  • Cryptococcal meningitis is a major cause of death in people with HIV/AIDS, but there's still a limited understanding of how the fungus changes during infection.
  • A study analyzed the whole genomes of 372 clinical isolates from patients in Malawi and Cameroon, revealing that Cameroonian isolates are more genetically uniform compared to their Malawian counterparts, and show different rates of mutations in key genes.
  • Longitudinal samples taken from patients in Cameroon highlighted genetic changes during infection and revealed mixed infections in 13% of patients, showcasing the evolutionary dynamics of the fungus in this context.

Background: Drug hypersensitivity reactions (DHRs) to platinum-based drugs are heterogenous and restrict their access, and drug desensitization (DD) has provided a ground-breaking procedure for their re-introduction, although the response is heterogeneous. We aimed to identify the phenotypes, endotypes, and biomarkers of reactions to carboplatin and oxaliplatin and their response to DD.

Methods: Seventy-nine patients presenting with DHRs to oxaliplatin (N = 46) and carboplatin (N = 33) were evaluated at the Allergy Departments of two tertiary care hospitals in Spain.

  • - Alzheimer’s disease is the leading cause of dementia globally, and recent therapies targeting amyloid and tau proteins show promise, with three drugs (lecanemab, donanemab, and aducanumab) nearing market release.
  • - A systematic review of 165 clinical trials revealed 40 published studies on anti-amyloid and anti-tau therapies, with several showing efficacy in reducing amyloid and tau levels in the brain.
  • - The discussion emphasizes that while anti-amyloid therapies dominate, effective patient selection for these drugs may depend on genetic factors, such as the presence of the apolipoprotein E gene's fourth allele.
