Alzheimer's disease (AD), the most prevalent form of dementia in the elderly, involves critical changes such as reduced aerobic glycolysis in astrocytes and increased neuronal apoptosis, both of which are significant in the disease's pathology. In our study, astrocytes treated with amyloid β1-42 (Aβ) to simulate AD conditions exhibited upregulated expressions of small ubiquitin-like modifier (SUMO)-specific protease 1 (SENP1) and Pumilio RNA Binding Family Member 2 (PUM2), alongside decreased levels of Nuclear factor erythroid 2-related factor 2 (NRF2). SENP1 is notably the most upregulated SUMOylation enzyme in Aβ-exposed astrocytes.

Metallothionein rescues doxorubicin cardiomyopathy via mitigation of cuproptosis.

Life Sci

January 2025

Department of Cardiology, Cardiac Arrhythmia Center, Affiliated Hospital of Nantong University, Nantong 226001, China. Electronic address:

Doxorubicin (DOX), a chemotherapeutic agent utilized in the management of cancer, provokes cardiotoxicity although effective remedy is lacking. Given that DOX provokes oxidative stress and cell death in cardiomyocytes, this study evaluated the possible involvement of cuproptosis, a newly identified form of cell death, in DOX-instigated cardiac remodeling and contractile dysfunction, alongside the impact of the heavy metal scavenger metallothionein (MT) on DOX cardiomyopathy. Cardiac-specific MT transgenic and wild-type (WT) mice were treated with DOX (5 mg/kg/wk.

Department of Neurosurgery, The First Medical Centre of Chinese PLA General Hospital, Beijing, China; Neurosurgery Institute, Chinese PLA General Hospital, Beijing, PR China. Electronic address:

Background: Chronic cortisol overexposure plays a significant role in the development of neuropathological changes associated with neuropsychiatric and neurodegenerative disorders. The hippocampus, the primary target of cortisol, may exhibit characteristic regional responses due to its internal heterogeneity. This study explores structural and functional alterations of hippocampal subfields in Cushing's disease (CD), an endogenous model of chronic cortisol overexposure.

Exosomes have emerged as a powerful biomarker for early cancer diagnosis, however, accurately detecting cancer-derived exosomes in biofluids remains a crucial challenge. In this study, we present a novel label-free electrochemical biosensor utilizing titanium dioxide nanotube array films (TiONTAs) for the sensitive detection of exosomes in complex biological samples. This innovative biosensor takes advantage of the excellent electrochemical properties of TiONTAs and their specific interactions with the phosphate groups of exosomes.

Physiological wound healing process can restore the functional and structural integrity of skin, but is often delayed due to external disturbance. The development of methods for promoting the repair process of skin wounds represents a highly desired and challenging goal. Here, a flexible, self-powered, and multifunctional triboelectric nanogenerator (TENG) wound patch (e-patch) is presented for accelerating wound healing through the synergy of electrostimulation and photothermal effect.

A meniscus injury is a common cartilage disease of the knee joint. Despite the availability of various methods for the treatment of meniscal injuries, the poor regenerative capacity of the meniscus often necessitates resection, leading to the accelerated progression of osteoarthritis. Advances in tissue engineering have introduced meniscal tissue engineering as a potential treatment option.

[Clinical progress in stem cell therapy for end-stage liver disease].

Zhonghua Gan Zang Bing Za Zhi

December 2024

Department of Infectious Disease Medicine, Fifth Medical Center, PLA General Hospital, National Clinical Research Center of Infectious Diseases, Beijing100039, China.

End-stage liver disease includes liver failure and decompensated cirrhosis resulting from various etiologies and often leads to patient mortality due to complications and clinical symptoms such as severe jaundice, ascites, hepatic encephalopathy, coagulopathy, and hepatorenal syndrome. Liver transplantation is currently regarded as the most effective treatment, but its clinical application is limited by the shortage of donors, elevated expenses, and post-transplant rejection. Stem cells are a group of cells with multidirectional differentiation potential and self-renewal ability, which can improve the clinical indicator outcomes through mechanisms such as immunoregulation and promotion of tissue repair in patients with end-stage liver disease.

Background: Both novel anti-human epidermal growth factor receptor 2 (HER2) antibody-drug conjugates (ADCs) and pertuzumab and trastuzumab (HP) combined with chemotherapy(C) regimens are the choice of treatment for HER2 positive metastatic breast cancer (MBC) after tyrosine kinase inhibitors (TKIs). Our team's previous research has shown significant therapeutic effects of novel anti-HER2 ADCs in patients with TKIs treatment failure. Unfortunately, there is currently no data available to compare novel anti-HER2 ADCs with HP combined with chemotherapy regimens.

The positive feedback loop between SP1 and MAP2K2 significantly drives resistance to VEGFR inhibitors in clear cell renal cell carcinoma.

Int J Biol Sci

January 2025

Department of Urology, the Fourth Affiliated Hospital of School of Medicine, and International School of Medicine, International Institutes of Medicine, Zhejiang University, Yiwu, China.

Clear cell renal cell carcinoma (ccRCC) is one of the most common and aggressive malignancies of the urinary system. Despite being the first-line treatment for advanced ccRCC, vascular endothelial growth factor receptor inhibitors (VEGFRis) face significant limitations due to both initial and acquired resistance, which impede complete tumor eradication. Using a CRISPR/Cas9 library screening approach, was identified as a resistance-associated gene for three prevalent VEGFRis (Sunitinib, Axitinib, and Sorafenib).
