Background: The optimal definition and risk stratification approach to identify high-risk patients with prediabetes and stable coronary heart disease has not been well studied to date. The objective of the current study is to compare the prognostic value of different definitions of prediabetes, and to explore the role of "very-high-risk" (VHR) criteria according to the 2018 American Heart Association/American College of Cardiology cholesterol guideline in the risk stratification of patients with prediabetes and stable coronary heart disease.
Methods And Results: This prospective large-cohort study enrolled a total of 7930 patients with stable coronary heart disease.

Alzheimer's disease (AD) is a debilitating systemic disorder that has a detrimental impact on the overall well-being of individuals. Emerging research suggests that long non-coding RNAs play a role in neural development and function. Nevertheless, the precise relationship between lncRNAs and Alzheimer's disease remains uncertain.

This study presents the synthesis and characterization of novel cocrystal structures of theophylline (THE) with the amino acids gamma-aminobutyric acid (GABA) and l-arginine (ARG). Despite a large number of reports about THE cocrystals, no crystallographic parameters of cocrystals formed by THE and amino acids have been reported. THE is characterized by low solubility, while amino acids as cocrystal co-formers (CCFs) are increasingly recognized for their high solubility and safety.

This multicenter, single-arm, phase II clinical trial (NCT04034589) evaluated the efficacy and safety of pyrotinib combined with fulvestrant in patients with HR-positive/HER2-positive metastatic breast cancer who had experienced trastuzumab treatment failure. A total of 46 patients were enrolled, receiving pyrotinib orally once daily and fulvestrant intramuscularly on days 1 and 15 of cycle 1, followed by monthly doses on day 1. The primary endpoint was progression-free survival (PFS), while secondary endpoints included overall survival (OS), objective response rate (ORR), disease control rate (DCR), and safety.

The clinical application of most natural antimicrobial peptides (AMPs) is hindered by their lack of a synergistic combination of high antibacterial efficacy, low toxicity, and stability, necessitating frequent complex modifications that incur significant labor and economic costs. Therefore, it is imperative to optimize the antibacterial properties of AMPs using some simplified approach. In this study, we designed a library of β-hairpin AMPs with identical β-turn sequences (-D-Pro-Gly-) and varying repetition units (IR, FR, and WK).

Colorectal cancer (CRC) is one of the most common cancers worldwide. Early detection and removal of colorectal polyps during colonoscopy are crucial for preventing such cancers. With the development of artificial intelligence (AI) technology, it has become possible to detect and localize colorectal polyps in real time during colonoscopy using computer-aided diagnosis (CAD).

Traditionally viewed as congenital conditions with structural abnormalities, CVMs have been treated primarily through resection, embolization, and stereotactic radiosurgery. While these approaches offer some efficacy, they often pose risks to neurological integrity due to their invasive nature.

Background: The aim of this study is to develop a machine learning-based model to predict postoperative overall survival (OS) in patients with soft tissue sarcoma (STS) that demonstrates superior comprehensive performance.
Methods: This analysis leveraged data from the SEER database spanning 2010-2020, alongside a STS cohort from the National Cancer Center. Machine learning methods were applied for predictor selection by wrapper methods and the development of the predictive model.

Understanding the pathobiology of critical illness is essential for patients' prognosis. Sepsis is a life-threatening organ dysfunction caused by a dysregulated host response to infection. As part of the host response, procoagulant responses, one of the most primitive reactions in biology, start at the very beginning of diseases and can be monitored throughout the process.

Objective: The objective of this study is to examine the epidemiological characteristics of very elderly patients (aged over 80 years) with cancer admitted to the intensive care unit (ICU), and to elucidate the association between Acute Physiology Score III (APS-III) and 28-day mortality.
Method: A retrospective analysis was conducted using data extracted from the Medical Information Mart for Intensive Care IV (MIMIC-IV) database. Patients aged 80 years and above were assigned to three groups: non-cancer group, non-metastatic cancer group, and metastatic cancer group, based on their cancer diagnosis and its extent, Kaplan-Meier curves were constructed among these patient groups.
Tumor immunotherapy has significantly transformed the field of oncology over the past decade. An optimal tumor immunotherapy would ideally elicit robust innate and adaptive immune responses within tumor immune microenvironment (TIME). Unfortunately, immune system experiences functional decline with chronological age, a process termed "immunosenescence," which contributes to impaired immune responses against pathogens, suboptimal vaccination outcomes, and heightened vulnerability to various diseases, including cancer.

Background: CDK4/6 inhibitors (CDK4/6i) have shown promising results in the treatment of hormone receptor-positive (HR+) metastatic breast cancer (MBC) when combined with endocrine therapy (ET). It is crucial to evaluate the actual effectiveness and safety of CDK4/6i in clinical practice, as well as to analyze the factors that can predict their outcomes.
Methods: Patients with HR+ MBC who received CDK4/6i-based therapy between May 2016 and May 2023 at Hunan Cancer Hospital were evaluated for progression-free survival (PFS).

Background: The commonly used international patient-reported outcome scales for breast cancer were developed before the advent of multiple targeted therapies and immunotherapies, rendering them potentially insufficient for current clinical practices. Therefore, it is necessary to develop a specific patient-reported outcome scale tailored for breast cancer patients in China to optimize the management model for these patients.
Methods: A comprehensive literature search was performed in the PubMed, Embase, Wanfang, and CNKI databases to extract dimensions and items for a potential patient-reported outcome scale.
Microbes play a significant role in human tumor development and profoundly impact treatment efficacy, particularly in immunotherapy. The respiratory tract extensively interacts with the external environment and possesses a mucosal immune system. This prompts consideration of the relationship between respiratory microbiota and lung cancer.

Opioids are the most effective and widely used treatments for acute and chronic pain in patients with cancer. This review focuses on the impact of opioids and mu-opioid receptors (MORs) on the stages of oncologic metastasis. Studies have shown that opioids can facilitate tumor progression and are related to a poor prognosis in patients with cancer.

Sulfilimines, a privileged class of -S(iv)[double bond, length as m-dash]N- functional groups found in nature, have been exploited as valuable building blocks in organic synthesis and as pharmacophores in drug discovery, and have aroused significant interest in the chemical community. Nevertheless, strategies for late-stage introduction of sulfilimines into peptides and proteins have still met with limited success. Herein, we have developed a method of introducing biological sulfilimine fragments into peptides by an intermolecular sulfur atom transfer cascade reaction, utilizing hydroxylamine condensed with the acid moieties of peptides and varied diaryl disulfides.

Visible-light photooxidation sensitized by surface attachment of small colorless organics on semiconductor photocatalysts has emerged as an economical method for photocatalytic synthesis or degradation. In particular, heteroatom (X = N and Cl)-containing substrates could undergo either C-N coupling or dechlorination degradation sensitizing TiO, but the mechanism in conducting the competitive visible-light sensitized photooxidations is still vague. Herein, the visible-light photooxidation of colorless 4-chlorobenzene-1,2-diamine (-CAN) on TiO was revealed, contributing to selective C-N coupling rather than dechlorination.
