and a favourable immunological profile compared with mRNAs, make them attractive candidates for RNA-based therapies. Here, we describe the properties of circRNAs, their therapeutic potential and technologies for their synthesis.

Significant progress has been made through the optimization of modelling and device architecture solar cells has proven to be a valuable and highly effective approach for gaining a deeper understanding of the underlying physical processes in solar cells. Consequently, this research has conducted a two-dimensional (2D) perovskite solar cells (PSCs) simulation to develop an accurate model. The approach utilized in this study is based on the finite element method (FEM).

Modelling mixed crop-livestock systems and climate impact assessment in sub-Saharan Africa.

Climate change significantly challenges smallholder mixed crop-livestock (MCL) systems in sub-Saharan Africa (SSA), affecting food and feed production. This study enhances the SIMPLACE modeling framework by incorporating crop-vegetation-livestock models, which contribute to the development of sustainable agricultural practices in response to climate change. Applying such a framework in a domain in West Africa (786,500 km) allowed us to estimate the changes in crop (Maize, Millet, and Sorghum) yield, grass biomass, livestock numbers, and greenhouse gas emission in response to future climate scenarios.

First out-of-bed mobilisation in adults with severe acquired brain injury in Scandinavian neurointensive care units: A survey of current clinical practice (FOOBScan).

Background: The harm-benefit balance for early out-of-bed mobilisation of patients with severe acquired brain injury (ABI) in neurointensive care units (neuro-ICUs) is unclear, and there are no clinical guidelines. This study aimed to survey the current clinical practice and perceptions among clinicians involved in first out-of-bed mobilisation in Scandinavian neuro-ICUs.

Methods: This was a cross-sectional, anonymous, web-based survey; the reporting follows the recommended CROSS checklist.

A new hypothesis to explain disease dominance.

The onset and progression of dominant diseases are thought to result from haploinsufficiency or dominant negative effects. Here, we propose transcriptional adaptation (TA), a newly identified response to mRNA decay, as an additional cause of some dominant diseases. TA modulates the expression of so-called adapting genes, likely via mRNA decay products, resulting in genetic compensation or a worsening of the phenotype.

Sex hormone-dependent host-microbiome interactions and cardiovascular risk (XCVD): design of a longitudinal multi-omics cohort study.

Introduction: Cardiovascular diseases (CVDs) present differently in women and men, influenced by host-microbiome interactions. The roles of sex hormones in CVD outcomes and gut microbiome in modifying these effects are poorly understood. The XCVD study examines gut microbiome mediation of sex hormone effects on CVD risk markers by observing transgender participants undergoing gender-affirming hormone therapy (GAHT), with findings expected to extrapolate to cisgender populations.

Peer reviewers' conflicts of interest in biomedical research: scoping review.

Background: Peer review may improve the quality of research manuscripts and aid in editorial decisions, but reviewers can have conflicts of interest that impact on their recommendations.

Objectives: The objective was to systematically map and describe the extent and nature of empirical research on peer reviewers' conflicts of interest in biomedical research.

Design: Scoping review METHODS: In this scoping review, we included studies investigating peer reviewers' conflicts of interest in journal manuscripts, theses and dissertations, conference abstracts, funding applications and clinical guidelines.

Pre-biologic disease trajectories are associated with morbidity burden and biologic treatment response in severe asthma.

Background: Biologics can induce remission in some patients with severe asthma, however, little is known about pre-biologic disease trajectories and their association with outcomes from biological treatment.
