Acceptability and feasibility of a urine-based tenofovir adherence assay for monitoring and providing feedback on PrEP adherence in Kenya.

Objective measures of oral PrEP adherence - especially point-of-care (POC) measures that enable real-time assessment, intervention, and feedback - have the potential to improve adherence. Our team previously developed and validated a novel urine-based POC metric of PrEP adherence. In this study, we sought to determine whether this assay is acceptable and feasible among women taking PrEP and PrEP providers in Kenya.

The earliest named stromatolite Cryptozoon Hall, 1884 (Late Cambrian, ca. 490 Ma, eastern New York State), was recently re-interpreted as an interlayered microbial mat and non-spiculate (keratosan) sponge deposit. This "classic stromatolite" is prominent in a fundamental debate concerning the significance or even existence of non-spiculate sponges in carbonate rocks from the Neoproterozoic (Tonian) onwards.

Germline inactivating mutations of the SLC25A1 gene contribute to various human disorders, including Velocardiofacial (VCFS), DiGeorge (DGS) syndromes and combined D/L-2-hydroxyglutaric aciduria (D/L-2HGA), a severe systemic disease characterized by the accumulation of 2-hydroxyglutaric acid (2HG). The mechanisms by which SLC25A1 loss leads to these syndromes remain largely unclear. Here, we describe a mouse model of SLC25A1 deficiency that mimics human VCFS/DGS and D/L-2HGA.

Mitochondrial epigenetics, particularly mtDNA methylation, is a flourishing field of research. MtDNA methylation appears to play multiple roles, including regulating mitochondrial transcription, cell metabolism and mitochondrial inheritance. In animals, bivalves with doubly uniparental inheritance (DUI) of mitochondria are the exception to the rule of maternal mitochondrial inheritance since DUI also involve a paternal mtDNA transmitted from the father to sons.

Surgical management of complicated diverticulitis: systematic review and individual patient data network meta-analysis : An EAES/ESCP collaborative project.

Background: We performed a systematic review and network meta-analysis (NMA) of individualized patient data (IPD) to inform the development of evidence-informed clinical practice recommendations.

Methods: We searched MEDLINE, Embase, and Cochrane Central in October 2023 to identify RCTs comparing Hartmann's resection (HR), primary resection and anastomosis (PRA), or laparoscopic peritoneal lavage (LPL) among patients with class Ib-IV Hinchey diverticulitis. Outcomes of interest were prioritized by an international, multidisciplinary panel including two patient partners.

Biopreservation and reversal of oxidative injury during blood storage by a novel curcumin-based gel formulation.

Blood storage lesion induces cytosolic and membrane changes driven in part by hemoglobin (Hb) oxidation reactions within red blood cells (RBCs). A novel gel formulation containing the antioxidant curcuminoids in a biocompatible solvent system was used to deliver curcumin into RBCs. Incubation of peroxide treated RBCs stored in PBS with curcumin gel led to a reduction in prooxidant ferrylHb and recovery in ATP.

Spatial and temporal analysis of West Nile virus infection in mosquito and human populations based on arboviral detection testing data.

West Nile virus (WNV) is a mosquito-borne zoonotic flavivirus which often causes asymptomatic infection in humans but may develop into a deadly neuroinvasive disease. In this study, we aimed to investigate variables potentially associated with human WNV infection using human and mosquito WNV surveillance and monitoring datasets, established over 20 years, from 2003 to 2022, across the province of Ontario, Canada. We combined climatic and geographic data, mosquito surveillance data (n = 3010 sites), blood donation arboviral detection testing data in the human population, and demographic and socio-economic data from Canadian population censuses.

Hydrothermal biochar enhances the photosynthetic efficiency and yield of alfalfa by optimizing soil chemical properties and stimulating the activity of microbial communities.

Hydrothermal biochar has demonstrated potential in enhancing crop growth by improving soil properties and microbial activity; however, its effectiveness varies with application rate, with excessive amounts potentially inhibiting plant growth. This study employed a pot experiment approach to compare varying application rates of hydrothermal biochar (ranging from 0 to 50 t/ha) and to analyze its effects on alfalfa biomass, photosynthetic efficiency, soil nutrient content, and microbial community composition. Biochar application increased alfalfa dry weight by 12.

Transcriptomic and genetic profiling in a spontaneous non-human primate model of hypertrophic cardiomyopathy and sudden cardiac death.

Hypertrophic cardiomyopathy (HCM) afflicts humans, cats, pigs, and rhesus macaques. Disease sequelae include congestive heart failure, thromboembolism, and sudden cardiac death (SCD). Sarcomeric mutations explain some human and cat cases, however, the molecular basis in rhesus macaques remains unknown.

Resolvin D1 combined with exercise rehabilitation alleviates neurological injury in mice with intracranial hemorrhage via the BDNF/TrkB/PI3K/AKT pathway.

Resolvin D1 (RvD1) is an endogenous anti-inflammatory mediator that modulates the inflammatory response and promotes inflammation resolution. RvD1 has demonstrated neuroprotective effects in various central nervous system contexts; however, its role in the pathophysiological processes of intracerebral hemorrhage (ICH) and the potential protective mechanisms when combined with exercise rehabilitation remain unclear. A mouse model of ICH was established using collagenase, and treatment with RvD1 combined with three weeks of exercise rehabilitation significantly improved neurological deficits, muscle strength, learning, and memory in ICH mice while reducing anxiety-like behavior.

Volatile hydrocarbon exposures and immune-related illnesses among Deepwater Horizon oil spill workers.

Background: Despite evidence from experimental studies linking some petroleum hydrocarbons to markers of immune suppression, limited epidemiologic research exists on this topic.

Objective: The aim of this cross-sectional study was to examine associations of oil spill related chemicals (benzene, toluene, ethylbenzene, xylene, and n-hexane (BTEX-H)) and total hydrocarbons (THC) with immune-related illnesses as indicators of potential immune suppression.

Methods: Subjects comprised 8601 Deepwater Horizon (DWH) oil spill clean-up and response workers who participated in a home visit (1-3 years after the DWH spill) in the Gulf Long-term Follow-up (GuLF) Study.

Investigating the effects of stereotactic body radiation therapy on pancreatic tumor hypoxia and microvasculature in an orthotopic mouse model using intravital fluorescence microscopy.

Despite decades of improvements in cytotoxic therapy, the current standard of care for locally advanced pancreatic cancer (LAPC) provides, on average, only a few months of survival benefit. Stereotactic Body Radiation Therapy (SBRT), a technique that accurately delivers high doses of radiation to tumors in fewer fractions, has emerged as a promising therapy to improve local control of LAPC; however, its effects on the tumor microenvironment and hypoxia remain poorly understood. To explore how SBRT affects pancreatic tumors, we combined an orthotopic mouse model of pancreatic cancer with an intravital microscopy platform to visualize changes to the in vivo tumor microenvironment in real-time.

Infectious agent release and Pacific salmon exposure at Atlantic salmon farms revealed by environmental DNA.

The potential risk posed by infectious agents (IAs) associated with netpen aquaculture to wild fishes is determined based on the "release" of IAs from netpens into the environment, the "exposure" of the wild fish to those released agents, and the "consequence" for wild fish experiencing infection by those agents. Information available to characterize these three factors is often lacking, and the occurrence of transmission from aquaculture to wild fish as well as potential consequences of such transmission are difficult to observe. In this study, we utilized environmental DNA (eDNA) to characterize the release of dozens of IAs from, and exposure of Pacific salmon to, Atlantic salmon aquaculture.

Hypoxia-induced TPC2 transcription and glycosylation aggravates pulmonary arterial hypertension by blocking autophagy flux.

Pulmonary arterial hypertension (PAH) is a serious medical condition that causes a failure in the right heart. Two-pore channel 2 (TPC2) is upregulated in PAH, but its roles in PAH remain largely unknown. Our investigation aims at the mechanisms by which TPC2 regulates PAH development.

Postoperative atrial fibrillation following cardiac surgery in severe obesity: the added value of waist circumference.

Introduction: Obesity is an independent risk factor for postoperative atrial fibrillation (POAF) after coronary artery bypass grafting (CABG) surgery. POAF in patients with severe obesity (body mass index [BMI] ≥ 35 kg/m) is less studied. Whether waist circumference (WC) improves prediction of POAF independently of BMI among patients with severe obesity remains unknown.

Caftaric acid attenuates kidney and remote organ damage induced by renal ischemia-reperfusion injury.

Oxidative stress and inflammation are indispensable components of ischemia-reperfusion (IR) injury. In this study, we investigated the effects of low and high doses of caftaric acid (CA) on reducing kidney and remote organ damage induced by IR. We divided Wistar rats into four groups: sham, IR, low (40 mg/kg body weight (BW)), and high (80 mg/kg BW) CA groups.

Effects of cation types on physicochemical parameters and micro-structure of soft clay for electrochemical treatment.

To investigate the influence of cations on the microstructural characteristics of electrochemical reinforcement in soft clay, a study was conducted using three different cationic salt solutions-NaCl, CaCl₂, and FeCl₃-for grouting treatment. Four sets of indoor experiments were performed to examine the reinforcement mechanism of the electrochemical method. The findings indicate that increasing the valence of injected cations significantly affects the electrochemical reinforcement effect and the soil's microstructural properties.

The role of C-reactive protein and ferritin in the diagnosis of HLH, adult-onset still's disease, and COVID-19 cytokine storm.

Cytokine storm syndromes such as hemophagocytic lymphohistiocytosis (HLH), Adult-onset Still's disease (AOSD), and COVID-19 cytokine storm (CCS) are characterized by markedly elevated inflammatory cytokines. However clinical measurement of serum cytokines is not widely available. This study examined the clinical utility of C-reactive protein (CRP) and ferritin, two inexpensive and widely available inflammatory markers, for distinguishing HLH from AOSD and CCS.

Optimizing adult-oriented artificial intelligence for pediatric chest radiographs by adjusting operating points.

The purpose of this study was to evaluate whether the optimal operating points of adult-oriented artificial intelligence (AI) software differ for pediatric chest radiographs and to assess its diagnostic performance. Chest radiographs from patients under 19 years old, collected between March and November 2021, were divided into test and exploring sets. A commercial adult-oriented AI software was utilized to detect lung lesions, including pneumothorax, consolidation, nodule, and pleural effusion, using a standard operating point of 15%.

Effects of mycelium post-ripening time on the yield, quality, and physicochemical properties of Pleurotus geesteranus.

This study determined the effects of the mycelium post-ripening time on the growth of Pleurotus geesteranus and the substrate metabolism. The characteristic indexes and timing reflecting the physiological maturity of P. geesteranus mycelium were identified to facilitate precise cultivation in factories.

Nutrient loading as a key cause of short- and long-term anthropogenic ecological degradation of the Salton Sea.

The Salton Sea (SS), California's largest inland lake at 816 square kilometers, formed in 1905 from a levee breach in an area historically characterized by natural wet-dry cycles as Lake Cahuilla. Despite more than a century of untreated agricultural drainage inputs, there has not been a systematic assessment of nutrient loading, cycling, and associated ecological impacts at this iconic waterbody. The lake is now experiencing unprecedented degradation, particularly following the 2003 Quantification Settlement Agreement-the largest agricultural-to-urban water transfer in the United States.

Analysis of canopy light utilization efficiency in high-yielding rapeseed varieties.

The photosynthetic mechanism responsible for the differences in yield between different rapeseed varieties remains unclear, and there have been no consensus and definite conclusions about the relationship between photosynthesis and yield. Representation of the whole plant by measuring the photosynthetic performance at a single site may lead to biased results. In this study, we comprehensively analyzed the main photosynthetic organs of four high-yielding rapeseed varieties at the seedling, bud, flowering, and podding stages.
