Molecular Imaging and Therapy of Differentiated Thyroid Carcinoma in Adults.

Cancer J

May 2024

Assistant Professor, Director of Therapeutic Nuclear Medicine, Department of Radiology, UNM School of Medicine, MSC10 5530, 1 University of New Mexico, Albuquerque, NM 87131.

Differentiated thyroid carcinoma (DTC) has been increasing in incidence in the United States over the last several decades, although mortality rates have remained low. Radioactive iodine therapy (RAI-T) has been a mainstay of treatment for DTC since the 1940s. Imaging of DTC before and after RAI-T primarily focuses on molecular imaging of the sodium iodide symporter.

Background: Polygenic Risk Scores (PRS) based on results from genome-wide association studies offer the prospect of risk stratification for many common and complex diseases. We developed a PRS for alcohol-associated cirrhosis by comparing single-nucleotide polymorphisms among patients with alcohol-associated cirrhosis (ALC) versus drinkers who did not have evidence of liver fibrosis/cirrhosis.

Methods: Using a data-driven approach, a PRS for ALC was generated using a meta-genome-wide association study of ALC (N=4305) and an independent cohort of heavy drinkers with ALC and without significant liver disease (N=3037).

Objective: Gene therapy by convection-enhanced delivery of type 2 adeno-associated virus-glial cell derived neurotrophic factor (AAV2-GDNF) to the bilateral putamina seeks to increase GDNF gene expression and treat Parkinson's disease (PD).

Methods: A 63-year-old man with advanced PD received AAV2-GDNF in a clinical trial. He died from pneumonia after anterior cervical discectomy and fusion 45 months later.

Only ~20% of heavy drinkers develop alcohol cirrhosis (AC). While differences in metabolism, inflammation, signaling, microbiome signatures and genetic variations have been tied to the pathogenesis of AC, the key underlying mechanisms for this interindividual variability, remain to be fully elucidated. Induced pluripotent stem cell-derived hepatocytes (iHLCs) from patients with AC and healthy controls differ transcriptomically, bioenergetically and histologically.

Professor of Oncology, Johns Hopkins University School of Medicine, Clinical Director, BMT Program, The Sidney Kimmel Comprehensive Cancer Center at Johns Hopkins, Baltimore, Maryland, USA.

Allogeneic bone marrow transplantation is a curative intervention for both neoplastic and non-malignant conditions. However, not all patients have an HLA-matched donor. Therefore, the development of an approach that expand the donor pool was of paramount relevance.

PCSK9 inhibition attenuates alcohol-associated neuronal oxidative stress and cellular injury.

Brain Behav Immun

July 2024

Section on Clinical Genomics and Experimental Therapeutics, National Institute on Alcohol Abuse and Alcoholism, National Institutes of Health, Bethesda, MD, United States. Electronic address:

Alcohol Use Disorder (AUD) is a persistent condition linked to neuroinflammation, neuronal oxidative stress, and neurodegenerative processes. While the inhibition of proprotein convertase subtilisin/kexin type 9 (PCSK9) has demonstrated effectiveness in reducing liver inflammation associated with alcohol, its impact on the brain remains largely unexplored. This study aimed to assess the effects of alirocumab, a monoclonal antibody targeting PCSK9 to lower systemic low-density lipoprotein cholesterol (LDL-C), on central nervous system (CNS) pathology in a rat model of chronic alcohol exposure.

  • GM1 gangliosidosis (GM1) is a lysosomal disorder caused by mutations in the GLB1 gene, leading to serious neurodegeneration and currently has no approved treatments.
  • A study involving 41 patients with type II GM1 revealed distinct genetic variants and observed that many children had normal hearing and did not exhibit typical symptoms seen in type I GM1.
  • The research highlighted progressive brain atrophy and identified crucial correlations between brain chemistry changes and patient behavior scores, emphasizing the importance of this long-term study for improving diagnosis and developing future therapies.

Background: Lymphoedema is a progressive condition causing significant alterations to life, exerting pressures on unscheduled care from complications including cellulitis and wounds. An on the ground education programme (OGEP) was implemented to raise knowledge, competence and confidence in lymphoedema management in community clinical services. The aim of this study was to explore the impact and outcomes of the OGEP intervention.

Objective: To describe the frequency of GHE in a large cohort of patients with CNC, and to identify genotype-phenotype correlations.

Methods: Patients with CNC with at least one biochemical evaluation of GH secretion at our center from 1995-2021 (n=140) were included in the study.

Background: Alcohol cessation is the only intervention that both prevents and halts the progressions of alcohol-associated liver disease. The aim of this study was to assess the relationship between a return to alcohol use and consultation with hepatology in treatment-seeking patients with alcohol use disorder (AUD).

Methods: Two hundred forty-two patients with AUD were enrolled in an inpatient treatment program, with hepatology consultation provided for 143 (59%) patients at the request of the primary team.

Background: De-escalation strategies for newly-diagnosed p16-positive oropharyngeal squamous cell carcinoma (p16+ OPSCC), aim to reduce treatment-related morbidity without compromising disease control. One strategy is neoadjuvant cisplatin and docetaxel chemotherapy (NAC + S) before transoral robotic surgery, with pathology-based risk-adapted adjuvant treatment.

Methods: We examined the recurrence-free survival (RFS) for patients who received NAC + S.

Comparison of clinical outcomes between single metal-ceramic and zirconia crowns.

J Prosthet Dent

April 2024

Associate Professor and Clinical Director, Craniofacial Prosthetics Unit, Sunnybrook Health Sciences Centre, Toronto, Ontario, Canada; Associate Professor, Department of Prosthodontics, Faculty of Dentistry, University of Toronto, Toronto, Ontario, Canada.

Statement Of Problem: Evidence comparing the survival of zirconia crowns with metal-ceramic crowns is sparse. Knowledge of their survival and a comparison of their clinical outcomes would improve clinical decision making.

Purpose: The purpose of this university-based study was to compare the survival, failures, biological and technical complications encountered with zirconia and metal-ceramic crowns restored and followed up over a similar period.

The relationship between pain and alcohol use disorder (AUD) is complex and bidirectional. The current study examines risk factors for pain in a large comprehensively phenotyped sample including individuals from across the spectrum of alcohol use and misuse. Participants (n = 1101) were drawn from the National Institute on Alcohol Abuse and Alcoholism Natural History Protocol and included treatment-seeking AUD inpatients (AUD+Tx, n = 369), individuals with AUD not seeking treatment (AUD+, n = 161), and individuals without AUD (AUD-, n = 571).

Subcortico-Cortical Dysconnectivity in ADHD: A Voxel-Wise Mega-Analysis Across Multiple Cohorts.

Am J Psychiatry

June 2024

Office of the Clinical Director, NIMH, Bethesda, Md. (Norman, Shaw); Section on Neurobehavioral and Clinical Research, Social and Behavioral Research Branch, National Human Genome Research Institute, NIH, Bethesda, Md. (Sudre, Price, Shaw).

Article Synopsis
  • A mega-analysis of six neuroimaging datasets investigated the relationship between ADHD and subcortico-cortical connectivity, involving 1,696 youths with ADHD and 6,737 control subjects.
  • The study found that youths with ADHD exhibited increased connectivity between striatal seeds and various brain regions, particularly the caudate, and also between the amygdala and dorsal anterior cingulate cortex.
  • Although these results support links between ADHD and brain connectivity, the effect sizes were small, suggesting that resting-state subcortico-cortical circuits may not fully explain the complexities of ADHD pathology.
