Article Synopsis
  • Primary human trophoblast stem cells (TSCs) and those derived from human pluripotent stem cells (hPSCs) can be modeled in the lab, but how hPSCs differentiate into TSCs is not well understood.
  • This study shows that a specific primed pluripotent state can produce TSCs by activating certain pathways (like EGF and WNT) and inhibiting others (like TGFβ and ROCK), all without adding BMP4, referred to as the TS condition.
  • The researchers found that the TSCs generated under TS conditions can proliferate extensively and closely resemble first-trimester placental cells, suggesting that primed hPSCs can differentiate into TSCs through various

Background: The accurate prediction of genomic breeding values is central to genomic selection in both plant and animal breeding studies. Genomic prediction involves the use of thousands of molecular markers spanning the entire genome and therefore requires methods able to efficiently handle high dimensional data. Not surprisingly, machine learning methods are becoming widely advocated for and used in genomic prediction studies.

Sequential neural dynamics encoded by time cells play a crucial role in hippocampal function. However, the role of hippocampal sequential neural dynamics in associative learning is an open question. We used two-photon Ca imaging of dorsal CA1 (dCA1) neurons in the stratum pyramidale (SP) in head-fixed mice performing a go-no go associative learning task to investigate how odor valence is temporally encoded in this area of the brain.

Sleep is essential for physical and mental health. Polysomnography (PSG) procedures are labour-intensive and time-consuming, making diagnosing sleep disorders difficult. Automatic sleep staging using Machine Learning (ML) - based methods has been studied extensively, but frequently provides noisier predictions incompatible with typical manually annotated hypnograms.

Center for Mathematics, Computing and Cognition (CMCC), Federal University of ABC (UFABC), São Bernardo do Campo 09606-045, Brazil

Estimating durations between hundreds of milliseconds and seconds is essential for several daily tasks. Explicit timing tasks, which require participants to estimate durations to make a comparison (time for perception) or to reproduce them (time for action), are often used to investigate psychological and neural timing mechanisms. Recent studies have proposed that mechanisms may depend on specific task requirements.

: Given the wide spectrum of clinical and laboratory manifestations of the coronavirus disease 2019 (COVID-19), it is imperative to identify potential contributing factors to patients' outcomes. However, a limited number of studies have assessed how the different waves affected the progression of the disease, more so in Portugal. Therefore, our main purpose was to study the clinical and laboratory patterns of COVID-19 in an unvaccinated population admitted to the intensive care unit, identifying characteristics associated with death, in each of the first three waves of the pandemic.

Electronic address:

Background: Men who have sex with men (MSM) in the Asia-Pacific region have a disproportionately high burden of HIV infection compared with the general population. Although pre-exposure prophylaxis (PrEP) for HIV is highly effective at preventing new HIV infections, the cost-effectiveness of PrEP for MSM in different countries in the Asia-Pacific region with varying PrEP coverage and HIV testing frequencies remains unstudied. We aimed to analyse the economic and health benefits of long-acting injectable cabotegravir (CAB-LA) compared with oral PrEP in high-income countries and low-income and middle-income countries within the Asia-Pacific region.

Propagation of random excitations in the polar representation of strongly nonlinear four-wave systems of turbulence.

Phys Rev E

December 2023

Research Center for Mathematics, Beijing Normal University at Zhuhai 519087, China and Department of Mathematical Sciences, United International College (BNU-HKBU), Zhuhai 519087, China.

The present work is concerned with the uncertainty propagation of the wave turbulent system. In particular, we study the temporal development and long-term behavior of the probability with respect to the amplitude and phase of complex-valued waves constituting the generic four-wave system of turbulence. Our approach to approximating the target distribution function is via the three steps: (i) to grasp the physical process described by the true turbulence model as random process, (ii) to determine the stochastic differential equation whose solution exhibits statistically similar behavior with the underlying turbulent signal, and (iii) to solve the corresponding Kolmogorov forward equation.

scQA: A dual-perspective cell type identification model for single cell transcriptome data.

Comput Struct Biotechnol J

December 2024

Research Center for Mathematics and Interdisciplinary Sciences, Shandong University, Qingdao 266237, China.

Single-cell RNA sequencing technologies have been pivotal in advancing the development of algorithms for clustering heterogeneous cell populations. Existing methods for utilizing scRNA-seq data to identify cell types tend to neglect the beneficial impact of dropout events and perform clustering focusing solely on quantitative perspective. Here, we introduce a novel method named scQA, notable for its ability to concurrently identify cell types and cell type-specific key genes from both qualitative and quantitative perspectives.

Rice is a significant staple food in the basic diet of the global population that is considered to have a high glycaemic index. The study of the physical and chemical parameters in rice that are related to the starch digestion process, which allows us to quickly predict the glycaemic index of varieties, is a major challenge, particularly in the classification and selection process. In this context, and with the goal of establishing a relationship between physicochemical properties and starch digestibility rates, thus shedding light on the connections between quality indicators and their glycaemic impact, we evaluated various commercial rice types based on their basic chemical composition, physicochemical properties, cooking parameters, and the correlations with digestibility rates.

On committor functions in milestoning.

J Chem Phys

December 2023

Qingdao Institute for Theoretical and Computational Sciences, Institute of Frontier and Interdisciplinary Science, Shandong University, Qingdao, Shandong 266237, People's Republic of China.

As an optimal one-dimensional reaction coordinate, the committor function not only describes the probability of a trajectory initiated at a phase space point first reaching the product state before reaching the reactant state but also preserves the kinetics when utilized to run a reduced dynamics model. However, calculating the committor function in high-dimensional systems poses significant challenges. In this paper, within the framework of milestoning, exact expressions for committor functions at two levels of coarse graining are given, including committor functions of phase space point to point (CFPP) and milestone to milestone (CFMM).

This article investigates the emergence of phase synchronization in a network of randomly connected neurons by chemical synapses. The study uses the classic Hodgkin-Huxley model to simulate the neuronal dynamics under the action of a train of Poissonian spikes. In such a scenario, we observed the emergence of irregular spikes for a specific range of conductances and also that the phase synchronization of the neurons is reached when the external current is strong enough to induce spiking activity but without overcoming the coupling current.

Monitoring disease response after intensive chemotherapy for acute myeloid leukemia (AML) currently requires invasive bone marrow biopsies, imposing a significant burden on patients. In contrast, cell-free tumor DNA (ctDNA) in peripheral blood, carrying tumor-specific mutations, offers a less-invasive assessment of residual disease. However, the relationship between ctDNA levels and bone marrow blast kinetics remains unclear.
