Retinal ganglion cell vulnerability to pathogenic tau in Alzheimer's disease.

bioRxiv

September 2024

Department of Neurosurgery, Maxine Dunitz Neurosurgical Research Institute, Cedars-Sinai Medical Center, Los Angeles, CA, USA.

Article Synopsis
  • Pathological tau isoforms, particularly hyperphosphorylated tau at serine 396, and tau oligomers were found in the retinas of patients with mild cognitive impairment (MCI) and Alzheimer's disease (AD), indicating a potential link between tauopathy and retinal changes.
  • The study analyzed retinal cross-sections from 25 patients with MCI or AD and 16 cognitively normal controls, revealing a significant reduction in retinal ganglion cells (RGCs) and increased signs of cell distress in MCI and AD patients compared to controls.
  • Findings showed that higher amounts of pS396-tau in RGCs were strongly correlated with decreased RGC integrity and related to severity in cognitive decline, suggesting that retinal

Purpose: Recent advancements in the management of biochemical recurrence (BCR) following local treatment for prostate cancer (PCa), including the use of androgen receptor signaling inhibitors (ARSIs), have broadened the spectrum of therapeutic options. We aimed to compare salvage therapies in patients with BCR after definitive local treatment for clinically non-metastatic PCa with curative intent.

Methods: In October 2023, we queried PubMed, Scopus, and Web of Science databases to identify randomized controlled trials (RCTs) and prospective studies reporting data on the efficacy of salvage therapies in PCa patients with BCR after radical prostatectomy (RP) or radiation therapy (RT).

Article Synopsis
  • Liposomal amphotericin B (L-AmB) is a key treatment for severe fungal infections, but it may pose risks for kidney damage in critically ill patients.
  • A study analyzed 67 patients treated with L-AmB for over 48 hours to determine the incidence of acute kidney injury (AKI), revealing that 26.8% of patients developed AKI, with lower rates in those at higher initial risk.
  • Key factors contributing to AKI included norepinephrine use, SOFA scores, and certain laboratory results, indicating that while L-AmB appears safe, AKI development is influenced by multiple factors.

Background: The IDENTIFY study developed a model to predict urinary tract cancer using patient characteristics from a large multicentre, international cohort of patients referred with haematuria. In addition to calculating an individual's cancer risk, it proposes thresholds to stratify them into very-low-risk (<1%), low-risk (1-<5%), intermediate-risk (5-<20%), and high-risk (≥20%) groups.

Objective: To externally validate the IDENTIFY haematuria risk calculator and compare traditional regression with machine learning algorithms.
