Thermophilic proteins, mesophiles proteins and psychrophilic proteins have wide industrial applications, as enzymes with different optimal temperatures are often needed for different purposes. Convenient methods are needed to determine the optimal temperatures for proteins; however, laboratory methods for this purpose are time-consuming and laborious, and existing machine learning methods can only perform binary classification of thermophilic and non-thermophilic proteins, or psychrophilic and non-psychrophilic proteins. Here, we developed a deep learning model, PSTP-BERT, based on protein sequences that can directly perform Three classes identification of thermophilic, mesophilic, and psychrophilic proteins.

Tough and Functional Hydrogel Coating by Electrostatic Spraying.

Small

December 2024

Center for X-Mechanics, Department of Engineering Mechanics, Zhejiang University, Hangzhou, 310027, China.

Hydrogel coatings impart superior surface properties to materials, but their application on large and complicated substrates is hindered by two challenges: limited wetting conditions and intricate curing processes. To overcome the challenges, lyophilized adhesive hydrogel powders (LAHPs) are developed, which consist of poly(acrylic acid-co-3-(trimethoxysilyl)propyl methacrylate) crosslinked with chitosan. These powders are electrostatic sprayed onto substrates to address wetting issues and rehydrated to form bulk hydrogel coatings to circumvent curing challenges.

Neural networks for predicting etiological diagnosis of uveitis.

Eye (Lond)

December 2024

Department of Internal Medicine, Hôpital Universitaire de la Croix-Rousse, Hospices Civils de Lyon, University Claude Bernard-Lyon 1, Lyon, France.

Background/objectives: The large number and heterogeneity of causes of uveitis make the etiological diagnosis a complex task. The clinician must consider all the information concerning the ophthalmological and extra-ophthalmological features of the patient. Diagnostic machine learning algorithms have been developed and provide a correct diagnosis in one-half to three-quarters of cases.

Article Synopsis
  • - The study explores the potential of chlorquinaldol (CQ), an antimicrobial agent, as a specific inhibitor of the NLRP3 inflammasome, which is linked to various inflammatory diseases, highlighting its promise for drug repurposing.
  • - CQ effectively suppresses NLRP3 inflammasome activation in both mouse and human macrophages primarily by blocking the interaction between NLRP3 and ASC, while having minimal impact on other inflammasomes like NLRC4 and AIM2.
  • - In vivo tests showed that CQ significantly improves conditions in mouse models of LPS-induced peritonitis, DSS-induced colitis, and MSU-induced gouty arthritis, suggesting its therapeutic potential for treating NLRP3-related

The evaluation of the effectiveness of traditional Chinese medicine(TCM) has long been a "bottleneck" issue that restricts the healthy development and internationalization of TCM. This article systematically analyzes the differences in diagnostic and treatment principles, material composition, new drug development pathways, efficacy cognition, and evaluation methodology between Chinese and Western medicine from the underlying logic of medical perspective. On this basis, it integrates the evidence elements of clinical experience, experimental research, and clinical trials, and for the first time proposes and establishes a new strategy and methodology for the evaluation of the effectiveness of TCM: the integrated evidence chain-based effectiveness evaluation of traditional Chinese medicine(iEC-Eff), forming an integrated evaluation and confirmation model of "clinical experience-experimental research-clinical trial" for the effectiveness of TCM.
