  • - Comprehensive genomic profiling using circulating tumor DNA (ctDNA) has potential in capturing tumor diversity and improving therapy choices, but it's not fully utilized in clinical settings, especially for advanced solid tumors.
  • - The GOZILA study found that ctDNA profiling led to a 24% match rate for targeted therapies, significantly improving patient outcomes, with those receiving matched treatments experiencing better overall survival rates compared to those who were unmatched.
  • - Key ctDNA characteristics, like biomarker clonality and plasma copy number, serve as indicators of treatment effectiveness, suggesting that ctDNA can enhance precision in oncology and should be more widely used to optimize patient survival in advanced solid tumors.

Radiation-associated cardiovascular disease (RACD), a complex disease characterized with pericarditis, myocardial damage, valvular heart diseases, heart failure, vasculopathy and ischemic heart disease, has a generally poor prognosis. While RACD may be acute, it often manifests in the late years or even decades following radiation exposure to the chest. With an increasing number of cancer survivors, RACD is likely to become an important issue in cardio-oncology.

Article Synopsis
  • The study aims to distinguish between struma ovarii (SO) and mucinous carcinomas (MC) based on various imaging techniques, including CT and MRI.
  • Results show that SO displays more low intensity on T2-weighted images and hyperattenuation on non-contrast CT compared to MC, indicating significant differences in imaging characteristics.
  • The findings suggest that SO can be specifically identified through a high-risk time-intensity curve pattern and the presence of low signal intensity on T2-weighted images, without diffusion restriction, contributing to more accurate diagnoses.

Lynch syndrome (LS) results from pathogenic variants in mismatch repair genes and is the most common hereditary cancer syndrome. Some guidelines or studies recommend restricting screening according to endometrial cancer (EC) using endometrial biopsy. The pooled sensitivity and specificity of endometrial cytology for detecting endometrial atypical hyperplasia or cancer have been reported to be as high as the pooled sensitivity and specificity of endometrial biopsy.

Background: Stent migration and sludge formation remain significant problems associated with covered self-expandable metal stents (CSEMSs). The EGIS biliary stent fully covered flare type (EGIS biliary stent), a new type of polytetrafluoroethylene-coated self-expandable metal stent with low axial force and an anti-migration system, was developed to overcome these disadvantages. We conducted this study to evaluate the efficacy and safety of this stent in comparison with conventional CSEMS (c-CSEMS).

  • A study investigated the costs and survival outcomes associated with pegfilgrastim prophylaxis for febrile neutropenia (FN) in breast cancer patients undergoing chemotherapy in Japan.
  • The pegfilgrastim group had higher median total medical costs compared to the non-pegfilgrastim group but did not show a significant difference in FN hospitalization costs.
  • Despite increased costs, there was no significant difference in 3-year overall survival rates between the two groups, suggesting pegfilgrastim should be used selectively based on individual FN risk factors rather than routinely.

In identifying biomarkers for anticancer drugs, the lack of objectivity in selecting candidate factors makes interpretation difficult. We performed preclinical analysis and a translational validation study to identify candidate biomarkers for regorafenib efficacy in metastatic colorectal cancer (mCRC). Using in silico COMPARE analysis with a human cancer cell line panel, JFCR39, we selected candidate biomarkers whose expression correlates with regorafenib sensitivity.

  • The study focuses on using circulating tumor DNA (ctDNA) analysis to identify homologous recombination deficiency (HRD) and BRCA1/2 and ATM mutations in patients with advanced pancreatic cancer (APC).
  • Out of 702 APC patients analyzed, 4.8% had BRCA1/2 mutations and 4.4% had ATM mutations, with those having BRCA mutations showing significantly better response rates to platinum-based chemotherapy compared to those without.
  • ctDNA profiling is suggested as a valuable, non-invasive method for assessing mutation status and selecting appropriate treatments in advanced pancreatic cancer, offering a practical approach to HRD screening.

Therapeutic developments in the targeting of human epidermal growth factor receptor 2 (HER2)-expressing gastric cancer have followed the dramatic success of HER2-expressing breast cancer treatment, which has facilitated the expansion of indications for anti-HER2 agents to include not only conventional HER2-positive breast cancer, but also HER2-low and HER2-ultralow subgroups. The targetability of HER2-low gastric cancer, however, has yet to be established. Hence, further studies are needed to comprehensively understand the clinicopathological features, specific gene alterations, and distinct tumor immune microenvironment of HER2-low gastric cancer and compare them with those for HER2-positive or -negative gastric cancer.
