ICP27, which is involved in post-transcriptional regulation, is the only IE gene product conserved in all herpesviruses. We show here that the ICP27 transcript of the oncogenic Marek's disease virus shares the same polyadenylation signal as the bicistronic glycoprotein K-ICP27 transcript and is regulated by alternative promoter usage, with transcription from its own promoter (pICP27) or that of gK (pgK).

Recombinant equine LH/chorionic gonadotropin (eLH/CG) was expressed in the baculovirus-Sf9 insect cell system either as a single-chain with the C-terminus of the beta-subunit fused to the N-terminus of the alpha-subunit or as non-covalently linked heterodimers with or without a polyhistidine tag at various locations. All these non-covalently linked eLH/CG variants were secreted as stable heterodimers in the medium of infected Sf9 cells. To assess the influence of the presence and the position of polyhistidine tag on LH bioactivity, we expressed four non-covalently linked tagged heterodimeric eLH/CG variants that were secreted in threefold higher quantities than the single chain.

We collected paraffin-embedded myelocytomatoses induced by subgroup J avian leukosis virus (ALV-J) in French poultry from 1992 to 2000. We used nested PCR to obtain the U3 LTR and the E element sequences that encompass putative binding sites for transcription factors. We observed minor mutations in the U3 sequences that rarely affected transcription factor binding sites, thus preserving the transcriptional potential of the U3 LTR.

The pharyngeal (Ph) and palatine (Pa) tonsils, although located in different regions of the upper aero-digestive tract (UADT), are thought to protect the respiratory tract similarly against infections by inducing and disseminating T and surface IgA(+) (sIgA(+)) B cells. We investigated the factors controlling the migratory properties of T and sIgA(+) B lymphocytes in the UADT of pigs by comparing the expression of vascular addressins, homing receptors and chemokine transcripts in Ph/Pa tonsils, Peyer's patches (PP) and their draining lymph nodes (LN). The vascular addressin PNAd was detected on high endothelial venules in both tonsils, whereas mucosal addressin cell adhesion molecule-1, otherwise present in PP and mesenteric LN, was not detected.

Rabbit hemorrhagic disease virus (RHDV) and European brown hare syndrome virus (EBHSV) are caliciviruses that produce severe symptoms and are lethal to rabbits and hares. The folding of the capsid protein was studied by determination of the antigenic pattern of chimeric capsid proteins, composed of regions from RHDV and EBHSV capsid proteins. The anti-RHDV monoclonal antibody (MAb) E3, which is known to bind an external conformational epitope, recognized the RHDV C-terminal region.

Cryopreservation of cells appears to be a potential method of comparing chemotaxis of lymphocytes collected from different anatomical sites at one time in a single assay. Migration of cryopreserved lymphocytes from swine in the absence (spontaneous migration) or presence of N-formyl-methionyl-leucyl-phenylalanine (fMLP, induced migration) was compared to that of fresh lymphocytes, originating from inguinal (ILN) and mesenteric (MLN) lymph nodes, respectively, using a 48-well microchemotaxis chamber. Cryopreservation did not affect the optimal concentration of fMLP for maximal induced migration and did not impair the chemoattractant activity of fMLP as shown by checkerboard assay.

Recombinant porcine interferon gamma (IFN gamma) at neutral pH is characterized by a tryptophan (Trp) fluorescence maximum around 343 nm and a rigid conformation, evidenced from tryptophan polarization values. Guanidine HCl shifts the protein emission spectra further to the red and decreases the fluorescence polarization values, indicating denatured IFN gamma in these solutions. In acidic solutions (3 < pH < 4), the emission spectra show a blue shift and lower tryptophan polarization.
