CBX7 inhibitors affect H3K9 methyltransferase-regulated gene repression in leukemic cells.

Exp Hematol

November 2024

European Research Institute for Biology of Ageing (ERIBA), University Medical Center Groningen, Groningen, The Netherlands; Sanquin Research, Amsterdam, the Netherlands; Landsteiner Laboratory, Cancer Center Amsterdam, Amsterdam UMC, Location University of Amsterdam, Amsterdam, the Netherlands. Electronic address:

The epigenome of leukemic cells is dysregulated, and genes required for cell cycle arrest and differentiation may become repressed, which contributes to the accumulation of undifferentiated malignant blood cells. Here, we show that the Polycomb group protein CBX7 can interact with H3K9 methyltransferases EHMT1/2 and SETDB1. We aimed to assess whether combined interfering with these H3K9 methyltransferases and CBX7 could derepress target genes and thereby induce growth arrest of leukemic cells.

Article Synopsis
  • CDK4/6 inhibitors combined with endocrine therapy improve outcomes for patients with hormone-receptor positive, HER2-negative advanced breast cancer, but evidence comparing first-line and second-line use is limited.
  • The phase 3 SONIA trial involved 1,050 patients and showed no significant overall benefit for using CDK4/6 inhibitors first-line compared to second-line (31.0 vs. 26.8 months median progression-free survival).
  • Although first-line treatment led to longer duration of CDK4/6 therapy and more severe side effects, the quality of life was similar for both treatment timing, questioning the necessity for first-line CDK4/6 inhibitor usage in all cases.

Amsterdam UMC location Vrije Universiteit Amsterdam, Molecular Cell Biology and Immunology, De Boelelaan, Amsterdam 1117, The Netherlands; Cancer Center Amsterdam, Cancer Biology and Immunology, Amsterdam, The Netherlands; Amsterdam institute for Infection and Immunity, Cancer Immunology, Amsterdam, The Netherlands. Electronic address:

The immune system is a complex network of highly specialized microenvironments, denominated niches, which arise from dynamic interactions between immune and parenchymal cells as well as acellular components such as structural elements and local molecular signals. A critical, yet underexplored, layer shaping these niches is the glycome, the complete repertoire of glycans and glycoconjugates produced by cells. The glycome is prevalent in the outer membrane of cells and their secreted components, and can be sensed by glycan binding receptors on immune cells.

In B-cell malignancies, the overexpression of MYC is associated with poor prognosis, but its mechanism underlying resistance to immunochemotherapy remains less clear. In further investigations of this issue, we show here that the pharmacological inhibition of MYC in various lymphoma and multiple myeloma cell lines, as well as patient-derived primary tumor cells, enhances their susceptibility to NK cell-mediated cytotoxicity induced by conventional antibodies targeting CD20 (rituximab) and CD38 (daratumumab), as well as T cell-mediated cytotoxicity induced by the CD19-targeting bispecific T-cell engager blinatumomab. This was associated with upregulation of the target antigen only for rituximab, suggesting additional escape mechanisms.
