The purpose of this inter-laboratory study was to test the repeatability and reproducibility of an in vitro method aimed at analyzing the physicochemical properties under physiological conditions of β-glucans from foods. After evaluating β-glucans molar mass and quantification methods using five β-glucan controls, four laboratories ran six oat-based products through in vitro digestion, measured β-glucans solubility and viscosity and molar mass of solubilized β-glucans. The determination of the molar mass of β-glucan controls, their viscosity in solution and β-glucans content in food samples exhibited relative standard reproducibility of 20.

Background: Members of the Alternaria genus produce various toxins whose occurrence in agricultural commodities is a major concern for humans and the environment. The present study developed a simple and efficient matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) method for the rapid detection of Alternaria toxins.
Results: A new method for the detection of alternariol (AOH), alternariol monomethyl ether (AME) and tentoxin (TEN) by MALDI-TOF MS was developed.

The physiological cholesterol-lowering benefits of β-glucan have been well documented, however, whether modulation of gut microbiota by β-glucan is associated with these physiological effects remains unknown. The objectives of this study were therefore to determine the impact of β-glucan on the composition of gut microbiota in mildly hypercholesterolemic individuals and to identify if the altered microbiota are associated with bioactivity of β-glucan in improving risk factors of cardiovascular disease (CVD). Using a randomized, controlled crossover study design, individuals received for 5-week either a treatment breakfast containing 3 g high molecular weight (HMW), 3 g low molecular weight (LMW), 5 g LMW barley β-glucan, or wheat and rice.

The extreme climate of the Canadian Prairies poses a major challenge to improve yield. Although it is possible to breed for yield per se, focusing on yield-related traits could be advantageous because of their simpler genetic architecture. The Canadian flax core collection of 390 accessions was genotyped with 464 simple sequence repeat markers, and phenotypic data for nine agronomic traits including yield, bolls per area, 1,000 seed weight, seeds per boll, start of flowering, end of flowering, plant height, plant branching, and lodging collected from up to eight environments was used for association mapping.

Twenty-four isolates of Penicillium expansum Link from British Columbia (Canada) apples were cultured in yeast-extract sucrose (YES) at 25°C for 28 days to investigate production of patulin and citrinin. These isolates proved to be potent producers of citrinin, patulin, or in most cases, both mycotoxins. In every isolate, citrinin, patulin, or both compounds were produced at levels as high as 565 µg/mL (mean 269 µg/mL) and 100 µg/mL (mean 31 µg/mL), respectively.

Twenty-six samples of commercial ice-wine made from late-harvested grapes were assayed for the mycotoxins ochratoxin A and tenuazonic acid. Canadian wines originated in British Columbia (18) and Ontario (8). For comparison two German wines from Hesse were also studied.

Wheat streak mosaic virus (WSMV) infection reduces seed yield and quality in wheat. These losses can be alleviated significantly by exploiting genetic host plant resistance. A new source of temperature-sensitive resistance to WSMV, KS03HW12, and its parental lines (KS97HW29/ KS97HW131//KS96HW100-5) were evaluated in both greenhouse and field conditions.

Wheat yields often are limited by infection by Wheat streak mosaic virus (WSMV). Host plant resistance to WSMV can reduce losses. This study was conducted to characterize a new source of temperature-sensitive resistance found in CO960293 wheat.

Wheat with yellow head disease (YHD) (yellow heads and mosaic leaf symptoms) has been observed in Kansas since 1997. A pathogen was transmitted from the infected wheat to maize by vascular puncture inoculation and to Nicotiana benthamiana by rub inoculation. The original infected wheat and infected maize and N.

To assess production of the mycotoxin moniliformin, inoculation trials with four barley cultivars and local isolates of Fusarium avenaceum were performed at two different experimental farms in southern Manitoba, Canada, during 1997-99. In the 1997 study, moniliformin was detected in 11 of 16 barley rows at levels between 0.19 and 1.

Fusarium graminearum, Fusarium culmorum, and Fusarium avenaceum, isolated from Fusarium-damaged wheat harvested in western Canada, were cultured and evaluated for mycotoxin production. Extracts of the culture media were assayed for trichothecenes by gas chromatography-mass spectrometry and for moniliformin by liquid chromatography. Deoxynivalenol (DON) was found in 28 of 42 isolates of F.

Sorghum (Sorghum bicolor) was infected by a mechanically transmissible, flexuous, rod-shaped virus. Antiserum made against the purified virus reacted specifically in enzyme-linked immunosorbent assay to the virus and to the potexvirus foxtail mosaic virus (FoMV), indicating that the sorghum virus was an isolate of FoMV. Comparison of the sorghum isolate (H93) to FoMV PV 139 showed that H93 differed biologically by causing severe symptoms in sorghum, not readily infecting certain barley lines, and causing only faint symptoms in barley.

Feed samples from 94 cases involving fungal contamination and suspected mycotoxicosis of farm animals in western Canada were examined during 1982-1994 to assess the incidence of mycotoxins. Samples were analyzed for aflatoxins, ochratoxin A, citrinin, sterigmatocystin, and the fungal estrogen zearalenone. Samples infected with Fusarium fungi were additionally assayed for nivalenol, deoxynivalenol, fusarenone-x, 15-acetyl-deoxynivalenol, diacetoxyscirpenol, HT-2 toxin, and T-2 toxin.

Polyclonal antibodies against the mycotoxin citrinin were raised in rabbits after immunization with citrinin conjugated to keyhole limpet hemocyanin. The antibodies were used in a competitive indirect enzyme immunoassay (EIA) with citrinin coupled to glucose oxidase as the solid-phase antigen for coating microtiter plates. Detection limits of this indirect EIA for citrinin ranged from 0.
