Transthyretin (TTR) is a natively tetrameric thyroxine transporter in blood and cerebrospinal fluid whose misfolding and aggregation causes TTR amyloidosis. A rational drug design campaign identified the small molecule tafamidis (Vyndamax) as a stabilizer of the native TTR fold, and this aggregation inhibitor is regulatory agency approved for the treatment of TTR amyloidosis. Here we used cryo-EM to investigate the conformational landscape of this 55 kDa tetramer in the absence and presence of one or two ligands, revealing inherent asymmetries in the tetrameric architecture and previously unobserved conformational states.

Here we investigated the efficacy and safety of cadonilimab (PD-1/cytotoxic T lymphocyte antigen-4 (CTLA-4) bispecific antibody) plus chemotherapy as first-line treatment in G/GEJ adenocarcinoma. The dataset covers the basins of the Laurentian Great Lakes and the Ottawa River and extends over the period 2001-2018. The data consist of all variables (hourly fluxes and state variables) related to the water balance of GEM-Hydro's land-surface scheme (including precipitation, surface and sub-surface runoff, drainage, evaporation, snow water equivalent, soil moisture…) and mean daily streamflow at 212 gauge locations.

With rising incidence, mortality and limited therapeutic options, heart failure with preserved ejection fraction (HFpEF) remains one of the most important topics in cardiovascular medicine today. Characterised by left ventricular diastolic dysfunction partially due to impaired Ca homeostasis, one ion channel in particular, SarcoEndoplasmic Reticulum Ca-ATPase (SERCA2a), may play a significant role in its pathophysiology. A better understanding of the complex mechanisms interplaying to contribute to SERCA2a dysfunction will help develop treatments targeting it and thus address the growing clinical challenge HFpEF poses.

Methods: A systematic literature search was conducted in MEDLINE, EMBASE, the Cochrane library, Scopus, and ProQuest from inception to February 2023 to identify studies capturing the care of IBD among immigrants who moved from one country to another, irrespective of the timing of IBD diagnosis.
