Extracellular proteins play pivotal roles in both intracellular signaling and intercellular communications in health and disease. While recent advancements in proximity labeling (PL) methods, such as peroxidase- and photocatalyst-based approaches, have facilitated the resolution of extracellular proteomes, their in vivo compatibility remains limited. Here, we report TyroID, an in vivo-compatible PL method for the unbiased mapping of extracellular proteins with high spatiotemporal resolution.

CKMT1 deficiency contributes to mitochondrial dysfunction and promotes intestinal epithelial cell apoptosis via reverse electron transfer-derived ROS in colitis.

Cell Death Dis

March 2025

National Clinical Research Center for Digestive Diseases, Department of Gastroenterology, Changhai Hospital, Naval Medical University, Shanghai, China.

Mitochondrial dysfunction contributes to the pathogenesis of ulcerative colitis (UC). As a mitochondrial isozyme of creatine kinases, which control energy metabolism, CKMT1 is thought to be a critical molecule in biological processes. However, the specific role of CKMT1 in intestinal inflammation remains largely unknown.

Motile cilia and flagella are evolutionarily conserved organelles, and their defects cause primary ciliary dyskinesia (PCD), a disorder characterized by systemic organ dysfunction. The nexin-dynein regulatory complex (N-DRC) is a crucial structural component of motile cilia and flagella, present across various species from Chlamydomonas to humans. Defects in N-DRC components lead to multiple PCD symptoms, including sinusitis and male infertility.

The cascade reaction of lipopolysaccharides (LPS), cell-free DNA (cfDNA), and reactive oxygen species (ROS), drives the development of inflammatory bowel disease (IBD). Herein, we construct polyethylenimide (PEI)-L/D-tartaric acid (L/D-TA) complexes templated mesoporous organosilica nanoparticles (MON) (PEI-L/D-TA@MON) by mimicking biosilicification under ambient conditions within seconds. The chiral nanomedicines include four functional moieties, wherein PEI electrostatically attracts cfDNA, tetrathulfide bonds reductively react with ROS, silanol groups adsorb LPS, and L/D-TA enables chiral recognition and inflammatory localization.

Background: The aspartate aminotransferase-to-platelet ratio index (APRI) is an effective non-invasive marker for chronic liver dysfunction. Given that heat stroke patients often suffer from poor prognosis due to multi-organ involvement, with liver injury and coagulation dysfunction being of particular concern, this study aims to investigate whether APRI can comprehensively reflect liver injury and coagulation dysfunction in heat stroke patients and explore its relationship with 28-day mortality.

Methods: This retrospective study analysed electronic medical records from patients treated at 57 grade A tertiary hospitals in China from May 2005 to May 2024.

Background: Cardiac cachexia is a condition characterised by unintentional weight loss and muscle wasting in patients with heart failure. However, there is debate about the prognostic value of cardiac cachexia in these patients.

Objectives: This meta-analysis aimed to evaluate the prognostic value of cardiac cachexia in patients who had heart failure.

Background: Long-term patterns of depressive symptoms among patients with heart failure, specifically those with a preserved ejection fraction (HFpEF), and their relationship with prognoses are not well studied.

Methods: This analysis included 609 participants from the TOPCAT (Treatment of Preserved Cardiac Function Heart Failure with an Aldosterone Antagonist) trial. Depressive symptoms were assessed using the Patient Health Questionnaire-9 (PHQ-9) at baseline and at 1-year, 2-year and 3-year intervals.

January 2025

Department of Laboratory Medicine, Wenzhou People's Hospital, The Third Affiliated Hospital of Shanghai University, The Wenzhou Third Clinical College of Wenzhou Medical University, Wenzhou, Zhejiang Province, China

Objective: This study aimed to ascertain serum bile acid levels and explore the diagnostic value of differential bile acids in patients with intrahepatic cholestasis of pregnancy (ICP).

Methods: 94 cases of ICP patients admitted to Wenzhou People's Hospital from Oct 2020 to July 2023 were selected as the ICP group, and 184 women who underwent normal prenatal examination in the same period as the healthy control group. The baseline data and 15 kinds of serum bile acid spectrum characteristics of the two groups of pregnant women were compared, and the risk factors of serum bile acid spectrum causing ICP were analyzed by univariate and multivariate logistic regression.
