Background: Gambian sleeping sickness or HAT (human African trypanosomiasis) is a neglected tropical disease caused by Trypanosoma brucei gambiense transmitted by riverine species of tsetse. A global programme aims to eliminate the disease as a public health problem by 2020 and stop transmission by 2030. In the South of Chad, the Mandoul area is a persistent focus of Gambian sleeping sickness where around 100 HAT cases were still diagnosed and treated annually until 2013.

Background: Trypanosoma (Duttonella) vivax is a major pathogen of livestock in Africa and South America (SA), and genetic studies limited to small sampling suggest greater diversity in East Africa (EA) compared to both West Africa (WA) and SA.

Profiling of wild and laboratory tsetse populations using 16S rRNA gene amplicon sequencing allowed us to examine whether the "Wigglesworthia-Sodalis-Wolbachia dogma" operates across species and populations. The most abundant taxa, in wild and laboratory populations, were Wigglesworthia (the primary endosymbiont), Sodalis and Wolbachia as previously characterized. The species richness of the microbiota was greater in wild than laboratory populations.

Trypanosomatidae are a dangerous family of Euglenobionta parasites that threaten the health and economy of millions of people around the world. More precisely describing the population biology and reproductive mode of such pests is not only a matter of pure science, but can also be useful for understanding parasite adaptation, as well as how parasitism, specialization (parasite specificity), and complex life cycles evolve over time. Studying this parasite's reproductive strategies and population structure can also contribute key information to the understanding of the epidemiology of associated diseases; it can also provide clues for elaborating control programs and predicting the probability of success for control campaigns (such as vaccines and drug therapies), along with emergence or re-emergence risks.

To better understand the relative importance of the multi-purpose functions of small ruminants for their urban owners and related breeding practices including selection criteria, we undertook a comparative analysis across the West African cities of Kano (Nigeria), Bobo Dioulasso (Burkina Faso) and Sikasso (Mali). Semi-structured questionnaires were used to collect the required information from 301 sheep farmers (100, 102 and 99 in Kano, Bobo Dioulasso and Sikasso) and 306 goat farmers (100, 99 and 107 in Kano, Bobo Dioulasso and Sikasso). Sheep and goats were kept for a variety of reasons including income generation, insurance (sale for cash to meet unexpected expenditures) and economic security (sale for cash to support foreseeable expenses), social/religious functions and prestige in ownership.

Expected values for observed heterozygosity, genetic diversity, and inbreeding of individuals relative to inbreeding of the population (F(IS)) are derived in the case of one locus displaying homoplasy with K possible allelic states (KAM model) in a clonal diploid population. Heterozygosity (H(O)) and genetic diversity (H(S)) are substantially affected by homoplasy as long as the number of alleles K ⩽ 10, while F(IS) remains weakly affected in any case. Simulations suggest that in big populations, or in case of maximum homoplasy (K = 2), expected values can appear far from the observed ones because equilibrium takes too many generations to be reached at homoplasic markers in clonally propagating populations.

Background: Pathogens and their vectors are organisms whose ecology is often only accessible through population genetics tools based on spatio-temporal variability of molecular markers. However, molecular tools may present technical difficulties due to the masking of some alleles (allelic dropouts and/or null alleles), which tends to bias the estimation of heterozygosity and thus the inferences concerning the breeding system of the organism under study. This is especially critical in clonal organisms in which deviation from panmixia, as measured by Wright's FIS, can, in principle, be used to infer both the extent of clonality and structure in a given population.

Tsetse flies are the cyclical vectors of African animal trypanosomosis (AAT) and human African trypanosomosis (HAT). In March 2010, the Government of Ghana initiated a large scale integrated tsetse eradication campaign in the Upper West Region (UWR) (≈18,000 km(2)) under the umbrella of the Pan-African Tsetse and Trypanosomosis Eradication Campaign (PATTEC).
