Article Synopsis
  • The study investigates frontotemporal dementia linked to granulin (GRN) mutations, focusing on brain connectivity patterns using graph theory measures in different groups: symptomatic carriers, presymptomatic carriers, and non-carriers.
  • It finds that symptomatic carriers show significant global connectivity issues, while presymptomatic carriers display early signs of interhemispheric disconnection between parietal regions, despite not showing overall connectivity changes.
  • The findings imply that GRN-related frontotemporal dementia develops as a disconnection syndrome, starting with parietal areas before advancing to frontotemporal regions as symptoms become apparent.

Intratumoral heterogeneity of MYC drives medulloblastoma metastasis and angiogenesis.

Neuro Oncol

September 2022

German Cancer Consortium (DKTK), Partner Site Essen/Düsseldorf , Düsseldorf, Germany.

Background: Intratumoral heterogeneity is crucially involved in metastasis, resistance to therapy, and cancer relapse. Amplifications of the proto-oncogene MYC display notable heterogeneity at the single-cell level and are associated with a particularly dismal prognosis in high-risk medulloblastomas (MBs). The aim of this study was to establish the relevance of interclonal cross-talk between MYC-driven and non-MYC-driven MB cells.

Clinical outcomes of a cohort of 271 patients with lung metastases from differentiated thyroid carcinoma.

Clin Endocrinol (Oxf)

December 2022

Endocrinology Department, Instituto Português de Oncologia de Lisboa Francisco Gentil, Lisboa, Portugal.

Article Synopsis
  • Lung is the primary site for distant metastases in differentiated thyroid carcinoma (DTC), particularly from papillary thyroid carcinoma (PTC), which accounts for 83.4% of cases.
  • A study analyzed 271 patients with lung metastases (LM), highlighting that younger patients (under 55) and those with a micronodular pattern had better outcomes.
  • While most patients received radioactive iodine treatment (RAIT), complete remission was rare, and survival rates did not significantly differ between treated and untreated patients, although RAIT appeared to benefit those with iodine-avid lesions.

[Analysis of the Cochrane Review: Antiplatelet Agents for Preventing Pre-Eclampsia and Its Complications. Cochrane Database Syst Rev. 2019;10:CD004659.].

Acta Med Port

December 2021

Centro de Estudos de Medicina Baseada na Evidência. Faculdade de Medicina. Universidade de Lisboa. Lisboa. Cochrane Portugal. Lisboa. Portugal.

Pre-eclampsia is associated with deficient intravascular production of prostacyclin, a vasodilator, and excessive production of thromboxane, a vasoconstrictor and stimulant of platelet aggregation. These observations led to the hypotheses that antiplatelet agents, low-dose aspirin in particular, might prevent or delay development of pre-eclampsia. This Cochrane review aimed to assess the effectiveness and safety of antiplatelet agents, such as aspirin and dipyridamole, when given to women at risk of developing preeclampsia.

Eosinophilic fasciitis (EF) is a rare subacute fibrosing disorder of unknown aetiology, characterised by thickening of the muscular fascia and subcutaneous tissue, leading to swelling of limbs and trunk and sparing fingers and toes. Eosinophilic infiltration and degranulation may prompt tissue damage and consequent fibrosis due to the accumulation of collagen and extracellular matrix proteins. MRI is the best imaging modality for diagnosis, depicting fascial thickening and enhancement.
