July 2008

Department of Pathology, Chongqing University of Medical Sciences, Chongqing 400016, China.

Objective: To investigate whether the deletion of PTEN affects the expression of Cu/Zn SOD and the related biology.

Methods: Protein and mRNA expression levels of PTEN, P-Akt, Cu/Zn SOD in the control immortalized wild type mouse embryonic fibroblast cells (PTEN+/+) and PTEN-null cells (PTEN-/-) were evaluated by Western blot and Northern blot respectively. The level of superoxide anions were detected using fluorescent probes.

Characteristics of circulating T cell receptor gamma-delta T cells from individuals chronically infected with hepatitis B virus (HBV): an association between V(delta)2 subtype and chronic HBV infection.

J Infect Dis

December 2008

Key Laboratory of Molecular Biology for Infectious Diseases, People's Republic of China Ministry of Education, Institute for Viral Hepatitis, Chongqing University of Medical Sciences, No. 74 Lin Jiang Road, Chongqing, China.

Background: To date, few studies have been conducted to determine whether T cell receptor (TCR) gammadelta T cells are involved in hepatitis B virus (HBV) infection. This study was performed to assess the quantity and immune function of TCRgammadelta T cells in the blood of patients with chronic HBV infection and to analyze the relationship between proportions of TCRgammadelta T cells and both proportions of other immune cells and clinical parameters.

Methods: Flow cytometry was used to detect the proportions of TCRgammadelta T cells and other immune cells in the peripheral blood of 46 asymptomatic carriers (AsCs) of HBV, 95 patients with chronic hepatitis B (CHB), and 29 healthy donors (HDs).

Article Synopsis
  • The study investigates how 60% oxygen affects type II alveolar epithelial cells (AEC II) from premature rats, specifically focusing on oxidative damage and apoptosis.* -
  • AEC II cells were exposed to different conditions (air, hyperoxia, hyperoxia plus CGRP, and a combination with a CGRP antagonist) and various oxidative markers were measured after 24 hours.* -
  • Results showed that hyperoxia caused increased levels of reactive oxygen species and cell death, while CGRP significantly reduced these harmful effects, indicating its potential protective role.*

Objective: To investigate the inhibitory effect of hepatocyte growth factor (HGF) on the apoptosis of rat primary cultured hepatic stellate cells treated with platelet-derived growth factor BB (PDGF-BB).

Methods: Hepatic stellate cells were cultured and treated with PDGF-BB+HGF, HGF or nothing (controls). Apoptosis of these hepatic stellate cells was evaluated by AO/EB staining, TUNEL and flow cytometry.

Hepatocellular carcinoma (HCC) is an especially frequent malignancy in China. Radiofrequency ablation, percutaneous ethanol injection, transarterial chemoembolization, cryoablation, microwave coagulation, and laser-induced interstitial thermotherapy all offer potential local tumor control and occasionally achieve long-term disease-free survival. High-intensity focused ultrasound (HIFU), as a noninvasive therapy, can be applied to treat tumors that are difficult to treat with other techniques.

August 2008

Department of Orthopaedics, the Second Affiliated Hospital, Chongqing University of Medical Sciences, Chongqing, PR China.

Objective: To observe the effect of recombinant human osteoprotegerin (rhOPG) on osteoclasts which were stimulated by polyethylene particles and to investigate the feasibility of applying rhOPG for the prosthetic aseptic looseness.

Methods: The osteoclasts were isolated from the long bones of 5 New Zealand rabbits born within 24 hours, weighing 80-100 g, male or female, and were plated into the 24-well coverslips (10 mm x 10 mm) and bone slices (8 mm x 8 mm x 50 microm) at the density of 1 x 10(5)/mL. Based on the different concentration and density of rhOPG and polyethylene particles, the plates of culture were divided into 3 groups: the group with polyethylene particles of 1 x 109/mL (polyethylene group), the group co-cultured with polyethylene particles of 1 x 10(9)/mL and rhOPG of 100 ng/mL (polyethylene/rhOPG group) and the control group.

The theories of Shushu in the literature of Qin and Han periods are reorganized and expositions of foreigner scholars about ancient way of thought are consulted, and compared with the recordation about the ebb-flow of ying-qi and the length of meridians in ancient medical books to probe into the relationship between Shushu and these recordation. It is found that the ebb-flow of ying-qi and the length of meridians are completely conform with the principle of Shushu, i. e.

Primary hepatocellular carcinoma (HCC) is one of the most common cancers occurring in human, and there is strong epidemiological evidence suggesting that persistent hepatitis B virus (HBV) infection is the most important risk factor for its development.HBx gene was found to be a transactivator recently. Its continuous expression in hepatocytes may transactivate cellular genes which can play a certain role in development of HCC.

Objective: It has long been known that high intensity focused ultrasound (HIFU) can kill tissue through coagulative necrosis. However, it is only in recent years that practical clinical applications are becoming possible. Since the ribs have strong reflections to ultrasonic beams, they may affect the deposition of ultrasound energy, decreasing the efficacy of HIFU treatment and increasing the chance of adverse events when the intra-abdominal tumours concealed by ribs are treated.

Hepatitis B virus (HBV) replicates in most tumor tissues of patients with HBV-associated hepatocellular carcinoma (HCC). In the present study, we have shown that the expression of HBV in the HCC cell lines, HepG2 and Huh7, down-regulated the expression of MHC class I-related molecule A (MICA), a ligand of the NKG2D receptor. Inhibition of HBV expression by small interference RNAs (siRNAs) in HepG2.

Background: To assess the efficacy of topotecan, a topoisomerase I specific inhibitor in S-phase, the reagent-induced apoptosis and cytotoxicity as well as related proteins expression, had been preliminarily investigated in human hepatoblastoma HepG2 cells.

Methods: Microculture tetrazolium assay (MTT), HE staining, transmission electron microscopy (TEM), flow cytometry (FCM), quantitative immunocytochemistry (QI), gene tranfection and RNAi technology were employed to carry out the exploration.

Results: Topotecan could potently kill HepG2 cells via inducing apoptosis and demonstrated strong cytotoxicity in a time, dose-dependent manner with IC50 of about 95 mu g/L.
