Hepatitis D virus (HDV) depends on hepatitis B virus (HBV) to enter and exit hepatocytes and to replicate. Despite this dependency, HDV can cause severe liver disease. HDV accelerates liver fibrosis, increases the risk of hepatocellular carcinoma, and hastens hepatic decompensation compared to chronic HBV monoinfection.

Osteopontin is a prognostic circulating biomarker in patients with neuroendocrine neoplasms.

J Cancer Res Clin Oncol

September 2023

Department of Hepatology and Gastroenterology, Campus Virchow Clinic and Campus Charité Mitte, Charité University Medicine, Berlin, Germany.

Article Synopsis
  • The study investigates the role of Osteopontin (OPN) as a potential biomarker in patients with neuroendocrine neoplasms (NEN), given its elevated levels in various cancers and its involvement in tumor behavior.
  • Plasma OPN concentrations were analyzed in 38 patients with NEN at different stages of their disease and compared to healthy controls, revealing significantly higher levels in NEN patients, especially in high-grade tumors.
  • The findings suggest that high initial OPN levels (above 200 ng/ml) are linked to worse prognosis, indicating shorter progression-free survival, making OPN a promising prognostic biomarker for NEN patients.

Background/aims: Cells adapt to chronic extracellular hypotonicity by altering metabolism. Corresponding effects of sustained hypotonic exposure at the whole-person level remain to be confirmed and characterized in clinical and population-based studies. This analysis aimed to 1) describe changes in urine and serum metabolomic profiles associated with four weeks of sustained > +1 L/d drinking water in healthy, normal weight, young men, 2) identify metabolic pathways potentially impacted by chronic hypotonicity, and 3) explore if effects of chronic hypotonicity differ by type of specimen and/or acute hydration condition.

Update on Locoregional Therapies for Cholangiocellular Carcinoma.

Cancers (Basel)

April 2023

Department of Diagnostic and Interventional Radiology, Medical Faculty, University Dusseldorf, D-40225 Düsseldorf, Germany.

Locoregional therapy options for CCA are used, in particular, for non-resectable tumors and aim to reduce tumor viability or delay tumor growth and ultimately prolong overall survival. In addition to local ablative procedures such as radiofrequency- or microwave-ablation, transarterial procedures such as transarterial embolization (TAE), transarterial chemoembolization (TACE), or selective internal radiotherapy (SIRT) play a major role. In particular, in combination with advances in molecular medicine and immunotherapy, there has been a further development in the therapy of primary malignant liver tumors in recent years.

Genetic knockout of porcine GGTA1 or CMAH/GGTA1 is associated with the emergence of neo-glycans.

Xenotransplantation

November 2023

Leibniz Research Laboratories for Biotechnology and Artificial Organs (LEBAO), Hannover Medical School, Hannover, Germany.

Background: Pig-derived tissues could overcome the shortage of human donor organs in transplantation. However, the glycans with terminal α-Gal and Neu5Gc, which are synthesized by enzymes, encoded by the genes GGTA1 and CMAH, are known to play a major role in immunogenicity of porcine tissue, ultimately leading to xenograft rejection.

Methods: The N-glycome and glycosphingolipidome of native and decellularized porcine pericardia from wildtype (WT), GGTA1-KO and GGTA1/CMAH-KO pigs were analyzed by multiplexed capillary gel electrophoresis coupled to laser-induced fluorescence detection.

Intratumoral dendritic cells and T cells predict survival in gastroenteropancreatic neuroendocrine neoplasms.

Endocr Relat Cancer

July 2023

Department of Hepatology and Gastroenterology, Charité Universitaetsmedizin Berlin, Campus Virchow-Klinikum and Campus Charité Mitte, Berlin, Germany.

Clinical management of gastroenteropancreatic neuroendocrine neoplasms remains challenging. We recently introduced the FMS-like tyrosine kinase 3 ligand (FLT3LG) as a possible biomarker for a proinflammatory tumor microenvironment. Here, we put a spotlight on the quantitative assessment of classical dendritic cells (cDC) and T cells in the context of FLT3LG mRNA levels in a retrospective study on neuroendocrine tumor (NET) G2/G3 and neuroendocrine carcinoma (NEC) of pancreatic and gastric origin.

Background: Progress of cholangitis to cholangiosepsis is a frequent observation in patients with secondary sclerosing cholangitis in critically ill patients (SSC-CIP). Adequate biliary drainage may reduce episodes of cholangiosepsis and therefore stabilize liver function and improve survival. The primary objective of the BISCIT study is to demonstrate that scheduled biliary interventions will reduce incidence of cholangiosepsis, liver transplantation, or death in patients with SSC-CIP.

Article Synopsis
  • The study focuses on improving the understanding of "curative therapies" for hepatocellular carcinoma (HCC) by developing a statistical model to predict the chances of a cure after treatment.
  • Researchers analyzed data from 1571 HCC patients treated with radiofrequency or microwave ablation to determine the success rate of these curative treatments.
  • The findings indicated a cure rate of around 18.3%, which can increase to about 40% in patients with good liver function and small tumors, and also led to the creation of a calculator for clinicians to estimate individual patient cure probabilities.
