Cell-free synthetic biology aims at the targeted replication, design, and modification of life processes in open systems by breaking free of constraints such as cell membrane barriers and living cell growth. The beginnings of this systematized technology, which took place in the last century, were used to explore the secrets of life. Currently, with its easy integration with other technologies or disciplines, cell-free synthetic biology is developing into a powerful and effective means of understanding, exploiting, and extending the structure and function of natural living systems.

Alveolar bone defects have always been an urgent problem in the oral cavity. For some patients with periodontal disease or undergoing orthodontic treatment or implant restoration, alveolar bone defects can greatly inconvenience clinical diagnosis and treatment. Periodontal ligament stem cells (PDLSCs) are considered a promising source for stem cell therapy due to their high osteogenic differentiation capability.

Background: Many plants, including those commonly found in the Fagaceae family, produce more flowers and ovules than mature fruits and seeds. In , an ovary contains 16-24 ovules, but only one develops into a seed. The other ovules abort or otherwise fail to fully develop, but the reason for this is unknown.

Background: Patients with end-stage renal disease (ESRD) have increased insulin resistance (IR). The triglyceride glucose (TyG) index is a marker of IR and is associated with the prognosis of patients on maintenance hemodialysis (MHD). The aim of this study was to examine the relationship between the TyG index and the risk of death in patients on MHD.

Background: Individuals with high autistic traits exhibit characteristics like those of individuals with autism, including impairments in sociability and communication skills. Whether individuals with high autistic traits exhibit less cooperation remains debated.
Methods: This study employed the prisoner's dilemma game (PDG) to measure cooperation in 56 dyads, including 27 with high-low (HL) autistic traits and 29 with low-low (LL) autistic traits, using functional near-infrared spectroscopy (fNIRS) hyperscanning technique.

Background: The increasing prevalence of primary hypertension among children and adolescents is a global health concern, with inadequate sleep duration identified as a significant risk factor. This study investigates the impact of weekday-weekend sleep duration gap (WWSDG) on hypertension among American adolescents.
Methods: Using data from the NHANES 2017-2020 cohort, we analyzed sleep patterns and hypertension prevalence among 430 adolescents.

Postoperative delirium (POD) represents a common neurological complication encountered predominantly among the elderly cohort undergoing surgical intervention for hip fractures. This phenomenon, particularly commonplace in geriatric populations with heightened preoperative risk profiles, pronounced comorbidities, and later stages of lifespan, poses complex clinical challenges. The impact of perioperative pharmacological interventions and anesthetic strategies on POD's emergence cannot be understated, as it may profoundly affect the length of hospital stays, rehabilitation milestones, and the overall mortality hazard.

Purpose: Sepsis-associated liver injury (SALI) leads to increased mortality in sepsis patients, yet no specialized tools exist for early risk assessment. This study aimed to develop and validate a risk prediction model for early identification of SALI before patients meet full diagnostic criteria.
Patients And Methods: This retrospective study analyzed 415 sepsis patients admitted to ICU from January 2019 to January 2022.

Background: Transarterial therapy (TAT), bevacizumab (Bev), and immune checkpoint inhibitors (ICIs) have individually exhibited efficacy in treating advanced-stage hepatocellular carcinoma (HCC). This study aimed to assess the efficacy and safety of the combination of these three treatments as a neoadjuvant modality in patients with locally advanced HCC.
Methods: The primary endpoint is overall survival (OS).

Immune checkpoint inhibitors (ICIs) have revolutionized the treatment of systemic cancer therapy. During disinhibiting the antitumor responses of immune system, ICIs may also cause unique immune-related adverse events (irAEs) which could affect any organ. Here, we report a rare case of sintilimab-induced ureteritis/cystitis in a 55-year-old male undergoing neoadjuvant chemo-immunotherapy for gastric cancer.

Introduction: Non-small cell lung cancer (NSCLC) constitutes approximately 80-85% of cancer-related fatalities globally, and direct and indirect comparisons of various therapies for NSCLC are lacking. In this study, we aimed to compare the efficacy and safety of immune checkpoint inhibitors (ICIs) in patients with epidermal growth factor receptor (EGFR)-mutated NSCLC.
Methods: The electronic databases were systematically searched from inception until March 18, 2024.

Background: Squalene epoxidase (SQLE) is a key enzyme in cholesterol biosynthesis and has been shown to negatively affect tumor immunity and is associated with poor outcomes of immunotherapy in various cancers. While most research in this area has focused on the impact of cholesterol on immune functions, the influence of SQLE-mediated squalene metabolism within the tumor immune microenvironment (TIME) remains unexplored.
Methods: We established an immune-competent mouse model (C57BL/6) bearing mouse pancreatic cancer xenografts (KPC cells) with or without stable SQLE-knockdown (SQLE-KD) to evaluate the impact of SQLE-mediated metabolism on pancreatic cancer growth and immune functions.

Background: Post-stroke depression (PSD) is the most prevalent neuropsychiatric complication following a stroke. The inflammatory theory suggests that PSD may be associated with an overactive inflammatory response. However, research findings regarding inflammation-related indicators in PSD remain inconsistent and elusive.

Introduction: Vaccines against intracellular pathogens like require the induction of effective cell-mediated immunity. Adjuvants primarily enhance antigen-induced adaptive immunity by promoting the activation of antigen-presenting cells (APCs).This study is to develop an adjuvant targeted to dendritic cells (DCs), one of the main APCs, so as to assist in inducing a long-term cellular immune response to protein antigens.

Pulmonary large cell carcinoma (LCC) is a rare and aggressive subtype of non-small cell lung cancer (NSCLC) with poor prognosis. Surgical resection remains the cornerstone of treatment for resectable LCC; however, its efficacy is limited in advanced stages, necessitating adjuvant therapies to reduce postoperative recurrence risk. Recent advances in immunotherapy have shown promising survival benefits.

Background: Clinical studies have demonstrated the high efficacy of using chimeric antigen receptor (CAR)-T cells targeting B-cell maturation antigen (BCMA) and orphan G protein-coupled receptor, class C group 5 member D (GPRC5D) to treat relapsed or refractory multiple myeloma (RRMM). In this study, we compared the efficacy and safety of BCMA CAR-T-cell therapy (BCMA CAR-T) and GPRC5D CAR T-cell therapy (GPRC5D CAR-T) in patients with RRMM.
Methods: We retrieved and included eligible clinical trials of BCMA or GPRC5D CAR-T for RRMM patients.
Background: The addition of durvalumab or pembrolizumab to gemcitabine and cisplatin (GP) has been approved to statistically improve survival outcomes in patients with advanced biliary tract cancer. However, since the survival time was only prolonged by about two months, doubts have been raised. In this analysis, we aimed to evaluate the efficacy of combining durvalumab or pembrolizumab with GP chemotherapy.

Objectives: Traditional methods of treating allergies primarily revolve around avoiding allergens and promptly using rescue medications when allergic symptoms occur. However, this approach is known for its inefficiency and limited success in achieving long-term relief. Our aim was to conduct a comprehensive analysis of previously published randomized controlled trials (RCTs) that explore the effectiveness and safety of epicutaneous immunotherapy (EPIT) as a means to manage food allergies in children.
