Article Synopsis
  • - Researchers found that IL-34 plays a role in resistance to immune therapies in mice and wanted to see if this holds true in human tumors as well.
  • - They used a specific method to select an IL-34-neutralizing antibody and transplanted human tumor tissue that expresses both IL-34 and PD-L1 into mice that have an immune system similar to humans.
  • - This patient-derived xenograft model is designed to study how neutralizing IL-34 affects the immune response when combined with immune checkpoint blockade in human cancers, with more detailed information available from Hama et al. (2020).

Evolution and insights into the structure and function of the DedA superfamily containing TMEM41B and VMP1.

J Cell Sci

April 2021

Department of Biochemistry and Molecular Biology, Graduate School of Medicine, The University of Tokyo, Tokyo 113-0033, Japan.

TMEM41B and VMP1 are endoplasmic reticulum (ER)-localizing multi-spanning membrane proteins required for ER-related cellular processes such as autophagosome formation, lipid droplet homeostasis and lipoprotein secretion in eukaryotes. Both proteins have a VTT domain, which is similar to the DedA domain found in bacterial DedA family proteins. However, the molecular function and structure of the DedA and VTT domains (collectively referred to as DedA domains) and the evolutionary relationships among the DedA domain-containing proteins are largely unknown.

Three-dimensional (3D) hydrogel serves as a platform to identify potential markers of chondrocyte dedifferentiation by combining RNA sequencing.

Bioact Mater

September 2021

Institute for Translational Medicine, The Affiliated Hospital of Qingdao University, College of Medicine, Qingdao University, Qingdao, 266021, Shandong, PR China.

Dedifferentiation of chondrocyte greatly restricts its function and application, however, it is poorly understood except a small number of canonical markers. The non-cell-adhesive property endows polysaccharide hydrogel with the ability to maintain chondrocyte phenotype, which can serve as a platform to identify new molecular markers and therapeutic targets of chondrocyte dedifferentiation. In this study, the high-throughput RNA sequencing (RNA-seq) was first performed on articular chondrocytes at primary (P0) and passage 1 (P1) stages to explore the global alteration of gene expression along with chondrocyte dedifferentiation.

Interleukin-34 Limits the Therapeutic Effects of Immune Checkpoint Blockade.

iScience

October 2020

Division of Immunobiology, Institute for Genetic Medicine, Hokkaido University, Kita-15 Nishi-7, Sapporo 060-0815, Japan.

Interleukin-34 (IL-34) is an alternative ligand to colony-stimulating factor-1 (CSF-1) for the CSF-1 receptor that acts as a key regulator of monocyte/macrophage lineage. In this study, we show that tumor-derived IL-34 mediates resistance to immune checkpoint blockade regardless of CSF-1 existence in various murine cancer models. Consistent with its immunosuppressive characteristics, the expression of IL-34 in tumors correlates with decreased frequencies of cellular (such as CD8 and CD4 T cells and M1-biased macrophages) and molecular (including various cytokines and chemokines) effectors at the tumor microenvironment.
