Occup Environ Med

February 2024

Institut National de la Santé et de la Recherche Médicale (INSERM), Unité mixte de Recherche (UMR) 1086 ANTICIPE, Caen, France.

Article Synopsis
  • Ovarian cancer is rare but has a poor prognosis, with hormones and reproductive factors potentially linking it to endocrine disruptors in agricultural settings.
  • In a study involving 59,391 female farmers, the impact of specific agricultural exposures during different life stages on ovarian cancer risk was analyzed, revealing both increased and decreased risks associated with various farming activities.
  • Key findings indicated that involvement with pigs, fruit-growing, and potato seed treatment elevated ovarian cancer risk, particularly during puberty, while grain cereal farming showed a reduced risk; exposure to triazine herbicides was not linked to increased ovarian cancer.

Variants which disrupt splicing are a frequent cause of rare disease that have been under-ascertained clinically. Accurate and efficient methods to predict a variant's impact on splicing are needed to interpret the growing number of variants of unknown significance (VUS) identified by exome and genome sequencing. Here, we present the results of the CAGI6 Splicing VUS challenge, which invited predictions of the splicing impact of 56 variants ascertained clinically and functionally validated to determine splicing impact.

February 2024

PSL Research University, institut Curie, service de génétique, pôle médecine diagnostique & théranostique, 26, rue d'Ulm, 75248 Paris cédex 05, France. Electronic address:

Article Synopsis
  • Systematic screening for pancreatic cancer is important for high-risk individuals due to the generally poor outcomes of late-stage diagnoses, which are often advanced and inoperable.
  • Individual risk is assessed through genetic information and family history, with genetic testing recommended for those suspected of having a genetic predisposition.
  • While current screening involves annual MRI and endoscopic ultrasound, challenges remain due to the methods' lack of sensitivity and specificity, leading to potential missed diagnoses and unnecessary surgeries, highlighting the need for improved imaging techniques and biological research.

Article Synopsis
  • There is currently no reliable biomarker for predicting the effectiveness of EGFR inhibitors in head and neck squamous cell carcinoma (HNSCC), leading researchers to investigate afatinib, a promising treatment.
  • In a trial with 61 patients, afatinib showed a high metabolic response, with responders demonstrating specific biomarker expressions (higher pERK1/2 and lower pHER4 and pRB1 levels).
  • Further analysis indicated that responders had more tumor-infiltrating B cells, while non-responders had an overrepresentation of NF-kappa B signaling in their gene expression, suggesting potential new biomarkers for afatinib efficacy that require more research.*

With more than 60,000 new cases of breast cancer in mainland France in 2023 and 8% of all cancer deaths, breast cancer is the leading cancer in women in terms of incidence and mortality. While the number of new cases has almost doubled in 30 years, the percentage of patients at all stages alive at 5 years (87%) and 10 years (76%) testifies to the major progress made in terms of screening, characterisation and treatment. However, this progress, rapid as it is, needs to be evaluated and integrated into an overall strategy, taking into account the characteristics of the disease (stage and biology), as well as those of the patients being treated.
