We have developed the GReedy Accumulated strategy for Protein Engineering (GRAPE) to improve enzyme stability across various applications, combining advanced computational methods with a unique clustering and greedy accumulation approach to efficiently explore epistatic effects with minimal experimental effort. To make this strategy accessible to nonexperts, we introduced GRAPE-WEB, an automated, user-friendly web server that allows the design, inspection, and combination of stabilizing mutations without requiring extensive bioinformatics knowledge. GRAPE-WEB's robust performance and accessibility provide a comprehensive and adaptable approach to protein thermostability design, suitable for both newcomers and experienced practitioners in the field.

Smaller microorganisms outcompete larger ones in resistance and functional effects under disturbed agricultural ecosystems.

Body size is a key ecological trait of soil microorganisms related to their adaptation to environmental changes. In this study, we reveal that the smaller microorganisms show stronger community resistance than larger organisms in both maize and rice soil. Compared with larger organisms, smaller microorganisms have higher diversity and broader niche breadth to deploy survival strategies, because of which they are less affected by environmental selection and thus survive in complex and various kinds of environments.

Hybridization is widespread in nature and is a valuable tool in domestic breeding. The pre-initiation complex (pre-IC) has been proposed for two decades as an intermediate right before the maturation of the eukaryotic DNA replication fork. However, its existence and biochemical nature remain enigmatic. Here, through combining several enrichment strategies, we are able to isolate an endogenous dimeric CMG-containing complex (designated as d-CMG) distinct from traditional single CMG (s-CMG) and in vitro reconstituted dimeric CMG.

Chinese indigenous chickens (CICs) constitute world-renowned genetic resources due to their excellent traits, including early puberty, good meat quality and strong resistance to disease. Unfortunately, the introduction of a large number of commercial chickens in the past two decades has had an adverse effect on CICs. Using the chicken 60 K single nucleotide polymorphism chip, we assessed the genetic diversity and population structure of 1,187 chickens, representing eight Chinese indigenous chicken breeds, two hybrid chicken breeds, two ancestral chicken breeds, two commercial populations and additional red jungle fowl.

Smooth muscle myosin heavy chain (SM-MHC) is exclusively expresses in smooth muscle, which takes part in smooth muscle cell contraction. Here, we used an insertional mutation mouse whose heavy polypeptide 11 () gene has been disrupted and no SM-MHC protein has been detected. Compared to the wild-type and SM-MHC mice, the SM-MHC neonates had large round bellies, thin-walled giant bladders, and large stomachs with huge gas bubbles.
