March 2020

Calgary Stroke Program, Departments of Clinical Neurosciences, Radiology and Community Health Sciences, Hotchkiss Brain Institute, Cumming School of Medicine, University of Calgary, Calgary, Alberta, Canada.

Article Synopsis
  • - The study aimed to assess how different measures of blood clot permeability on non-contrast CT and CT angiography relate to the effectiveness of intravenous alteplase in achieving recanalization in patients with anterior circulation occlusion.
  • - Four hundred eighty patients were analyzed, revealing that various measuring methods had similar abilities to distinguish between those who experienced recanalization and those who did not, with a notable finding that a specific measurement threshold (HU ≥ 89) offered optimal discrimination.
  • - Ultimately, the research concluded that simpler methods for gauging thrombus permeability are just as effective as more sophisticated techniques in identifying patients likely to achieve recanalization.

Article Synopsis
  • The study investigates how genetic variants in a specific gene region contribute to ethnic differences in the risk of Alzheimer's disease (AD) associated with the ε4 allele among various populations, including East Asians, Europeans, and African Americans.
  • Analysis of 19,398 East Asian individuals and 15,836 individuals of European ancestry revealed that certain SNPs, particularly rs405509, significantly impact AD risk, particularly among ε4 homozygotes, with risk increasing in a dose-dependent manner.
  • Results suggest that higher expression linked to specific genotypes might reduce AD risk, highlighting the importance of genetic variation in understanding ethnic disparities in AD association with the ε4 allele.

A peptide containing Noxa mitochondrial-targeting domain induces cell death via mitochondrial and endoplasmic reticulum disruption.

Biochem Biophys Res Commun

October 2019

Department of Biochemistry and Molecular Biology, Chosun University School of Medicine, 309 Pilmoon-Daero, Dong-Gu, Gwangju, 61452, Republic of Korea. Electronic address:

Noxa is a weak apoptosis activator consisting of a BH3 domain and a mitochondrial-targeting domain (MTD). BH3 binds Mcl-1 and Bcl2A1 and inactivates their anti-apoptotic activities, while MTD delivers BH3 to mitochondria. Previously we revealed that MTD may also function as an inducer of necrosis via conjugation with octa-arginine, which induces cytosolic Ca influx from mitochondria.

During ligand-mediated receptor endocytosis, the small GTPase Rab5 functions in vesicle fusion and trafficking. Rab5 activation is known to require interactions with its guanine nucleotide-exchange factors (GEFs); however, the mechanism regulating Rab5 interactions with GEFs remains unclear. Here, we show that the SH3-adapter protein SPIN90 participates in the activation of Rab5 through the recruitment of both Rab5 and its GEF, Gapex5, to endosomal membranes during epidermal growth factor (EGF)-mediated endocytosis.

Noxa mitochondrial targeting domain induces necrosis via VDAC2 and mitochondrial catastrophe.

Cell Death Dis

July 2019

Department of Biochemistry and Molecular Biology, Chosun University School of Medicine, 309 Pilmoon-Daero, Dong-Gu, Gwang-Ju, 61452, Korea.

Noxa, a Bcl-2 homology 3 (BH3)-only protein of the Bcl-2 family, is responsive to cell stresses and triggers apoptosis by binding the prosurvival Bcl-2-like proteins Mcl1, Bcl, and Bcl2A1. Although the Noxa BH3 domain is necessary to induce apoptosis, the mitochondrial targeting domain (MTD) of Noxa functions as a pronecrotic domain, an inducer of mitochondrial fragmentation, and delivery to mitochondria. In this study, we demonstrate that the extended MTD (eMTD) peptide induces necrotic cell death by interaction with the VDAC2 protein.

BMAP-27, a member of cathelicidin family, plays an important role against microorganisms, including bacteria and fungi. BMAP-27 may exert antimicrobial effects through membrane integrity disruption, but the exact molecular mechanism remains unclear. To identify the structural features important for antimicrobial activity and propose a mechanism underlying antibacterial effects, we determined the nuclear magnetic resonance structure of BMAP-27 in a membrane-mimetic environment and investigated its interactions with lipid membranes.

Background: Spontaneous isolated posterior inferior cerebellar artery (PICA) dissection has been reported more frequently since high-resolution vessel wall magnetic resonance imaging (HR vw-MRI) was introduced to the field. The intimal flap or double lumen, which is commonly reported to be a direct sign of the dissection, is not easily detectable on HR vw-MRI because the size of the PICA is very small and tortuous.

Case Presentation: Two patients with posterior circulation ischemic stroke due to spontaneous isolated PICA dissection underwent HR vw-MRI.
