Exposure-response relationship of cabozantinib in patients with metastatic renal cell carcinoma treated in routine care.

Br J Cancer

April 2024

Department of Medical Oncology, Institut du Cancer Paris CARPEM, Assistance Publique Hôpitaux de Paris, Hôpital Georges Pompidou, Paris, France.

Article Synopsis
  • The study examines how individual differences in drug absorption (pharmacokinetics) impact the effectiveness and side effects of cabozantinib in patients with metastatic renal cell carcinoma (mRCC).
  • Out of 78 patients analyzed, 67% experienced dose-limiting toxicity, and higher drug concentration in the blood was identified as a significant risk factor for these side effects.
  • The research suggests that monitoring drug levels early in treatment could help optimize cabozantinib therapy, particularly for frail patients starting on lower doses.

Vibrational spectra of a series of gas-phase metal 1+ and 2+ ions solvated by acetone molecules are collected to investigate how the metal charge, number of solvent molecules and nature of the metal affect the acetone. The spectra of Cu(Ace)(N), Cu(Ace), and M(Ace), where M = Co, Ni, Cu, and Zn are measured photodissociation by monitoring fragment ion signal as a function of IR wavenumber. The spectra show a red shift of the CO stretch and a blue shift of the C-C antisymmetric stretch.

Sci Rep

December 2023

Department of Nephrology and Organ Transplantation, Reference Centre for Rare Kidney Diseases (SORARE), French Intensive Care Renal Network (FIRN), University Hospital of Toulouse, 1, Avenue du Pr. Jean Poulhes, 31059, Toulouse Cedex, France.

Rhabdomyolysis is a risk factor for acute kidney injury, transition towards chronic kidney disease, and death. The role of calcium phosphate deposits in the mechanisms of rhabdomyolysis-induced acute kidney injury (RAKI) is still unclear. Better insight of the role calcium in RAKI could lead to new therapeutic avenues.

Many people with hemophilia A (PwHA) undergo surgery in their lifetime, often because of complications of their disease. Emicizumab is the first bispecific monoclonal antibody prophylactic therapy for PwHA, and its efficacy and safety have been previously demonstrated; however, there is a need to build an evidence base on the management of PwHA on emicizumab undergoing surgery. Data from the HAVEN 1-4 phase 3 clinical trials were pooled to provide a summary of all minor and major surgeries in PwHA with or without factor VIII (FVIII) inhibitors who were receiving emicizumab prophylaxis.

Time-Controlled Adaptive Ventilation Does Not Induce Hemodynamic Impairment in a Swine ARDS Model.

Front Med (Lausanne)

May 2022

CHRU Nancy, Service de Médecine Intensive et Réanimation, Hôpital Brabois, Vandœuvre-lès-Nancy, France.

Article Synopsis
  • The study investigates the effectiveness of a new ventilation strategy called time-controlled adaptive ventilation (TCAV) compared to traditional low tidal volume ventilation in pigs with severe acute respiratory distress syndrome (ARDS).
  • While both methods showed similar hemodynamic effects, TCAV resulted in significantly higher levels of total PEEP and lower driving pressure and lung elastance.
  • The conclusion suggests that TCAV does not cause negative hemodynamic effects and may be more beneficial for the lungs compared to conventional protective ventilation in ARDS.

Background: Optimal management of severe trauma patients with active hemorrhage relies on adequate initial resuscitation. Early administration of coagulation factors improves post-traumatic coagulation disorders, and four-factor prothrombin complex concentrate (PCC) might be useful in this context. Our main hypothesis is that four-factor PCC in addition to a massive transfusion protocol decreases blood product consumption at day 1 in severe trauma patients with major bleeding.
