Background: Type 2 diabetes (T2D) and older age are well-known risk factors for dementia. Indeed, there is evidence that older adults not diagnosed, but at-risk for T2D can show early signs of cognitive decline, further exacerbated by excessive body weight or high blood glucose levels. Such a finding would have implications for early treatment strategies; however, the evidence is still sparse.

Background: Frontotemporal dementia (FTD) is a diverse spectrum of neurodegenerative disorders impacting the frontal and temporal lobes, primarily manifesting as behavioral and/or language issues. Accurate diagnosis is crucial for effective management, potential treatment, and care planning. While speech analysis has shown promise in detecting cognitive markers of Alzheimer's disease (AD), its exploration in FTD is limited.

Background: Autonomic dysfunction has been linked to empathy deficits in symptomatic frontotemporal degeneration (FTD), but less is known about pre-symptomatic FTD mutation carriers (preFTD+). Our prior work found that increasing resting heart rate (RHR) over time predicts decline in emotional empathy in preFTD+. Here, we replicate previous findings in a large, multi-site consortium sample and assess relationships between RHR and empathy loss across disease stages.

Background: Lewy body (LB) diseases can present with overlapping prodromal, cognitive, motor, autonomic or neuropsychiatric symptoms. Intuitively, greater symptom severity should correlate with greater pathological burden, but this has not been consistently shown. LB pathology does not translate to clinical expression in Incidental LB disease.

Background: Language impairment is common in progressive supranuclear palsy (PSP) and is often overlooked due to the severity of the motor symptoms. We investigated whether language can be used to predict PSP prognosis.
Methods: One hundred-forty-six patients with a diagnosis of possible or probable PSP from the Tilavonemab (ABBV-8E12) clinical trial were evaluated at baseline and week 32 using the Repeatable Battery for the Assessment of Neuropsychological Status (RBANS), the PSP rating scale (PSPRS) and the Schwab and England Activities of Daily Living Scale (SEADL).
Background: Test-retest reliability for existing cognitive screening tests is typically poor and most have ceiling effects and restricted score ranges that mask the presence of subtle decline. The Brief Assessment of Cognitive Health (BACH) is a computerized cognitive screening tool that patients complete independently. It includes a complex memory test without ceiling effects and brief mood and history questions.

Background: Psychological factors such as repetitive negative thinking, proneness to experience distress, and perceived stress are associated with increased risk of neurodegeneration and clinical dementia, whereas having a sense of life-purpose, self-reflection, and dispositional mindfulness may be protective. However, whether combinations of these risk and protective factors may inform distinct psychological profiles, which may be differential associated with age-related health outcomes is currently unknown.
Method: We included 742 middle-aged (mean age 51.

Percutaneous tumor ablation has become a widely accepted and used treatment option for both soft and hard tissue malignancies. The current standard-of-care techniques for performing these minimally invasive procedures require providers to navigate a needle to their intended target using two-dimensional (2D) US or CT to obtain complete local response. These traditional image-guidance systems require operators to mentally transpose what is visualized on a 2D screen into the inherent three-dimensional (3D) context of human anatomy.

Background: The ALLFTD (ARTFL-LEFFTDS Longitudinal Frontotemporal Lobar Degeneration) study is an NIH-funded effort to prepare for clinical trials in sporadic (s-FTLD) and familial (f-FTLD) FTLD syndromes by characterizing cohorts, developing new clinical trial outcome measures, and evaluating disease progression. To understand disease trajectories in the context of potential preventative or disease-modifying therapeutic agents, comprehensive evaluation across multiple time-points is crucial.
Method: ALLFTD evaluates participants with FTLD spectrum disorders (bvFTD, svPPA, nfvPPA, FTD-ALS, CBS, PSP), with strong family histories of FTLD, or known FTLD-associated genetic variants within the family.

Background: The Uniform Data Set (UDS) neuropsychological battery, administered across Alzheimer's Disease Centers (ADC), includes memory tests but lacks a list-learning paradigm. ADCs often supplement the UDS with their own preferred list-learning task. Given the importance of list-learning for characterizing memory, we aimed to develop a harmonized memory score that incorporates UDS memory tests while allowing centers to contribute differing list-learning tasks.

Background: Amnestic initial symptoms are common in Alzheimer's disease(AD). However initial non-amnestic (language, executive or visuospatial) symptoms are seen in ∼20% and associated with faster progression. Given racial differences in clinical and biomarker characteristics in AD, we explored if initial cognitive symptoms differed by race of the participants among autopsy confirmed AD, Lewy body pathology (LBP) and mixed pathology (AD-LBP).

Introduction: Identifying tuberculosis infection (TBI) using interferon-gamma release assays (IGRAs) is a primary component of clinical and public health efforts to prevent pediatric tuberculosis. Pediatric data comparing the two IGRAs in the United States are very limited. We compared the performance of the two IGRAs among a large pediatric cohort tested for TBI and assessed whether discordance might be due to quantitative results close to test cut-off values.
