To study single nucleotide polymorphisms (SNP) in A-922G, T-786C and G894T of endothelial nitric oxide synthase (NOS3) and to correlate the distribution of their allelic combinations with hypertension in Chinese Han nationality population, genomic DNA was isolated from venous blood leukocytes from 192 unrelated patients with hypertension (95 females and 97 males) and 122 healthy unrelated individuals (46 females and 76 males) as controls. SNPs of NOS3 A-922G, T-786C and G894T were genotyped by allele-specific primer (ASP) PCR. The distribution of genotype combinations of three SNPs was determined by clustering analysis.

Novel vaccines for the treatment of chronic HBV infection based on mycobacterial heat shock protein 70.

Vaccine

February 2006

Key Laboratory of Molecular Biology for Infectious Diseases, Ministry of Education, Institute for Viral Hepatitis, Chongqing University of Medical Sciences, PR China.

Immunogenic peptide-based vaccines can raise significant cellular immune responses. Although cytotoxic T lymphocytes (CTL) peptide epitopes are generally poor immunogens, heat shock protein 70 from Mycobacterium tuberculosis (TBhsp70) can overcome this problem since it is a potent adjuvant that links innate and adaptive immune responses. Our goal is to use TBhsp70 as an adjuvant for development of therapeutic vaccines for chronic Hepatitis B virus infection (HBV).

Bacillus Calmette-Guerin (BCG) vaccination can protect animals from asthma, but the effect of BCG on childhood asthma prevention is controversial in humans. To verify the hypothesis that the BCG anti-asthma effect in childhood might be reversed by a respiratory virus infection, newborn BALB/c mice were divided into five groups. Control and ovalbumin (OVA) groups were mock vaccinated and mock infected.

Sheng Wu Yi Xue Gong Cheng Xue Za Zhi

December 2005

Department of Biomedicine Engineering, Chongqing University of Medical Sciences, Chongqing 400016, China.

Based on theoretic analysis, a new method for measuring linearity of physiological instrument and its inner unit circuits is provided. In this method, the sawtooth wave signal, which includes a base wave and its odd-even times harmonic waves, is used to act as input signal, then the linearity of physiological instrument and its inner unit circuits can be measured by analyzing the shape and frequency property of the output signal. The results of experiment show this method is quick and convenient for measuring the linearity of physiological instrument and its inner unit circuits.

Chromosomal aneuploidy was a notable cytogenetic character in cancer cells. However, the mechanism of aneuploidy aberration is not clear up to the present. In order to probe into the mechanism of aneuploidy aberration of cancer cells, Centromeric dots (Cd) variation on chromosomes of BGC823 and A549 cells were studied by a simultaneous silver staining of both NOR and Cd from centromeric dots variation.

We have evaluated the effects of a 2 week treatment with pioglitazone (Pio, 4mg/kg x d) on hepatic and peripheral insulin sensitivity, plasma adiponectin, and resistin concentrations in lipid-infused rats. Lipid infusion caused a large (60% in 4h) decrease in whole-body insulin sensitivity. Hepatic and peripheral insulin resistance contributed about equally to the whole-body insulin resistance.

Purpose: To assess whether surgical manoeuvre or resection of lung cancer could lead to haematogenous dissemination of malignant cells. In the mean time, the relationship between the sequence of vessel ligation and the haematogenous dissemination of cancer cells during operation was determined.

Methods: Exploiting cytokeratin 19 (CK19)/carcinoembryonic antigen (CEA) mRNA as markers, 69 peripheral blood samples were collected from 23 consecutive patients with non-small cell lung cancer (NSCLC) who underwent surgical resection with curative intention in preoperative, intraoperative and postoperative period, respectively.

Parabiosis theory clearly expounds the change course of stimulatiom, namely, parabiotic equalization phase, parabiotic deregulation phase and parabiotic inhibition phase, and it can clearly explain that the effective stimulating amount is directly related to the reinforcing or reducing, and establishment of the therapeutic course. That is, the large effective stimulating amount is reducing method and the small effective stimulating amount is reinforcing method. In clinical application, for deficiency syndrome and chronic diseases, the effective intensity of acuti puncture should be smaller and the needle should be retained for 20-30 min, and for excess syndrome and acute disd eases, the effective stimulating intensity of acupuncture should be larger and the needle should be retained for longer time.

Background: RNA interference (RNAi), which has been demonstrated as having great potentional in the fields of gene function and gene therapy, was applied to inhibit the expression of some endogenous genes. Human telomerase reverse transcriptase (hTERT) is highly expressed in hepatocellular carcinoma cells.

Materials And Methods: In combination with DNA vector-based RNAi, quantitative real-time reverse transcription polymerase chain reaction, telomeric repeat amplification protocol-enzyme-linked immunosorbent assay, 3-(4,5 dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide assay, and xenograft tumor animal techniques, we first constructed three pTZU6 + 1-shRNA-hTERT vectors and their corresponding site-mutated vectors, then transfected them into hepatocarcinoma HepG2, SMMC-7721 cells, and normal liver L02 cells, respectively, injected them into xenograft hepatocarcinoma tumor tissues to induce RNAi, and then detected the alteration of cell and tumor proliferation, telomerase activity, hTERT, and c-myc expression in each treatment.
